    Notes: Summary Protoplasts were isolated enzymatically from suspension cultures derived from embryogenic calli induced by leaf base culture of Iris germanica. In protoplast culture, the effects of glucose concentration, different sugars and combinations of 2,4-D and KIN on protoplast division and colony formation were examined. N6 medium supplemented with 0.1–1 mg/l 2,4-D, 1 mg/l KIN, 200mg/l casein hydrolysate, 250 mg/l proline, 0.2 M glucose and 20 g/l agarose was suitable for protoplast division and colony formation. When colonies formed were transferred onto hormone-free MS medium, many plantlets were regenerated through somatic embryogenesis. Thus, we could establish a plant regeneration system from protoplasts of I. germanica.

    Notes: Dense sands, overconsolidated clays and soft rocks exhibit strain-hardening and strain-softening behaviour in a certain range of confining pressure. The aim of the present paper is to construct a constitutive model of soft rock that can describe not only the strain-hardening behaviour, but also the strain-softening behaviour. An elasto-plastic constitutive model for soft rock is derived by introducing a stress history tensorThe preliminary idea was first reported in the reference, the conference paper16. This paper is a complete version of the theory with interpretations and experimental validations. The model is evaluated through a comparison of the calculated results and the experimental results of tests on sedimentary soft rock (porous tuff). In addition, the applicability of the model to numerical analyses is discussed in relation to the uniqueness of the solution in initial and boundary value problems.

    Notes: A two-dimensional consolidation analysis of clay deposits was made with an elasto-viscoplastic constitutive model and Biot's consolidation theory. One- and two-dimensional consolidation problems were analysed numerically by the finite element and finite difference methods. Results show that the proposed method can describe the effect of sample thickness and aging on consolidation phenomena. The two-dimensional behaviour of a clay foundation during the construction of embankments also was simulated.
