Notes: The 4-benzylidene-3-oxo[1,3]oxathiolan-5-ones 13-15, which were derived from the 3-oxo[1,3]oxathiolan-5-ones 9-11 by Knoevenagel condensation with the aldehydes 12, cyclize in an intramolecular hetero Diels-Alder reaction with high yield and excellent endo/exo as well as induced diastereoselectivity to give the hetero Diels-Alder adducts 16-18. The preferred formation of the Knoevenagel products 13-15 with a (Z) configuration was predicted with DFT calculations (B3LYP-6-311+G*) using the model systems 28 and 29. In addition B3LYP-6-31G*/sB3LYP/3-21G(*) calculations on transition structures for the hetero Diels-Alder reaction of 29 and 30 allowed a good correlation with the experimental results, which show that an endo attack of the dienophile syn to the S-O group in 13-15 leads to the main products.

Notes: -Several new approaches for the enantioselective synthesis of the chromane moiety of vitamin E are described. Sonogashira coupling of 3a with the alkyne 4 and subsequent elimination gave 6, which was bis(hydroxylated) in 93% yield and with 85% ee. Recrystallization gave enantiopure 7a, which was hydrogenated and transformed into the vitamin E precursor 11. The bis(hydroxylation) of 18 and 21 to give 9 and 22, respectively, was less than satisfactory, proceeding with ee values of 28% and 18%. In contrast, stereoselective allylation of ketone 15 followed by removal of the protecting group or ozonolysis of the allyl moiety furnished the allyl alcohol 26 and the aldehyde 27, respectively, in almost enantiopure form, which again could be used as precursors for vitamin E. Partial hydrogenation of 5a gave the alkene 32a and that of 28 the alkene 30b, both of which show interesting atropisomerism.

Notes: Intramolecular Lewis or Brønsted acid-catalyzed cyclization reactions of steroid arylimines 6 yielded either tetrahydroquinolines condensed to the estrane skeleton 9 or N-arylamino-D-homosteroids 12-16, depending on the substituent of the arylimino group.

Notes: An efficient three-step biomimetic synthesis of the four diastereomeric 18,19-dihydroantirhines 4a-d starting from the tetrahydrocarboline aldehydes 5a and 5b is described. Domino reaction of the aldehydes 5a and 5b, respectively, with Meldrum's acid (6) and the enol ethers 8a and 8b in the presence of catalytic amounts of ethylenediammonium diacetate leads to the strictosidine analogues 10a-d and 11a-h, respectively, in 74-86% yield, hydrogenation of which gives mainly 15 and 16 with the skeleton of the vallesiachotamine indole alkaloids (55-86%). In addition, small amounts of 17 and 18 with the corynanthe skeleton are also formed (10-12%). Reduction of both 15 and 16 with LiAlH4 yields the diastereomeric rac-18,19-dihydroantirhines 4a-d in 78-86% yield.

Notes: Enantiopure homoallylic alcohols 5, which are important building blocks in organic synthesis, are obtained with an ee of greater than 99% and a yield of 75-95% by cleavage of the secondary homoallylic ethers 4 using sodium in liquid ammonia. The ethers 4 are formed with excellent diastereoselectivity and in 52-89% yield by treatment of the aldehydes 1 with the trimethylsilyl ether of N-trifluoroacetylnorpseudoephedrine (2) in the presence of a catalytic amount of TMS triflate or TMS borontriflate, followed by addition of allylsilane 3. Nearly all achiral aliphatic aldehydes employed gave a diastereoselectivity of over 99:1. With the chiral aldehydes 24, the difference between matched and mismatched pairs was low; this reveals that there is strong reagent control.

Notes: The stereodivergent asymmetric hetero-Diels-Alder reaction of achiral and chiral 1-oxa-1,3-butadienes carrying an oxazolidine moiety with various enol ethers in the presence of different Lewis acids is described as a highly stereoselective and efficient approach to dihydropyrans, which can be used for the synthesis of carbohydrates. In the cycloaddition of the achiral oxabutadiene very good endo/exo selectivity was possible, and with the chiral oxabutadienes excellent 1,6-asymmetric induction was additionally observed. In the processes a reversal of facial selectivity occurs by changing the Lewis acid, allowing the synthesis of both enantiomers of the dihydropyrans with the same auxiliary. Thus, cycloaddition of 1 to 2 in the presence of Me2 AlCl gives predominantly the endo product 3 (3:4 = 10:1), whereas with SnCl4 the exo product 4 is obtained (3:4 = 1:15). The reaction of 7 and 1 a in the presence of Me2AlCl as promoter nearly exclusively yields the endo-I adduct 16 a (16 a+17 a:18 a+19 a = 〉 50:1; 16 a:17 a = 60:1), whereas with TMS-OTf the endo-II-product 17 a was obtained as the main component (16 a+17 a:18 a+19 a = 〉 50:1; 16 a:17 a = 1:7.9). The use of SnCl4 leads to a mixture of endo and exo, again, however, with excellent induced selectivity. A similarly good induction was obtained with the oxabutadiene 9 containing the new auxiliary 8. Also, other enol others 1 b-g were used, some of which afforded excellent induction. Mechanistic considerations are used to explain the results.

Notes: The synthesis of the ganglioside LM1 (1) has been performed by glycosidation of 2 with azidosphingosine 3, followed by reduction of the azido group, acylation with stearine anhydride (4), and solvolysis. 2 was obtained from sialyl xanthate 5 and tetrasaccharide 6 in the presence of phenylsulfenyl triflate. The tetrasaccharide acceptor 6 was prepared from the lactose derivatives 7 and 8.

Notes: Intermolecular hetero Diels-Alder reactions of the enantiopure sulfinimine 11 with the enol ethers 12-16 at 20 °C and 11 kbar lead to the tetrahydropyridines 18-22 in high yield, with very good to good endo/exo selectivities, and with an induced diastereoselectivity of up to 2.1:1. The sulfinyl group in the adducts can be removed with MeLi followed by treatment with acetyl chloride or dimethyl sulfate. According to this procedure, 21 gives either N-acetyltetrahydropyridine 29 or the N-methyldihydropyridine 32. The intramolecular Diels-Alder reaction of 26 yields 27, which is transformed into 33 and 34, respectively.

Notes: The facial selective allylation of alkyl methyl ketones 1a-f in the presence of chiral 2-amino alcohol derivatives 2a-p by reaction with allylsilane 3 and a catalytic amount of TfOH to give the tertiary homoallylic ethers 8a-o and 9a-e is described. The best results were obtained with the 2-amino alcohol derivative 2p which affords a stereoselectivity of 18:1 even for the allylation of ethyl methyl ketone. The ethers 8 and 9, which contain a phenyl group at C-1 of the amino alcohol moiety, can be cleaved to give the corresponding homoallylic alcohols 5 by reduction with sodium or lithium in liquid ammonia.
