    Notes: Summary A monoclonal antibody (Ted-R-1) to the keratohyalin granules (KHGs) of the newborn rat epidermis was developed by immunizing mice with a pH 4.7 hematoxylin-stainable fraction of the extract in a 50 mM Tris-HCl buffer, pH 7.3, containing 10 μ/ml phenylmethylsulfonyl fluoride (PMSF) from the newborn rat whole epidermis. Using an indirect immunofluorescence technique, Ted-R-1 was located in two places: (a) the KHGs and (b) the cell membrane region of the stratum corneum. Under immunoelectron microscopy, the antigenic materials were located at the periphery of the KHGs. Immunoblotting analyses of the antigenic pH 4.7 fraction, the Tris-HCl extract from the stratum corneum, and the Tris-8M urea extract from freeze-dried whole skin demonstrated that there were 55 kd, 43 kd, and 78 kd polypeptides, respectively, with which Ted-R-1 reacts. No possitive spot was seen in the dermal extract. The major amino acids of the 55 kd protein were glycine (24.4%), serine (17.3%), glutamic acid (14.1%), and alanine (7.7%), while the histidine residue was only 3.1%. On the basis of its amino acid composition, the 55 kd protein is the third component of KHGs — in addition to histidine-rich and cystinerich proteins.

    Notes: Abstract In order to identify the endogenous protease associated with stratum corneum (SC) desquamation, we examined properties of proteases in the stratum corneum of normal human skin. SC were obtained by tape stripping, washed in toluene and then dried. The proteolytic activity in SC was measured using peptidyl 4-methyl-coumaryl-7-amides (MCAs). The SC was dispersed uniformly in the reaction mixture with dimethylformamide and Triton X-100 and incubated with the peptidyl MCAs. The protease in the SC hydrolysed both Boc-Phe-Ser-Arg-MCA and Boc-Gln-Ala-Arg-MCA (substrates for trypsin) very effectively. The hydrolytic activity was inhibited by the serine protease inhibitors diisopropyl fluorophosphate (DFP), aprotinin, antipain and leupeptin, but not by chymostatin, a chymotrypsin inhibitor. These results show that one or more trypsin-like serine protease is present in the SC of normal human skin. Casein-acrylamide electrophoresis showed that the molecular weight of this serine protease was about 30 kDa. We have previously shown that cells dissociate from human SC sheets in a detergent mixture (N,N-dimethyldodecylamine oxide and sodium lauryl sulphate). This cell dissociation was inhibited by aprotinin and leupeptin. In addition, the proteolytic activity in the outer SC was higher than that in the inner SC, and the activity in the SC of scaly skin induced by SLS treatment was higher than that of untreated skin. These results strongly suggest that the trypsin-like serine protease described here is involved in SC desquamation.

    Notes: The interaction of RecA protein with short single-stranded oligonucleotides is characterised by flow linear dichroism (LD), isoelectric focusing (IEF) and electron microscopy (EM). From LD and EM it is evident that RecA forms long filaments with at least some 50 oligonucleotides in a ‘train formation’. The tendency to form trains is substantially lower when an amino group is attached to the 5′ end of the oligonucleotide, suggesting that the modification impairs protein-protein interactions at the interface between two oligomers. From LD it is also evident that no bridging occurs between RecA-Oligonucleotide complexes containing more than one oligomer strand per RecA filament. This property make them manageable in polyacrylamide gels, hence allowing characterisation by IEF. RecA was found acidic with a pI of 5.0. The pI was not dependent on the presence of bound cofactor (ATPγS) and oligonucleotides suggesting that protonation of the protein readily occurs to compensate for the negative charges provided by bound cofactor and DNA.

    Notes: The perturbation expansion coefficients for the ground-state energy of the half-filled one-dimensional Hubbard model with N = 4ν + 2, (ν = 1, 2,…) sites and satisfying cyclic boundary conditions were calculated in the Hückel limit (U/β → 0), where β designates the one-electron hopping or resonance integral, and U, the two-electron on-site Coulomb integral. This was achieved by solving - order by order - the Lieb-Wu equations, a system of transcendental equations that determines the set of quasi-momenta {ki} and spin variables {τα} for this model. The exact values for these quantities were found for the N = 6 member ring up to the 20th order in terms of the coupling constant B = U/2β, as well as numerically for 10 ≤ N ≤ 50, and the N = 6 Lieb-Wu system was reduced to a system of sextic algebraic equations. These results are compared with those of the infinite system derived analytically by Misurkin and Ovchinnikov [Teor. Mat. Fiz. 11, 127 (1972)]. It is further shown how the results of this article can be used for the interpolation by the root of a polynomial. It is demonstrated that a polynomial of relatively small degree provides a very good approximation for the energy in the intermediate coupling region. © 1995 John Wiley & Sons, Inc.

    Notes: The finite-order many-body perturbation theory using the localized Wannier orbital basis is applied to the problem of bond length alternation in the Pariser-Parr-Pople model of cyclic polyenes CN HN, N = 4v + 2, which may be regarded as a simplified model of polyacetylene. Both the Møller-Plesset and the Epstein-Nesbet-type partitionings of the model Hamiltonian are employed. The localized orbital basis enables an efficient truncation of the perturbation theory summations over the intermediate states as well as an elimination of energetically unimportant diagrams, thus enabling one to obtain the fourth-order Møller-Plesset-perturbation energies with a relatively small computational effort even for large polyenes. The results obtained with the second-, third-, and fourth-order Møller-Plesset and with the third-order Epstein-Nesbet perturbation theories yield very similar bond length distortions (about 0.05 Å) and stabilization energies per site (about 0.04 eV) as obtained earlier with the RHF, one-parameter AMO, and delocalized orbital perturbation theories. The effects of truncation and diagram elimination in the fourth-order Møller-Plesset perturbation theory and the abnormal behavior of the second-order Epstein-Nesbet perturbation theory results in the localized Wannier basis near the instability threshold of the RHF solutions are discussed.
