    Notes: Summary A semi-quantitative analysis of 2-deoxy-(1-14C) glucose (2 DG) incorporation in the supra-chiasmatic nucleus (SCN) was performed in male, castrated female, and castrated oestradiol treated female rats (OVX-E2) during the light period (from 9.00 h to 18.00 h). Unilateral orbital enucleation was carried out on each group. In males and castrated females the 2 DG incorporation in the rostral and caudal SCN was constant throughout the studied period. On the contrary, in OVX-E2 rats, which display a clear cut circadian rhythm of LH secretion, the 2 DG incorporation in the rostral SCN was higher in the afternoon than in the morning. After serotonin (5 HT) depletion, the level of morning labelling reached that of the afternoon. These results suggest the existence of a steroid-linked rhythm of SCN metabolic activity during the light period which might be related to the LH surge and dependent on 5 HT innervation.

    Notes: Summary The uptake of 5-hydroxytryptophan (3H-5-HTP) and serotonin (3H-5-HT) and their distribution have been demonstrated, at the ultrastructural level, in the secretory rudimentary photoreceptor cells (SRP cells), in the pineal organ of the parakeet and the duck. In vivo experiments. When the parakeets were injected with 3H-5-HTP alone or pretreated with drugs interfering with monoamine metabolism (e.g. R04-4602, nialamide) prior to injection of the label, the SRP cells always exhibited specific radioautographic reactions, in the regions of the dense secretory granules of the cell body and asynaptic pedicles. The labeling concentration varied quantitatively with time (interval between injection of the label and sacrifice) and drugs used. In vitro experiments. The uptake of 3H-5-HTP and 3H-5-HT in the pineal organ of the duck has also been studied, using an incubation procedure. The results with the two precursors confirmed those obtained “in vivo” with the parakeet. From these results it is suggested that the uptake of precursors in some compartments of SRP cells is qualitatively independent of: a) the experimental period (experiments performed in winter, spring, and summer; uptake of precursors between 11.00 a.m. and 12.00) b) the precursor (3H-5-HTP or 3H-5-HT) c) the observation times. It is concluded that the SRP cell, an important step in the cytological and functional evolution of the pineal organ (Collin, 1969, 1971), is responsible for the synthesis of 5-HT. The metabolism of 5-HT seems also possible in such cells.

    Notes: Summary The morphological substrate of putative serotonin (5-HT)/neuropeptide Y (NPY) interactions in thé suprachiasmatic nucleus (SCN) was investigated by combined radioautography and immunocytochemistry after intraventricular administration of (3H)5-HT in the rat. In the ventral portion of the SCN, the distribution of (3H)5-HT uptake sites overlapped closely the NPY-immunoreactive terminals. Previous investigations have shown that the dense 5-HT and NPY innervations of the SCN originate in different structures, i.e., the midbrain raphe nuclei and the ventral lateral geniculate nucleus, respectively. Accordingly, in the present study, destruction of 5-HT afferents by 5,7-dihydroxytryptamine was not found to induce any modification in NPY staining and, in ultrastructural immuno-radioautographic preparations, two distinct pools of axonal varicosities could be identified. Both 5-HT and NPY terminals established morphologically defined synaptic junctions, sometimes on the same neuronal target. Some cases of direct axo-axonic appositions between the two types of terminals were also encountered. These data constitute additional criteria for characterizing the cytological basis of the multiple transmitter interactions presumably involved in the function of the SCN as a central regulator of circadian biological rhythms.
