    Notes: Six types of hemocytes were identified in fifth instars of the Indian meal moth, Plodia interpunctella. The morphology of these cells was characterized by phase contrast and electron microscopy, with Sudan black B, Giemsa, Janus green B, and periodic acid-Schiff staining. Reaction of the hemocytes with seven fluorescing lectin conjugates revealed distinctive binding patterns by their plasma and nuclear membranes and cytoplasmic inclusions. A direct line of descent from prohemocytes to plasmatocytes to granulocytes is suggested from these morphological observations.

    Notes: Small swellings near the base of the radial vein in each fore wing of the green lacewing, Chrysopa carnea, resemble typical insect tympanal organs, but some important differences are apparent. The swellings are bounded dorsally and laterally by thick cuticle and ventrally by thin, membranous cuticle. The ventral membrane is formed by a single, thin sheet of exocuticle with flattened hypodermis internally, but lacks the tracheal component that forms part of the tympanum in the typical insect tympanal organ. The portion of the membrane beneath each swelling is rippled while proximally it is smooth. In contrast to typical insect tympanal organs, the swellings in C. carnea are largely fluid-filled since an unexpanded trachea runs through each organ. A distal and a proximal chordotonal organ composed of typical chordotonal sensory units are associated with each swelling. The distal organ contains from five to seven units while the proximal organ is composed of from 18 to 20 units. Each sensory unit is composed of three readily identifiable cells. Distally, an attachment cell unites with the membrane and is contiguous with the scolopale cell, which surrounds the dendrite of the bipolar neuron. On the basis of the morphological evidence, one would not expect these swellings to function as sound receptors. However, the results of physiological and behavioral experiments, presented elsewhere, show that these organs are receptors for ultrasound.

    Notes: A model describing two consecutive gas-liquid reactors in a continously stirred-tank reactor is developed with reaction factors used to account for the interactions between the chemical and transport rate processes. A parametric numerical study revealed several interesting effects, including the existence of up to seven steady state solutions. The predictions of the model adequately describe the steady state multiplicity observed during the chlorination of liquid n-decane. It is shown that the steady state multiplicity can serve as an excellent tool for discriminating among rival models and estimation of kinetic parameters.

    Notes: A model of an adiabatic continuously stirred-tank reactor in which a single exothermic second order-gas liquid reaction occurs has been developed. The interactions between the rate of the chemical reaction, the diffusional resistances, and the solubility may cause the occurrence of up to five steady state solutions, even though no more than three solutions can be attained when the same reaction is carried out in a sinle phase CSTR. A parametric study is used to examine the sensitivity of the model to the value of several parameters. Topological arguments are used to obtain a simple instability criterion.

    Notes: The progressive differentiation of both normal rat osteoblasts and HL-60 promyelocytic leukemia cells involves the sequential expression of specific genes encoding proteins that are characteristic of their respective developing cellular phenotypes. In addition to the selective expression of various phenotype marker genes, several members of the heat shock gene family exhibit differential expression throughout the developmental sequence of these two cell types. As determined by steady state mRNA levels, in both osteoblasts and HL-60 cells expression of hsp27, hsp60, hsp70, hsp89α, and hsp89β may be associated with the modifications in gene expression and cellular architecture that occur during differentiation.In both differentiation systems, the expression of hsp27 mRNA shows a 2.5-fold increase with the down-regulation of proliferation while hsp60 mRNA levels are maximal during active proliferation and subsequently decline post-proliferatively. mRNA expression of two members of the hsp90 family decreases with the shutdown of proliferation, with a parallel relationship between hsp89α mRNA levels and proliferation in osteoblasts and a delay in down-regulation of hsp89α mRNA levels in HL-60 cells and of hsp89β mRNA in both systems. Hsp70 mRNA rapidly increases, almost twofold, as proliferation decreases in HL-60 cells but during osteoblast growth and differentiation was only minimally detectable and showed no significant changes. Although the presence of the various hsp mRNA species is maintained at some level throughout the developmental sequence of both osteoblasts and HL-60 cells, changes in the extent to which the heat shock genes are expressed occur primarily in association with the decline of proliferative activity. The observed differences in patterns of expression for the various heat shock genes are consistent with involvement in mediating a series of regulatory events functionally related to the control of both cell growth and differentiation.

    Notes: Ruber hapatoma cells are useful cultured lines for the study of insulin action, lipid and lipoprotein metabolism, and the regulation of acetyl-CoA carboxylase (ACC), the rate-limiting enzyme of faty acid biosynthesis. During investigations in deifferent clonal lines of these of these cells, we have uncovered marked intercellular variability in the activity, enzyme content, and insulin regulation of ACC paralleled by differences in cellular neutral lipid (triglyceride) content. Two contrasting clonal liners, Fao and H356A-1, have been studied in detail. Several features distinguish these two lines, including differences in ACC activity and enzyme kinetics, the content of the two major hepatic ACC isozymes (M, 280,000 and 265,000 Da) and their heteroisozymic complex, the extent of ACC phosphorylation, and the ability of ACC to be activated on stimulation by insulin and insulinomimetic agonists. As studied by Nile Red staining and fluorscence-activated cell sorting, these two lines also display marked differences in neutral lipid content, which correlates with both basal levels of ACC activity and inhobition of ACC by the fatty acid analog, 5-(tetradecyloxy)-2-furoic and (TOFA). Thsese results emphasize the importance of characterization of any particular clonal line of Reuber cells for studies of enzyme regulation, substrate metabolism, and hormone action. With respect to ACC, studies in contrasting colonal lines of Ruber cells could provide valuable clues to understanding both the complex mechanisms of intracellular ACC regulation in the absence and presence of hormones and its regulatory role (s) in overla hepatic lipid metabolism.
