    Notes: Abstract The effect of gibberellic acid (GA3) on ion ratios in a dwarf maize mutant (Zea mays L. d1) exhibiting normal growth after hormone treatment has been investigated by electron microprobe analysis. Gibberellic-acid treatment increased the ion content in chloroplasts and vacuoles whereas no change of the ion content was found in the cytoplasm. The relation of these observations to the action of the hormone is discussed.

    Notes: Abstract A 68-kDa heat-stress protein (HSP68) has been purified from cell-suspension cultures of tomato (Lycopersicon peruvianum L.). Antibodies raised against HSP68 cross-react with the Escherichia coli heat-stress protein DnaK. HSP68 was found to be a hydrophilic, ATP-binding protein. Immunological analysis of subcellular fractions and immunogold-labelling of ultrathin sections showed consistently that HSP68 is localized in the mitochondrial matrix. In-vitro translation experiments indicated that HSP68 is synthesized as a precursor protein. Immunoscreening of cDNA libraries from tomato and potato (Solanum tuberosum L.) led to the isolation of corresponding cDNA clones. The deduced amino-acid sequences show strong relationships to the DnaK-like proteins from bacteria and organelles of eukaryotic cells. The protein HSP68 is constitutively expressed, but its synthesis is increased during heat stress in all cells of higher plantes investigated so far.

    Notes: Abstract Cell cultures of Lycopersicon peruvianum L. stressed with CdSO4 (10−3M) show typical changes in the ultrastructure, starting with the plasmalemma and later on extending to the endoplasmic reticulum and the mitochondrial envelope. Part of the membrane material is extruded, with the formation of osmiophilic droplets which increase in size and number during the stress period. After 4 h, about 20‰ of the cells are dead. A short heat stress preceeding the heavy-metal stress induces a tolerance effect by preventing the membrane damage. The cells show a normal ultrastructure with one exception: cytoplasmic heat-shock granules are formed. This protective effect can be abolished by cycloheximide. Cadmium uptake is not markedly influenced by the heat stress. Cadmium is found together with sulfur in small deposits in the vacuoles of stressed cells. The precipitates contain an excess of sulfur, evidently due to the stress-induced formation of phytochelatins. The role in heavy-metal tolerance of heat-shock proteins in the plasmalemma (HSP70) and in cytoplasmic heat-stress granules (HSP17, HSP70) is discussed.
