    Notes: Abstract The apparent molal volumes (φv) and adiabatic compressibilities [φK(S)] of La2(SO4)3 solutions have been determined from density and sound speed data at 25°C. The large positive deviations of φv and φK(S) of La2(SO4)3 from the limiting law have been attributed to the formation of the ion pair LaSO 4 + . The observed values of φv and φK(S) have been used to estimate the change in the apparent molal volume and adiabatic compressibility for the formation of LaSO 4 + from $$\Delta \phi (LaSO_4^ + ) = [\phi (obs.) - \phi (2La^{3 + } ,3SO_4^{2 - } )]/\alpha$$ where ϕ(2La3+, 3SO 4 2− ) is the apparent molal volume or adiabatic compressibility of the free ions, and α is the degree of association. The value of $$\Delta \phi _v^o (LaSO_4^ + ) = \Delta \bar V^o (LaSO_4^ + ) = 22.8 \pm 1cm^3 - mole^{ - 1}$$ and $$\Delta \phi _{K(S)}^o (LaSO_4^ + ) = \Delta \bar K_S^o (LaSO_4^ + ) = 85 \pm 20 \times 10^{ - 4} cm^3 - mole^{ - 1} - bar^{ - 1}$$ at infinite dilution are in reasonable agreement with the values determined from the high-pressure conductance data of Fisher and Davis. The number of hydrated water molecules (ca. 11) associated with the formation of LaSO 4 + determined from the volume and compressibility data are in good agreement.

    Notes: Abstract Density data for dilute aqueous solutions of RbNO 3 and TlNO 3 at 25°C lead to the following equations for the apparent molal volumes: RbNO3: ΦV = 43.07 + 2.46c 1/2 + 0.15c (0−0.41M) TINO3: ΦV = 39.50 + 10.83c 1/2 − 25.46c + 26.0c 3/2 (0.03−0.25M) From these expressions and a semiempirical equation for Φv of a completely dissociated 1:1 electrolyte, values of ΔΦv for ion pair formation at c=0.10 are calculated to be+5 ml-mole−1 for RbNO 3 and+12 ml-mole−1 for TlNO 3 . Comparison of ΔΦv with the electrostriction volume effect suggests that virtually all of the electrostriction is removed in the formation of the TlNO 3 ion pair but only about half with RbNO 3 . Values of ΔΦv are in poor agreement with those estimated from electrostatic theories of ion pair formation.

    Notes: Abstract In recent years, a number of workers have studied the stability of deep lakes such as Lake Tanganyika, Lake Baikal and Lake Malawi. In this paper, the methods that can be used to determine the effect that the components of lakes have on the equation of state are examined. The PVT properties of Lakes have been determined by using apparent molal volume data for the major ionic components of the lake. The estimated PVT properties (densities, expansibility and compressibilities) of the lakes are found to be in good agreement with the PVT properties (P) of seawater diluted to the same salinity. This is similar to earlier work that showed that the PVT properties of rivers and estuarine waters could also be estimated from the properties of seawater. The measured densities of Lake Tanganyika were found to be in good agreement (± 2 × 10-6 g cm-3) with the values estimated from partial molal properties and the values of seawater at the same total salinity (ST = 0.568‰). The increase in the densities of Lake Tanganyika waters increased due to changes in the composition of the waters. The measured increase in the measured density (45 × 10-6 g cm-3) is in good agreement (46 × 10-6 g cm-3) with the values calculated for the increase in Na+, HCO3 -, Mg2+, Ca2+ and Si(OH)4. Methods are described that can be used to determine the conductivity salinity of lakes using the equations developed for seawater. By combining these relationships with apparent molal volume data, one can relate the PVT properties of the lake to those of seawater.

    Notes: Abstract The densities and the sound speeds of aqueous NaHCO3 and Na2CO3 solutions were measured from 0.05 to 1.0m and from 0 to 45°C. These data were fitted to functions of molality and temperature and were used to calculate the apparent molal volumes Vϕ and compressibilites κφ of these solutions. Polynomial expression for Vϕ and κφ as functions of molality and temperature have been determined. The partial molal volumes and compressibilities of these solutions and literature data have been used to determine the volume ΔV and compressibility Δκ changes for the ionization of carbonic acid in aqueous solutions. These values of ΔV and Δκ have been used to estimate the effect of pressure on the ionization constants for carbonic acid from 0 to 45°C. The calculated pressure coefficients are in good agreement with the measured values.

    Notes: When skeletal, cardiac, and smooth muscle is exhaustively extracted with a protein-unfolding reagent such as 6 M guanidine HCl and a disulfide-reducing reagent such as 5% β-mercaptoethanol, a tissue ghost remains intact and retains the characteristic shape and dimensions of the tissue before extraction. In the case of chicken pectoral muscle, the tissue ghost contains 1% of the original muscle proteins. Guanidine HCl extraction followed by collagenase treatment of glycerol-extracted chicken pectoral muscle releases a clean preparation of elongated structures containing 0.2% of the original protein and representing the covalently cross-linked remnants of the muscle fibers. The material of these muscle fiber ghosts extends throughout the interior of the cell. Antibodies raised against the tissue ghosts of smooth muscle cross-react with glycerol extracted skeletal myofibrils, forming a banding pattern which coincides with the banding pattern observed when myofibrils are reacted with antibodies against titin. Titin, a large and soluble protein found in skeletal muscle, cross-reacts with our antigizzard antibody. However, amino acid analysis of the muscle fiber ghosts indicates that titin cannot be the only subunit of the insoluble polymer, but that one or more proteins with a very high glycine and alanine content and a very low basic and acidic amino acid content must also form part of the covalently cross-linked matrix. The possibility is presented that this matrix may be the basis of the superthin 2-3-nm filaments which have been observed in a variety of cell types.

    Notes: Resection of malignant tumors about the knee, followed by reconstruction of the lower limb by rotating it through an arc of 180 degrees is becoming a popular method of treatment of osteosarcoma in growing children. Following rotation, the rotated ankle functions as the knee joint, powering a custom-made prosthesis. The functional result is superior to above-knee amputation with better propioceptive feedback and the absence of phantom limb pain. The children are able to climb stairs well and participate in running sports.The surgical technique is demanding, requiring a precise knowledge of the surgical anatomy. To be successful, the sciatic, peroneal, and tibial nerves must be preserved. The femoral and popliteal artery may be resected and reconstructed if necessary. The remaining growth of the child is determined so that the rotated ankle can be positioned at the level of the contralateral normal knee.

    Notes: A method for correlative studies of early fertilization events that integrates techniques of intracellular electrophysiological recording, video-imaging, and electron microscopy is described. A key feature of the method is its ability to identify the fertilizing sperm and to record the moment of egg excitation. Since the site of gamete interaction is recognizable throughout all stages of preparation, difficulties associated with locating the site of fertilization and determining specimen orientation for microtomy and electron microscopic examination are eliminated. Virtually all samples yield useful information. An example of interacting gametes fixed 4 sec after initiation of the fertilization potential and serial sectioned is described. The method is applicable to systems other than fertilizing eggs when functional, temporal, and spatial relationships of individual cells need to be correlated with changes in ultrastructure.

    Notes: The perinuclear theca is a novel cytoskeletal consisting of a densely layered lamina that surrounds the nucleus of mammalian sperm. Using antibodies specific for the multiple band polypeptides present in the perinuclear theca of bull sperm, we show that a heterogeneous group of immunological related proteins are present in the sperm heads of other mammals with greatly different morphologies, including guinea pig, hamster, rat, and mouse. In none of the species were identical groups of immunoreactive polypeptides found, although immunoreactive proteins of molecular weights 65,000 to 80,000 were present in the sperm heads of all species examined. Immunoreactive proteins less than Mr 55,000 were prominent in rat sperm heads and mouse sperm; guinea pig, hamster, and rat sperm heads and mouse sperm had one band in common at approximately Mr 50,000. Different immunoreactive proteins were present in isolated sperm tails. The perinuclear theca first appeared in the subacrosomal space of round to elongating spermatids. Later, with the caudal movement of the manchette, the postacrosomal segment of the perinuclear theca was deposited in a cephalad to caudal direction along the sperm nucleus. Concomitantly, the cytoplasmic space between the nuclear envelope and the plasma membrane narrowed such that only the theca occupied this portion of the sperm head. Immunoreactivity accompanied the ultrastructural appearance of the subacrosomal layer and the postacrosomal segment. The periods of spermiogenesis, in which sub- and post-acrosomal components of the perinuclear theca are formed and the morphogenesis of sperm organelles with which these elements are associated, suggest that components of this cytoskeletal structure function to join the acrosome and the postacrosomal plasma membrane to the nucleus.

    Notes: Using colloidal gold tagged ligands we have identified the ultrastructural site of ARIS binding to intact and acrosome-reacted starfish sperm. In intact sperm, colloidal gold conjugated ARIS was specifically localized to a single domain (0.1-0.3μ in diameter) on the plasma membrane. This site was located on the anterior-lateral aspect of the sperm head, that is, just peripheral to the region occupied by the acrosomal vesicle and periacrosomal components. When sperm were labeled with colloidal gold conjugated ARIS, washed to remove unbound label, and then induced to undergo the acrosome reaction, the labeled patch remained associated with the plasma membrane and was positioned just lateral to the acrosomal process. However, when sperm were suspended in labeled ARIS and induced to undergo the acrosome reaction, label was observed along the entire anterior aspect of the sperm head with the exception of the acrosomal process. Labeling along the entire anterior aspect of the sperm head in this case was deemed to be nonspecific and due to binding of colloidal gold tagged molecules to components formerly located within the acrosomal vesicle, as the same pattern was obtained using colloidal gold tagged bovine serum albumin. Quantitative and qualitative aspects of ARIS binding observed here by electron microscopy are in agreement with measured binding characteristics previously reported (Ushiyama et al., 1993a: Zygote 1:121-127; Ushiyama et al., 1993b: J Reprod Dev 39:53-54), and indicate that the site of labeled ARIS binding represents a specific plasma membrane domain occupied by ARIS receptors. © 1995 Wiley-Liss, Inc.

    Notes: Expression of the Cylla cytoskeletal actin gene is a marker of differential gene activation in the aboral ectoderm of the early sea urchin embryo. Gene transfer expreiments have defined a 2,300 nucleotide cis-regulatory domain required for the correct spatial and temporal control of this gene. This domain includes at least 20 sites at which relatively stable DNA-protein complexes form in vitro on reaction with embryo nuclear extracts. We report the nucleotide sequence of the whole regulatory domain and map the sites at which high-specificity DNA-protein interactions occur. These were located initially by gel shift assays carried out on progressive restriction digests of given subfragments of the large regulatory domain and were located more exactly by oligonucleotide gel shift competitions. Eight of the sites of specific interaction are unique within the Cyllla regulatory domain, and the remainder consist of five different sites that occur more than once. We observe some well known sequences also found in regulatory regions of other genes, e.g., “CCAAT” and “octamer” elements. The various sites have been classified regarding putative biological function in other work, and the present studies permit an assessment of the number and complexity of interactions constituting each functional class and of the relative locations of sites of each class.
