    Notes: Summary Selection for a character controlled by additive genes induces linkage disequilibrium which reduces the additive genetic variance usable for further selective gains. Additive x additive epistasis contributes to selection response through development of linkage disequilibrium between interacting loci. To investigate the relative importance of the two effects of linkage disequilibrium, formulae are presented and results are reported of simulations using models involving additive, additive x additive and dominance components. The results suggest that so long as epistatic effects are not large relative to additive effects, and the proportion of pairs of loci which show epistasis is not very high, the predominant effect of linkage disequilibrium will be to reduce the rate of selection response.

    Notes: Summary There is an inherent bias in intraclass correlations since the expectation of a ratio does not equal the ratio of expectations. A simple accurate approximation for this bias is derived, and it is found that the inherent bias is usually negligible. Selection of sires is known to bias half-sib heritability estimates, and appropriate formulae are given and discussed.

    Notes: Summary Prediction of response to selection for traits with direct and maternal components is described for discrete and overlapping generations. Expected phenotypic response is the sum of direct and maternal contributions, the latter having a genetic and an environmental component. With overlapping generations the selection differentials achieved on these components are added to respective updated vectors containing age-sex distributions with values of previous selection rounds. An example demonstrates that in the early stages, results may be considerably affected by environmental correlations between direct and maternal effects. The method could be helpful in interpreting phenotypic changes in a population selected for traits with maternal effects.

    Notes: Films consisting of a rigid-rod polymer and thermoset resin matrixes were prepared. Poly{(benzo[1,2-d : 5,4-d′]bis(oxazole-2,6-diyl))-1,4-phenylene} (PBO) in polyphosphoric acid (PPA) was blended with 2,6-bis(4-benzocyclobutene) benzo[1,2-d : 5,4-d′]bis(oxazole) (1), and films were extruded from these solutions. The coagulated films were soluble in methanesulfonic acid (MSA). After heat treatment at 300°C, the films became insoluble in MSA. Crosslinked films were homogeneous and did not show phase segregation between the two components. These were composite films at the molecular level. Transmission electron microscopy (TEM) showed enhanced interlayer integrity and reduced microfibril separation for the molecular composite films as compared to normal PBO film. These films had significantly better torsion and tension delamination resistance. The incorporation of a second component did not sacrifice the tensile properties of PBO film.     Notes: Uniquely among class A β-lactamases, the RTEM-1 and RTEM-2 enzymes contain a single disulfide bond between Cys 77 and Cys 123. To study the possible role of this naturally occurring disulfide in stabilizing RTEM-1 β-lactamase and its mutants at residue 71, this bond was removed by introducing a Cys 77 → Ser mutation. Both the wild-type enzyme and the single mutant Cys 77 → Ser confer the same high levels of resistance to ampicillin in vivo to Echerichia coli; at 30°C the specific activity of purified Cys 77 → Ser mutant is also the same as that of the wild-type enzyme. Also, neither wild-type enzyme nor the Cys 77 → Ser mutant is inactivated by brief exposure to p-hydroxymercuri-benzoate. However, above 40°C the mutant enzyme is less stable than wild-type enzyme. After introduction of the Cys 77 → Ser mutation, none of the double mutants (containing the second mutations at residue 71) confer resistance to ampicillin in vivo at 37°C; proteins with Ala, Val, Leu, Ile, Met, Pro, His, Cys, and Ser at residue 71 confer low levels of resistance to ampicillin in vivo at 30°C. The use of electrophoretic blots stained with antibodies against β-lactamase to analyze the relative quantities of mutant proteins in whole-cell extracts of E. coli suggests that all 19 of the doubly mutant enzymes are proteolyzed much more readily than their singly mutant analogues (at Thr 71) that contain a disulfide bond. Thus, the disulfide bond of the RTEM-1 β-lactamase seems able to reduce the destabilizing effect of mutations at Thr 71. These results also emphasize the unique and essential role that Thr 71 performs in the stable folding of RTEM-1 β-lactamase and presumably the other class A β-lactamases.

    Notes: To our benefit, the modelling of fluid-solid catalyzed reaction-reactor systems has long been the keen concern of Hanns Paul Hofmann. In the light of his instruction we remark upon key aspects of modelling for the (a) fixed bed (b) fluid bed and (c) moving bed/transport line reactors. There emerges from such analyses a properly renewed awareness of the need for a more sophisticated respect for reliable physical-chemical data. As Hanns has taught us, our models are no btter than our data.

    Notes: The majority of the fluidized-bed reactor models are models are rooted in the tacit assumption that transport between the bubble, cloud, and emulsion phases occurs in series with chemical reaction. A more realistic model that anticipates simultaneous transport and reaction is presented in terms of a fluid-bed effectiveness factor which embraces the limits between chemical and mass transport control. Analysis of the predictive capacity of this model vis-a-vis the Kunii-Levenspiel model reveals signal differences in chemical conversion.

    Notes: The paper describes the application of a stochastic separated flow model for the dispersed phase to the prediction of a particle-laden turbulent air jet discharging at 13 m/s from a 15 mm nozzle into stagnant surroundings. Emphasis is placed on the stochastic element of the Lagrangian particle tracking part of the model and on the importance of particle initial conditions over the first 20 jet diameters. Calculations are presented for 80 m̈m sized glass particles which clarify how particles with turbulent Stokes number less than unity acquire axial turbulence much larger than radial. Far from being only a response to the gas-phase turbulence as implied by the model, the axial turbulence is shown to be also produced by an interaction between particle radial turbulence fluctuations and cross-stream spatial gradients in particle mean velocity, here referred to as "fanspreading". In addition, initial particle turbulence levels remain identifiable for about 10 jet diameters; the initial radial turbulence reinforces the fan-spreading contribution and leads to extra generation of axial turbulence farther downstream. In general, the results agree well with experimental measurements.

    Notes: The 1H and 13C{1H} NMR spectra of 4-formylcyclohexene in 50% CF2Cl2-50% CHF2Cl decoalesce at very low temperatures and, at 108 K, have sharpened into a major (77%) and a minor (23%) subspectrum. Based on the NMR spectra and molecular mechanics calculations, the major subspectrum is assigned to a family of equatorial conformations and the minor subspectrum to a family of axial conformations. The free energy of activation for conversion of the equatorial conformations to the axial forms is 5.6 kcal mol-1 (1 kcal  =  4.184 kJ) at 117 K. Within a family of conformations, interconversion occurs rapidly at 108 K via formyl group rotation. The conformational preference in 4-formylcyclohexene is solvent dependent. In 50% CF2Cl2-50% CHF2Cl, CHF2Cl and CF2Cl2, the respective ratios of equatorial to axial conformations are 77:23, 77:23, and 89:11 at 108 K. The 1H and 13C{1H} NMR spectra of 4-methylcyclohexene and 4-vinylcyclohexene in 50% CF2Cl2-50% CHF2Cl show no evidence of decoalescence at very low temperatures but do show differential broadening and subsequent sharpening of various resonances characteristic of exchange between strongly dominant equatorial conformations and axial conformations present at too low a concentration to be detectable by NMR. A lower limit on the free energy preference for the equatorial conformations is estimated to be 1.0 kcal mol-1. Molecular mechanics calculations also predict a stronger preference for equatorial conformations in 4-methylcyclohexene and 4-vinylcyclohexene than in 4-formylcyclohexene. © 1998 John Wiley & Sons, Ltd.
