    Notizen: Summary In this study, we have used an α1-adrenergic receptor photoaffinity ligand, 2-[4-(4-azido-3-iodo-benzoyl)-piperazin-1-yl]-4-amino-6, 7-dimethoxyquinazoline (125I-APD), to label covalently the α1-adrenergic receptor in a smooth muscle cell line. Our results indicate that in the absence of light, (125I)APD binds reversibly to a site in the DDT1 MF-2 cell membranes having pharmacological characteristics of an α1-adrenergic receptor. Following incorporation of (125I)ADP into partially purified membranes a single labeled band of protein with a Mr of 81 000 was visualized by autoradiography following sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Incorporation of (125I)-APD into this band was affected by adrenergic agonists and antagonists in a manner consistent with an α1-adrenergic interaction. Prazosin (α1-selective) blocked incorporation of the label into the Mr = 81 000 protein while yohimbine (α2-selective) did not. Of the adrenergic agonists, (−)-epinephrine and (−)-norepinephrine but not (−)-isoproterenol blocked labeling of the Mr − 81 000 protein. We conclude that the ligand binding site of the DDT1 MF-2 cell α1-adrenergic receptor resides in a Mr = 81 000 protein.

    Notizen: Abstract We have shown that glucocorticoids induce the appearance of β2-adrenergic receptors in membranes of the ductus deferens smooth muscle cell line (DDT1 MF-2). A concomitant increase in isoproterenol stimulated adenylate cyclase activity in the absence of exogenously applied GTP was observed as was a significantly increased (p 〈 0.05) sensitivity of the adenylate cyclase system to exogenously applied GTP. However, no significant difference in the maximal velocity of adenylate cyclase between control and steroid treatment was measurable in the presence of sodium fluoride. Induction of β2-adrenergic receptors in DDT1 MF-2 cells is correlated with the presence of steroid receptors (androgen and glucocorticoid) in the cells since estrogens and progesterones had no effect on receptor levels. Finally, utilizing dense amino acid labeling of cells to measure old versus newly synthesized receptor sites by a density shift method, we have documented that glucocorticoid induction of β2-adrenergic receptors involves synthesis of new receptor protein.

    Notizen: The temperature dependence of relative modulus observed in filled thermoset, thermoplastic, and polyelectrolyte salt matrices is explained on the basis of induced stresses produced by the differences in the thermal expansion coefficients of the constituent materials. The analysis is based on the assumption that the modulus of the matrix in a filled polymer is less than that of the unfilled polymer. The temperature dependence of relative modulus is expressed as a function of the difference in thermal expansion coefficients, the volume fraction, the relative modulus in the unstressed state, and mechanical properties of the phases. Agreement is good between the analysis and experimental results for three systems: epoxy and glass, polyethylene and wollastonite, and a polyelectrolyte salt with mica and asbestos.

    Notizen: By the use of simple models of filled plastics, approximate equations are derived for the elongation to break in the case of perfect adhesion between the phases and for the tensile strength in the case of no adhesion between the polymer and filler phases. By combining these equations with equations for the modulus (assuming Hookean behavior) all the stress-strain properties can be derived, including rough estimates of the impact strength, as a function of filler concentration. Among other things, the theory predicts a very rapid decrease in elongation to break as filler concentration increases, especially for the case of good adhesion. It is also predicted for the case of good adhesion that the tensile strength of a filled polymer can be greater than that of an unfilled polymer.

The form of this equation is different from the equations used to predict the properties of composites with one continuous phase and one dispersed phase. A simple model is used to correlate the morphology of systems having two continuous phase with the parameter n of the mixture rule. The connectivity of the phase varies with concentration. The properties, such as elastic modulus, depend primarily upon the modulus of the material with the higher modulus. In general, the properties depend very little on the morphology of the system.

    Notizen: The feasibility of using a copolymer composed of styrene, acrylonitrile, and acrylamide or methacrylamide in the formation of a high-strength thermally and chemically stable polymer concrete for use in geothermal environments has been demonstrated. Specimens produced with the copolymer in conjunction with an aggregate containing sand and portland cement had compressive strengths in the range of 25,000 to 30,000 psi at 20°C and were thermally stable up to ∼240°C. A study of the effect of monomer concentration on the properties of the polymer concrete indicated that the optimum concentration is in the range of 12 to 13 wt. %. Increased monomer concentrations lead to a nonuniform polymer distribution within the composite, resulting in a deterioration of the properties. The optimum properties are obtained when the monomer is used in conjunction with an aggregate containing 70 to 80 wt. % silica sand and 20 to 30 wt. % portland cement. The results from laboratory and field evaluations in progress indicate that the materials can be used for pipe or as protective liners on pipe and vessels in electric generating and direct utilization geothermal processes.

    Notizen: Domains, or areas of inhomogeneities and imperfections, may be made visible in polystyrene by treatments of the plastic with certain liquids and vapors. Different types of domains are revealed by different techniques, depending upon the solvent power of the liquids and vapors. One type of domain reveals itself as craze cracks when polystyrene is first soaked in a nonsolvent such as methanol, and then exposed to the vapors of a solvent such as hexane or benzene. A second type of domain may be revealed by exposing polystyrene to certain mixtures of solvents and nonsolvents which produce a rough, whitened surface on the polystyrene. For the first type of crazing, the size of the domains is influenced by the frozen-in thermal stresses, and the shape of the domains is influenced by molecular orientation. The domains of the second type are due to heterogeneity of molecular orientation and are not found in annealed specimens. A theory has been proposed which explains crack crazing in terms of adsorbed vapors in cracks and areas of imperfections in the polystyrene.

    Notizen: The creep and dynamic mechanical properties of rubbers with a wide range of degree of crosslinking have been correlated with the swelling properties of the rubbers. Both butyl and styrene-butadiene rubbers were studied. Mechanical damping steadily decreases as the crosslinking increases. A damping test could replace the usual swelling tests as a measure of the tightness of the network structure. Lightly crosslinked rubbers have a minimum in the damping-temperature curve while highly crosslinked rubbers do not. Very small degrees of crosslinking dramatically decrease the creep rate, however, even for highly crosslinked rubbers, creep does not completely stop. The Nutting equation often gives a very good correlation between damping and creep rate.

    Notizen: Thermogravimetric (TGA) and infrared (IR) analyses were used to determine the thermal stability of a polymer system consisting of styrene-acrylonitrile-acrylamide-trimethylolpropane trimethacrylate. This study indicates that increases in curing temperature and the use of two initiators with different activities decreases the percentages of unreacted monomers. This increases the thermal stability of the polymer. To prove this, dynamic and isothermal TGA were used. The change in typical functional groups found by IR spectra also confirms this observation.
