    Notes: The ultrastructure of spermatogenesis and of the spermatozoon of Acanthopagrus schlegeli (Sparidae) are described. The testis is of the unrestricted type. Germ cells are surrounded by cyst cells. Spermiogenesis involves conspicuous modifications such as intracellular movements (diplosome and mitochondria migration, nuclear rotation, and depression) and structural changes (chromatin condensation, shape of mitochondria, and loss of cytoplasm). The mature spermatozoon has a spherical nucleus with a deep, axial nuclear fossa, and an unusual notch, shaped like a bow tie. The short midpiece contains four spherical mitochondria and encircles the basal body of the flagellum. It is concluded that the A. schlegeli spermatozoon is of a primitive type, but that it is characterized by a unique feature which may provide a useful systematic character. © 1993 Wiley-Liss, Inc.

    Notes: The present study investigated the ontogeny of 3H-uridine incorporation into RNA as a measure for RNA synthesis in preimplantation porcine embryos from the two-cell stage up to the stage of the newly hatched blastocyst. A total of 568 embryos were cultured in vitro for 3 hr in medium (KRB plus lamb serum) containing 9 μM 3H-uridine. After disruption of cell membranes, RNA was isolated on DEAE cellulose filters, and the radioactivity was taken as a measure for the rate of RNA synthesis. No RNA synthesis was detected at the two-cell stage. From the four-cell to the morula stage, 3H-uridine incorporation per embryo increased about ninefold (P 〈 0.001) in blastocyst stages, the increase between developmental stages was not statistically significant. Hatched blastocysts had the highest genomic activity. On a per cell basis, 3H-uridine incorporation was not different from the four-cell stage up to the zona pellucida-intact blastocyst and amounted to 0.29-0.37 fmol 3H-uridine incorporation/cell/3 hr. In hatched blastocyst, 3H-uridine incorporation per blastomere was increased (P 〈 0.01 compared with younger stages) and amounted to 0.86 fmol 3H-uridine incorporation/cell/3 hr. It is concluded that 1) the rate of uridine incorporation depends on the cell stage in zona pellucida-intact porcine embryos and 2) uridine incorporation per blastomere is significantly increased in hatched blastocysts compared with earlier stages.

    Notes: The shedding of epidermis in this species, as in most amphibia, is periodic. A single outer layer of cells separates nearly simultaneously over the whole body; two hours to ten days may be required for the process. In the first three months of laboratory existence the average frequency of molting was 9 per cent. In nature the frequency was only 2 per cent; this increased during the life in confinement in five months up to 25 per cent. Death in the laboratory was usually preceded by numerous molts.The time of a molt is not normally determined by any one of a number of ordinary external conditions which were modified; but nevertheless several means were found of inducing molting. These were, in order of their efficacy: bandaging with gauze on body or tail, wrapping the body with thread, stitching the body wall, applying hot water, and transplanting skin. Molting frequencies up to 98 per cent may thus be obtained.Since molting is periodic and simultaneous, it is evidently coordinated. Experimental induction requires a latent period of six to ninety-six hours, depending upon the stimulus. Recent skin grafts molt with the host, so that the coordination is probably chemical. The primary significance of molting in all groups of animals is possibly the getting rid of unautolyzable waste materials, i.e., worn epithelial structures.

    Notes: The alimentary tract of the freshwater snail Lymnaea stagnalis, except the salivary glands and the digestive gland, was studied with histochemical and ultrastructural methods. The main focus was on the epithelia. Previous studies have indicated that the digestive gland, where intracellular digestion occurs, plays the major role in the digestion process. The main cell types of the tract are ciliated cells and cells with microvilli. The presence of ciliated cells reflects transport of food particles and faeces. In particular areas, viz. the prooesophagus and the pylorus, the epithelium is almost exclusively composed of ciliated cells. Both cell types show signs of endocytosis, lysosomal breakdown of substances, and storage of reserve materials (glycogen, lipid, protein). These features are very prominent in the crop and in the first part of the prointestine and they would indicate that absorption of energy-rich substances can occur in the whole tract. Ten types of gland cells were distinguished. Their possible functions are discussed on the basis of their histochemical properties and ultrastructure of the secretion products. The observations suggest that some of the cell types produce two different substances, one of which may have enzymatic properties. The epithelium of the gizzard consists of cells possessing stout microvilli, which extend into a special coating produced by these cells. The morphology of the epithelium reflects the function of the gizzard, which is comminution of food particles. In the mid- and postintestine occur areas composed of villi cells that possess an extensive basal labyrinth. The morphology of the cells indicates that they may be involved in absorption of water and ions from the faecal pellets. The whole tract is richly innervated. Numerous neurons occur in the connective tissue and muscle layers. Furthermore, in several areas numerous intraepithelial neurons were observed.

    Notes: The integument of the vermilion-spotted newt contains three kinds of pigment--yellow, red, and black. The yellow is uniform and continuous over the whole body; the red and black are discrete, and are chiefly in large spots. The pattern does not change measurably in an adult individual.Functional changes in contraction of the scattered dorsal melanophores were uncommon. The only efficient single factors found were a long subjection to low temperature (expansion) and injection of pituitrin (contraction).Removal of skin was followed by a rapid mass migration of dermal elements into the wound area. Melanophores were thus furnished to a dorsal wound, and after several months black spots formed from these. Red pigment never regenerated nor migrated into a wound. Yellow pigment was formed in situ after the wound had completely healed.Auto-, homo-, and heterotransplants lost their pigment patterns and were gradually reorganized so as to conform in gross appearance to the surrounding pattern of the individual host's skin. The transformation was never complete in eight months, but new black spots and new yellow pigment eventually formed.The behavior of melanophores during morphogenesis differed for specific areas of the integument. Their movements and aggregation were highly coordinated among themselves.

    Notes: The cells of the follicular epithelium of the ovarioles of the mosquito, Aedes aegypti (L.), and of the stable fly, Stomoxys calcitrans (L.), have been found to be interconnected by cytoplasmic bridges. Cytoplasmic bridges have been reported between sibling gamete cells of both male and female insects, but these bridges have not been previously reported to exist between somatic cells of insects.

    Notes: In both photosynthetic (Pyrodinium bahamense, Gonyaulax polyedra, Pyrocystis Iunula, P. noctiluca, P. fusiformis) and nonphotosynthetic (Noctiluca miliaris) bioluminescent dinoflagellates chemical stimulation can by-pass mechanical stimulation. The effective ions are Ca++, K+, NH4+ and H+. Other chemicals found effective are those implicated in Ca++ transport or binding. There are interspecies differences in degrees of mechanical and chemical stimulability. Photoinhibition of mechanical stimulability is the result of two effects, the first a reduction in total bioluminescence potential and the second a decrease in mechanical stimulability resulting experimentally in a decreased rate of light emission. This latter effect can be reversed with Ca++ ions. Chemicals which bind Ca++ or displace Ca++ can mimic the effects of photoinhibition. The chemical inhibition of mechanical stimulability is also reversed by Ca++ ions. A scheme is proposed which is consistent for all species examined.

    Notes: Cells from thigh muscles of fetal rats proliferate readily in vitro in medium containing homologous adult rat “plasma.” As the donor animals mature, these cells become unable to make DNA and proliferate in “plasma” medium, but retain an ability to proliferate in medium containinig fetal bovine serum (FBS). This age-related loss of the ability to proliferate in “plasma”-medium is due to an increasing need by the cells for an exogenous prereplicative promoter which is found in FBS and a crude preparation of bovine luteinizing hormone. Adult cells (and possibly fetal cells) also require a “cycle-completion” factor which is found in FBS and adult rat “plasma.” The requirements for such external proliferative promoters is completely eliminated by neoplastic transformation in vivo, and neoplastic adult cells isolated from a nickel sulfide-induced mixed rhabdomyo-fibro-sarcoma can make DNA and proliferate in vitro in the complete absence of exogenous growth factors.

    Notes: Mechanically stimulable bioluminescence and photoinhibition of sensitivity to mechanical stimulation in the marine dinoflagellate Gonyaulax polyedra can be mimicked by a number of cations, proportional to the logarithm of their external concentrations. The data are consistent with mechanical stimulability as a membrane depolarization resulting in an increase in H+ions at bioluminescence sites and with photoinhibition as a hyperpolarization of the cell membrane.
