Notes: Abstract  The intrathecal synthesis of borrelial-specific IgM- and IgG-antibodies was studied in 67 patients with neuroborreliosis and in 14 patients with neurosyphilis (controls). Antibody concentrations in serum and in the cerebrospinal fluid were determined by an enzyme immunoassay (EIA) using, as antigens, a sonicate of Borrelia burgdorferi, the recombinant 14 kDa flagellin fragment, the outer surface protein C (22 kDa), and the high molecular mass protein p83 (83 kDa). In the sonicate EIA, IgG- and/or IgM-antibodies to Borrelia burgdorferi in serum were detected in all patients with neuroborreliosis and in 71% of patients with neurosyphilis. Intrathecal synthesis of borrelial-specific IgG- and/or IgM-antibodies was demonstrated in 82% of patients with neuroborreliosis and in 71% of patients with neurosyphilis. Immunoglobulin G- and/or IgM-antibodies in serum against any of the recombinant antigens were detected in 92% of patients with neuroborreliosis and in none of those with neurosyphilis. Intrathecal synthesis of IgG- and/or IgM-antibodies to individual recombinant antigens was demonstrated in 67% of patients with neuroborreliosis and in none of those with neurosyphilis. The sensitivity of the recombinant antigens in serum was almost equal to that of the sonicate EIA, whereas the recombinant antigens were clearly less sensitive in the estimation of the intrathecal specific immune response. It was concluded that in suspected cases of neuroborreliosis, the estimation of high specific antibodies in the recombinant EIA will be helpful in confirming the diagnosis.

Notes: Abstract Neuroborreliosis, a manifestation of infection with the spirochete Borellia burgdorferi, has become the most frequently recognised arthropod-borne infection of the nervous system in Europe and the USA. The best criterion of an early infection with B. burgdorferi is erythema migrans (EM), but this is present in only about 40–60 % of patients with validated borreliosis. Therefore use of the duration of the disease as a classification criterion for neuroborreliosis is increasing, the chronic form being distinguished from the acute when symptoms persist for more than 6 months. The diverse manifestations of neuroborreliosis require that it be included in the differential diagnosis of many neurological disorders. In Europe, meningopolyradiculoneuritis (Bannwarth’s syndrome) represents the most common manifestation of acute neuroborreliosis, with the facial nerve being affected much more frequently than the other cranial nerves. Clinical symptoms affecting the central nervous system are rarely observed and then mostly in chronic courses. By far the most common manifestation of chronic neuroborreliosis is encephalomyelitis with spastic-ataxic disturbances and a disturbance of micturition. The current diagnosis of neuroborreliosis is a clinical one, which has to be confirmed by laboratory testing. In most patients, examination of the cerebrospinal fluid (CSF) reveals lymphocytic pleocytosis, damage to the blood-CSF-barrier and an intrathecal synthesis immunoglobulin (Ig) M, IgG, and sometimes IgA. Confirmation of a borrelial infection of the nervous system requires demonstration of an intrathecal synthesis of borrelial-specific antibodies in the CSF or detection of borrelial DNA in the CSF by polymerase chain reaction (PCR). There is no generally accepted therapeutic regime for the treatment of neuroborreliosis, but recent studies have shown ceftriaxone 2 g/day and cefotaxime 6 g/day to be effective in acute and chronic courses. Penicillin G 20 mega units/day and doxycycline 200 mg/day may be suitable for uncomplicated meningopolyneuritis, without involvement of the central nervous system. The duration of treatment – at least 2 weeks in the acute forms and 3 weeks in the chronic forms of neuroborreliosis – is very important for successful treatment. Corticosteroids are recommended only for patients with severe pain that does not respond to antibiotics an analgesics.

Notes: Saturated fluorocarbon elastomers are very resistant to chemical reactions, including those of a crosslinking nature. In order to study proper methods of preparing these materials, Viton GLT was mixed with various levels of trimethylol propane trimethacrylate, TMPTM, and triallyl isocyanurate, Diak #7. After molding, the materials were subjected to electron beam irradiation. The glass transition, rubbery modulus, viscoelastic behavior, and equilibrium swelling were used to characterize the materials as a function of crosslinker and irradiation level. In general, the modulus and gel fraction increased with both crosslinker content and irradiation level. Mixtures of both crosslinking monomers produced an unexpected synergism, yielding higher tensile strength, and earlier onset of gelation.

Description / Table of Contents: Summary A 20-year-old man without vascular risk factors presented with paraesthesia of the left side of the body with acute onset. Cerebral magnetic resonance imaging showed an infarction in the right thalamus. Intra-arterial digital subtraction angiography revealed stenosis of the right thalamic vessels. Recent infection by Borrelia burgdorferi was demonstrated by typical findings in the cerebrospinal fluid: lymphocytic pleocytosis and intrathecal synthesis of borrelial-specific antibodies. The diagnosis of a borrelial-induced vasculitis with secondary thalamic infarction was made from these findings. After antibiotic treatment with cefrtriaxone, the patient was discharged without residual complaints.

Notes: The transverse NMR relaxation times of hydrogen nuclei of water absorbed in white spruce sapwood [Picea glauca (Meunch) Voss] were measured for moisture contents in the range from 5 to 176%. The spin echo amplitudes resulting from the Carr-Purcell sequence decay nonexponentially suggesting the possibility of at least two different relaxation times for water in wood. A simplified structural model of the wood-water mixture is used to estimate the rates of chemical exchange at room temperature of hydrogen nuclei between various sites in the system. The high-resolution NMR line shape is discussed briefly in terms of this proposed model.

Description / Table of Contents: Saturated and unsaturated oligomers are formed by a side reaction during the thermal polymerization of styrene. MAYO has suggested that these oligomers are closely associated with the initiation step.Recently the structures of the dimer fraction were published by BROWN. By independent synthesis of the dimers we could confirm his results. The main components of the dimer fraction are trans- and cis-1.2-diphenylcyclobutane in a ratio of 3:1 ; also smaller amounts of 1.3-diphenylbutene-(3) and I-phenyltetralin were found. The aim of this work was to clarify the structures of the trimers. The trimer fraction contains about 30% of 1.3.5-triphenylhexene-(5), the structure of which was confirmed by independent synthesis. Furthermore we were able to show, that about 65% of the trimer fraction consists of the 4 optical inactive isomers of 1-phenyl-4-[1′-phenyläthyl-(1′)]-1.2.3.4.-tetrahydronaphthalene. Their structures were confirmed by dehydrogenation to 1-phenyl-4-[1′-phenyläthyl-(1′)]-naphthalene, which was identical to an independently synthesized sample.

Notes: Somatostatin (SS) and two glucagon selective analogs [D-Cysl4]-SS and [D-Trp8, D-Cys14]-SS have been synthesized in gram quantities by the solid-phase procedure. A general modification of Monahan and Gilon's procedure for esterification of the first protected amino acid onto the chloromethylated resin as well as a general protocol for solid-phase peptide synthesis on Beckman 990 automatic synthesizer are described. A new general procedure for disulfide formation, which involves the adaptation of the “high-dilution” principle to the ferricyanide oxidation and the optimization of the sequence of purification steps as applied to somatostatin and its analogs, yields highly purified peptides (≥ 199% pure) as checked by reverse-phase high-pressure liquid chromatography - which is shown to be a highly sensitive, resolutive, and quantitative analytical tool for evaluation of the homogeneity of peptides.
