    Notes: The crystallized rennin of N. J. Berridge was found to be not quite homogeneous at a pH of 7 in the ultracentrifuge and in diffusion measurements. An approximate molecular weight of 40,000 was calculated for the main fraction from the sedimentation and diffusion constants. The rennin behaved uniformly during paper-electrophoresis between pH 5 and 8 and at 2. The isoelectric point lies in the neighbourhood of 4,5. In conclusion, the quantitative amino acid composition was determined with the chromatographic method of Stein & Moore.

    Notes: The influence of a range of acid pH values on human oxy- and carboxyhemoglobin was thoroughly investigated. Above pH 4 at most a very slow conversion into hemiglobin (in the presence of O2) takes place. Denaturation sets in below pH 4 unmasking approximately 34 proton-binding groups per mole of oxyhemoglobin. Heme always appears as a cleavage product with the acid-denaturation, the cleavage becoming more and more quantitative on lowering the pH value. The split-off heme or hemin can be extracted with acetone which simultaneously precipitates the protein. After reneutralization to pH 7.3, it is soluble in water, the solubility percentage increasing with decreasing pH values of the cleavage process. The extent of renaturation which occurs on neutralization of the hydrochloric acid globin increases with increasing completeness of removal of the pigment. The cleavage velocity of oxy- and carboxyhemoglobin at various pH values was spectroscopically measured.

    Notes: Bei der elektrophoretischen Untersuchung von Caseinatlösungen verschiedener Konzentration (0,3 bis 3,8%) findet man bei Auswertung der absteigenden Gradienten prozentual um so weniger β-Casein, je konzentrierter die Lösung ist. Dies ist weniger durch Grenzflächenanomalien als durch die reversible Komplexbildung zwischen α- und β-Casein bedingt, die in erwarteter Weise konzentrationsabhängig ist. Das gebundene β-Casein verschwindet auf den Diagrammen im α-Casein-Gradient. Interpolation der Werte auf die Konzentration gleich Null ergibt einen Maximalwert für das β-Casein von 25%. Dies dürfte dem wirklichen Gehalt des normalen Caseins entsprechen.

    Notes: During the rennet curdling of milk, the amount of nitrogen (NPN) which is not precipitated by 12% trichloroacetic acid (TCE) increases markedly. The course of this splitting reaction can be followed quantitatively. The NPN/time curves show clearly that a very specific reaction is involved which terminates before visible clotting occurs and which differs distinctly from the slow but general proteolytic breakdown effected by the enzyme.

    Notes: Under suitable conditions it is possible to introduce into dissolved denatured gelatin molecules intracatenar cross-links. Working with 2% solutions of commercial gelatin (Mn approximately 40000) at 40° and pH 5,75 and with water-soluble carbodiimid as a condensing agent we succeeded in inserting 7-8 intracatenar amide bonds per molecule. The number of cross-links introduced was determined by the decrease of free —NH2 and —COOH groups.Using 4% solutions a gel melting point of the cross-linked preparations below 0° was obtained as compared with the gel melting point of untreated gelatin at 28°.The low viscosity and the behaviour in gel chromatography on Sepharose 6B show that the cross-linked peptide chains are fixed as rather dense coils.A small amount of the cross-linking reagent remains covalently bound to the protein, as was shown with tritium labeled carbodiimid. The side reactions possibly responsible for this occurrence are discussed. The bonds between reagent and gelatin are fairly stable at acidic pH, but labile at alkaline pH.The results of optical measurements (UV. and CD.) on cross-linked gelatin will be reported in another paper.
