    Notes: An expression cassette for continuous high-level expression of secreted glycoproteins by transformed lepidopteran insect cells has been developed as an alternative to baculovirus and mammalian cell expression systems. The expression cassette utilizes the promoter of the silkmoth cytoplasmic actin gene to drive expression from foreign gene sequences, and also contains the ie-1 transactivator gene and the HR3 enhancer region of BmNPV to stimulate gene expression. Using an antibiotic-resistance selection scheme, we have cloned a Bm5 (silkmoth) cell line overexpressing the secreted glycoprotein juvenile hormone esterase (JHE-KK) at levels of 190 mg/L in batch suspension cultures. A baculovirus (AcNPV) expressing the same gene under the control of the p10 promoter of AcNPV produced only 4 mg/L active JHE in static cultures of infected Sf21 cells. A cloned Bm5 cell line overexpressing a soluble isoform of the α-subunit of the granulocyte-macrophage colony stimulating factor receptor (solGMRα) was also generated and produced five times more solGMRα in static cultures than a cloned BHK cell line obtained by transformation with a recombinant expression cassette utilizing the human cytomegalovirus (CMV) enhancer-promoter system. Finally, we show that recombinant protein expression levels in transformed Bm5 cells remain high in serum-free media, that expression is stable even in the absence of antibiotic selection, and that lepidopteran cells other than Bm5 may be used equally efficiently with this new expression cassette for producing recombinant proteins. © 1998 John Wiley & Sons, Inc.     Notes: The synthesis of several dibenzo-18-crown-6 derivatives, and their acid catalyzed incorporation into crosslinked styrene and 4-methoxystyrene based copolymers is described. Some of these reactions were carried out in the presence of chiral ammonium cations acting as non-covalent templates. The polymers have been characterized by IR, and by 1H and 13C solution state and magic angle spinning NMR.

    Notes: Curing reactions of certain laminates involve a series of chemical structure changes which occur with increasing temperature and/or pressure. This paper describes the use of a midinfrared chalcogenide fiber to monitor the lamination of polymer prepregs in an autoclave. The fiber optic was used both as a wave guide and as a sensing element for acquiring evanescent wave spectra. The spectra were measured by embedding the fiber optic in a polymer prepreg package. The polymer prepregs studied include epoxy, polyimide, and bismaleimide triazine (BT) resins. The degree of cure and an understanding of the kinetic processes were obtained from the absorbance changes in specific bands. This in situ sampling technique demonstrates a new area of Fourier transform infrared (FTIR) spectroscopy for following the formation of polymeric bonds. © 1995 John Wiley & Sons, Inc.

    Notes: Raman spectra of H2S isolated in N2 at M/A ratios of 30, 50, 100, and 300 and in Ar at M/A ratios of 50, 100, 300, and 1000 have been measured in the S-H stretching region. The spectral results confirm the infrared measurements on H2S isolated in N2; however, the symmetric stretching vibration, v1, of the H2S monomer is at about the same frequency, 2619 cm-1, in the spectra of Ar matrices. This disagrees with previous infrared measurements of Ar matrices. Raman bands in the spectra of both N2 and Ar matrices due to a dimer of H2S are consistent with those observed in the infrared spectrum of N2 matrices, and they support a donor-acceptor model for the dimer.
