    Notes: Abstract The expression of human placental-type glutathione S-transferase (GST-π) was investigated in human cutaneous squamous cell carcinoma (SCC) and normal skin using Northern blot and immunohistochemical analysis. In Northern blot examination, the expression of GST-π transcript was recognized in all instances, and SCC showed a significantly higher expression of GST-π than normal skin. In immunohistochemical examination, GST-π was stained well in the cytoplasm of all cells of the stratum granulosum, many cells of the stratum spinosum and a few cells of the stratum basale in normal skin. Some cells of the stratum spinosum and almost all cells of the stratum basale showed only a weakly positive or almost negative reaction for GST-π. No nuclear staining of GST-π was obvious in normal epidermal cells. In SCC, many cells showed strong positivity for GST-π in the cytoplasm, and some were obviously accompanied by nuclear staining of GST-π. These findings suggest that GST-π exists mainly in many cells in the upper layers of the normal epidermis and that GST-π is involved in the process of carcinogenesis.

    Notes: Abstract  The histogenesis of phyllodes tumour (PT) and that of fibroadenoma (FA) of the breast appear to be closely related. FA is thought to be hormonally responsive, while the hormone-responsiveness of PT is uncertain. To gain insight into hormone-responsiveness of PT, we performed immunohistochemical analysis of oestrogen-regulated pS2 and androgen-regulated prostate-specific antigen (PSA) protein expression and also of oestrogen receptor (ER), progesterone receptor (PgR) and androgen receptor (AR) expression in paraffin sections obtained from 50 female PT patients. Paraffin sections taken from 50 female fibroadenoma (FA) patients were analysed for comparison. ER, PgR, pS2, AR and PSA expression were detected in 32%, 96%, 20% 98% and 4.0% of PT sections and in 28%, 96%, 42% 80% and 10% of FA sections, respectively. No correlations were detected among ER, PgR and pS2 expression or between AR and PSA expression in PT or FA sections. PgR expression was significantly associated with AR expression in PT (P〈0.0001). The present investigations indicate that PT and FA have almost similar hormone receptor status. However, different positivities of pS2 expression suggest that oestrogen-responsiveness may differ between PT and FA. In addition, a wide-ranging co-expression of AR and PgR in PT sections suggests that these receptors may play an important part in the proliferation, although the functional significance of these receptors should be elucidated.

    Notes: Abstract To clarify whether p53 protein expression is involved in multistep carcinogenesis or the progression of mammary ductal carcinoma, we investigated p53 protein expression in 83 invasive ductal carcinomas (IDC), 10 IDC with a predominant intraductal component, 13 non-invasive ductal carcinoma (NIDC), 16 atypical ductal hyperplasia (ADH) and 39 benign epithelial hyperplasia (EH), using immunohistochemistry. Expression of p53 protein was detected in 24 (28.9%) cases of IDC, 5 (50%) cases of IDC with a predominant intraductal component and 1 (7.6%) case of NIDC. No expression was observed in either ADH or EH. In IDC, including cases with a predominant intraductal component, p53 protein expression was associated with a higher histological grade (P〈0.0001) or mitotic index (P〈0.0005). Although overexpression of c-erbB-2 protein has also shown a similar association with these prognostic indicators, expression of p53 protein correlated regardless of the status of c-erbB-2 overexpression. Completely coordinated expression of p53 protein was seen in both intraductal and invasive components. The intraductal component in IDC including cases with a predominant intraductal component which expresses p53 protein had significantly higher histological grade (P〈0.0005) or more comedo-subtypes (P〈0.0001). These results suggested that p53 protein expression occurs at a stage of NIDC with high histological grade or in comedo-subtypes. Its expression is maintained throughout invasion.

    Notes: In order to extend the application of grafting for the modification of natural polymers, the graft polymerization of methyl methacrylate onto gelatin by radical initiators was studied in aqueous solution at temperatures between 60°C and 80°C. Among the initiators used (peroxy-sulfates, α,α′-azobisisobutylonitrile, and benzoyl peroxide), potassium peroxysulfate was found to be the most efficient initiator in this particular graft polymerization. From the kinetic data with this initiator, it was shown that (1) efficiency of grafting is higher at lower temperature; (2) a sharp increase in the efficiency of grafting occurs at the later period of the polymerization at high temperature, which is attributable to the combination between homopolymer and backbone gelatin; and (3) generally, the number of branches was small and the molecular weight of the branch polymer was high in this polymerization.

    Notes: Polyurethanes with various NCO/OH ratios and kraft lignin contents were synthesized by polymerization of kraft lignin (free of the low and high molecular weight fractions), a propylene-oxide-based polyether triol and polymeric MDI in tetrahydrofuran solution. Films were made by solvent casting and tested with respect to swelling behavior and tensile properties. The main findings were: At low NCO/OH ratios, kraft lignin contributed effectively to the formation of the three dimensional network; under particular conditions of NCO/OH ratio and kraft lignin content, polyurethanes of considerable toughness were obtained; at high kraft lignin contents, the obtained polyurethanes were hard and brittle regardless of the NCO/OH ratio used.
