    Notizen: Summary The majority (about 90%) of children developing Type 1 (insulin-dependent) diabetes mellitus do not have a first-degree relative with the disease. Nearly all (389/405, 96%) children (0–14 years) in Sweden, who developed diabetes during one year, were therefore studied to compare islet cell, thyroid peroxidase, thyroglobulin, and gastric H+, K+-ATPase antibodies with 321 age, sex, and geographically matched, but non-related, control children. Islet cell (cytoplasmic) antibodies were found in 81% (316/389) of the patients and in 3% (9/321) of the control children (p〈0.001). The median islet cell antibody levels were 70 (range 3–8200) Juvenile Diabetes Foundation (JDF) Units in the islet cell antibody positive patients, and 27 (range 17–1200) JDF Units in the control children (NS). Autoantibodies against thyroid peroxidase (8%), thyroglobulin (6%), and gastric H+, K+- ATPase (3%) were all increased in the patients compared with the control children, being 2% (p〈0.001), 2% (p〈0.01), and 0.3% (p〈0.01), respectively. During an observation time of 20–34 months, two of the nine islet cell antibody positive control children developed Type 1 diabetes, after 8 and 25 months respectively, while the others remained healthy and became islet cell antibody negative. None of the islet cell antibody negative control children developed diabetes during the same time of observation. This first investigation of an unselected population of diabetic children and matched control children shows: that islet cell antibodies are strongly associated with newly diagnosed childhood diabetes, that other autoantibodies are more frequent among diabetic children than control children, and that the frequency of islet cell antibodies in the background population of children is higher than previously documented, and could also be transient, underlining that factors additional to islet cell antibodies are necessary for the later development of Type 1 diabetes.

    Notizen: Summary Enantiomeric separation of mosapride and a structurally related compound was performed using chiral chromatography and experimental design. Unique effects of mobile phase pH and column temperature made it possible to control the elution order of the enantiomers when using Chiral-AGP as the solid phase. At a low mobile phase pH (〈6) the (R)-enantiomer of mosapride elutes before the (S)-form whereas the (S)-enantiomer elutes first at a high mobile phase pH (〉6). By using a mobile phase pH around 6, the column temperature could also be used to control the elution order of the enantiomers of mosapride. Similar effects of mobile phase pH and column temperature were obtained for the enantiomers of a structurally related compound, a metabolite (M1). Isocratic chromatographic systems made it possible to determine enantiomeric impurities less than 0.1% in the respective enantiomer of mosapride. The enantiomers of mosapride as well as the enantiomers of M1 could easily be separated simultaneously using Chiral-AGP and a simple gradient elution.

    Notizen: Summary A strategy for general optimization of the mobile phase in LC-systems is described in this study. The influence on column efficiencies and capacity factors of three uncharged modifiers, two charged modifiers, pH and column temperature was evaluated with experimental design and multivariate analyses. A factorial design was used and the experimental results demonstrated that the uncharged modifiers and the pH were important for the capacity factors while column temperature and pH were important for the column efficiency. Two different stationary phase materials were used and compared, one was a traditional silica based material and the other a silica covered with a polymer. Both were reversed phase materials. A partial least squares model obtained from the results from the polymer column, Chromspher B, was used to predict retention data.

    Notizen: Summary A strategy for the optimisation of direct chiral separation of omeprazole and a metabolite, hydroxi-omeprazole, in reversed phase liquid chromatography is described. A factorial design was used, where mobile phase pH, concentration of a mobile phase modifier, ionic strength and column temperature were tested as the variables and enantioselective retention, column efficiency and asymmetry factor as the responses. The experimental results were evaluated with multivariate analyses, which demonstrated that the column temperature and content of mobile phase acetonitrile were by far the most important variables. The enantiomers of omeprazole and one of its metabolites were baseline resolved within 15 minutes. The optimised chromatographic system was used for a separation of the enantiomers of omeprazole and its main metabolite in a patient plasma sample.

    Notizen: Summary In this paper chemometrics have been used to study and characterize peak compression phenomena in packed column SFC. A carbon dioxide/2-methyl-1-propanol mobile phase was used in the experimental design (modifier concentration, temperature and pressure) and modelling part of the investigation. A cubic interaction term was needed in the model to obtain a reasonable fit, suggesting that all three parameters are of significance in terms of controlling peak compression. At the optimum conditions derived from the model a narrow peak was obtained as predicted.
