    Notes: Abstract Mitochondrial DNA fragments of two nearly isogenic lines of sugar beet (Beta vulgaris L.) were amplified by RAPD analysis. A number of fragments, most of them unique to either the male-sterile or the male-fertile cytoplasm, were selected for cloning and sequencing. One fragment was present in the PCR fingerprint pattern of both cytoplasms, whereas five of the selected fragments were specifically amplified from only one type of cytoplasm. The mitochondrial origin of all cloned RAPD fragments was confirmed by Southern hybridization. One fragment resulted in a hybridization pattern that suggests its repetitive presence in the mitochondrial genome of sugar beet. Four out of the five cytoplasm-specific RAPD fragments were shown to hybridize specifically to one type of cytoplasm only. One fragment hybridizing with the mtDNA from N-cytoplasm also revealed hybridization signals with both total and nuclear DNAs of N- as well as S-cytoplasm. Sequence alignments of this clone showed strong homologies with a part of the plastidal ndhC gene of higher plants, indicating that the male-fertile-specific mtDNA RAPD fragment is derived from chloroplast DNA. Sequence analysis of an amplified sterile-specific fragment revealed the presence of an open reading frame of 288 bp. Northern hybridization showed a transcription signal specific for the male-sterile cytoplasm. No sequence homology of the open reading frame to any known sequences was found. The results reveal an extremely high degree of sequence variability between the mtDNA of the N- and S-cytoplasm of Beta vulgaris.

    Description / Table of Contents: Zusammenfassung Bei 121 Patienten mit isolierten, nicht resezierbaren colorectalen Lebermetastasen wurde eine intraarterielle regionale Chemotherapie mittels implantierbarer Pumpen oder Infusionskammern durchgeführt. In unterschiedlicher Dosierung und Applikationsart erfolgte die Therapie mit FUDR allein oder in Kombination mit 5-FU und Mitomycin C. Eine partielle Tumorremission wurde in 35–71% beobachtet. Die mediane Überlebenszeit seit Therapiebeginn betrug bisher 8-16 Monate. Extrahepatische Metastasen traten in 45%, technische Komplikationen in 23% (Pumpe) und 46% (Infuse-à-port) auf. Die lokale Toxizität war abhängig von der Dosierung und dem Applikationsmodus. Diese Ergebnisse stimmen mit denen von 647 Patienten aus verschiedenen deutschen Kliniken und einer Sammelstatistik der amerikanischen Literatur (n = 490) überein.
    Notes: Summary Intraarterial regional chemotherapy for non-resectable, isolated colorectal metastases to the liver was performed in 121 patients via implantable pumps or infusea-ports. FUDR alone or in combination with 5-FU and Mitomycin C was administered in different modalities. Partial response rate ranged between 35 and 71%. Median survival since starting chemotherapy was so far 8-16 months. Extrahepatic relaps occurred in 45%, technical complications in 23% (pump) and 46% (infuse a port). Local toxic side effects strongly depended on drug dosage and application modus. These results corresponded with those of 647 German collective cases and 490 cases from American literature.

    Notes: Abstract The technique of random amplified polymorphic DNA (RAPD) offers a broad range of applications in the investigation of plant genomes. A promising prospect is the use of RAPD products as genetic markers. We have investigated a possible organellar source of fragments in RAPD patterns of total DNA. Two nearly-isogenic lines of cytoplasmic male-sterile and male-fertile sugar beet (Beta vulgaris L.) were subjected to RAPD analysis with six different primers. Total, nuclear, mitochondrial (mt), and chloroplast (cp), DNA from each line were investigated. Reproducible DNA fingerprints could be obtained from both organellar DNAs. Differences in band patterns of mtDNA between cytoplasmic male-sterile and -fertile lines were observed with five out of six primers, whereas different cpDNA patterns were generated by one of the primers. Consequently, the RAPD technique can be used to discriminate between different cytoplasms. Clear evidence is provided for the organellar origin of fragments in genomic (total DNA) RAPD patterns. The consequences of these results for the interpretation of RAPD analyses are discussed.

    Description / Table of Contents: Zusammenfassung Die Wertigkeit der intraarteriellen Chemotherapie von Lebermetastasen speziell mit implantierbaren Applikationssystemen ist statistisch noch nicht gesichert. Die kontrollierte Behandlung von 29 Patienten in den letzten 2 Jahren jedoch ist ermutigend: 86% Ansprechrate (CT, CEA), Lebenszeitverhingerung 8 Monate bei fortgeschrittenem Tumorstadium, fehlende systemische Beeinträchtigung, geringe Komplikationen bei standardisiertem Verfahren. Vor der endgültigen statistischen Klärung sollte die Methode nur bei detaillierten methodischen Kenntnissen, bei Klassifizierung der Erkrankung (Frankfurt-Klassifikation) und unter Studienbedingungen angewandt werden. Eine multizentrische randomisierte Studie wurde begonnen.
    Notes: Summary The advantage of intra-arterial chemotherapy of liver metastases, especially with implantable pumps, has not yet been statistically proven. However, controlled FUDR treatment of 29 of our own patients during the last 2 years has been encouraging: an 86% response rate (CT, CEA), prolongation of the survival time by 8 months in advanced tumor stages, lack of systemic interference, and a low complication rate in a standardized procedure. Prior to final statistical evaluation the method should only be used with technical expertise as a prerequisite, classification of the disease (Frankfurt class), and under controlled conditions. A randomized multicenter trial has been started.
