    Notes: Abstract The relationship among stomatal density, photosynthetic rate, leaf conductance, plant growth, bean yield and kaempferol triglucoside (K9) in the leaves of soybean (Glycine max (L.) Merr.) was examined in two field tests. K9 in the leaves was associated with reduced stomatal density, reduced photosynthetic rate, reduced stomatal conductance, reduced plant weight and lower bean yield. Plants with high stomatal frequency (lacking K9) were better able to take advantage of increased water supply by increasing stomatal conductance (upper surface), transpiration and bean yield. Plants with low stomatal frequency (with K9) were unresponsive to irrigation and in this sense were more tolerant of water stress, but their overall yield was low.

    Notes: Summary A gas chromatograph/mass selective detector method is presented for the analysis of the major metabolic residues of metolachlor by acid hydrolysis of tomato fruit. The two major hydrolysis products, CGA-37913 (2-[2-ethyl-6-methylphenyl]amino-1-propanol) and derivatised CGA-49751 (4-[2-ethyl-6-methylphenyl]-2-hydroxy-5-methyl-3-morpholinone) were monitored in the selective ion mode atm/z 162 andm/z 160, respectively. Recoveries from 87 to 96% were obtained for tomato fortified at 50 to 200μg kg−1. The proposed method of analysis will allow for a detection limit of 11 μg kg−1 for CGA-37913 and 12 μg kg−1 for the derivatised CGA-49751.

    Notes: Summary An HPLC method is described for the determination of kaempferol glycosides in soybean leaf extracts. The method is rapid and can provide qualitative and relative quantitative results for 9 kaempferol glycosides. The flavonol glycosides are extracted from soybean leaves by shaking the samples in 50% methanol; the extracts are filtered, evaporated to dryness and reconstituted in methanol before further purification through a C-18 Sep-Pak column. The samples are injected onto a C-18 HPLC column, separated by gradient elution with a 1% phosphoric acid: methanol mixture and detected on a UV/VIS diode-array detector. Flavonols were monitored at 265 and 348 nm and spectra from 180 to 400 nm were stored and used as an aid in identification. Relative quantities of the kaempferol glycosides among soybean cultivars were calculated from their proportion of peak area in the chromatograms. Total kaempferol concentration of the extracts was calculated after acid hydrolysis of the kaempferol glycosides to the aglycone and comparison of peak areas to kaempferol standards.
