    Notes: Abstract The Fourier transform infrared spectra and subtraction spectra of poly C film in the 4000 to 800 cm−1 region at 22°C were measured systematically as a function of relative humidity (from 0% to 100%) at static equilibrium. The peak position and intensity changes of infrared bands prove that the hydration of poly C has happened mainly on the-NH2 and C=O group of cytidine base, phosphate and ribose ring. The ~1664 cm−1 peak (NH2) and the ~1655 cm−1 peak (C=O) show the strongest variation in intensity starting from R.H. 〈12%. In comparison with the hydration of poly A which we studied, it indicates: (1) The polynucleotide molecules are surrounded by hydration water at their active sites. (2) Poly C can more easily be hydrated than poly A, since the polar group C=O of cytidine plays an important role during hydration.
