Notes: Summary Ten patients with moderate to severe Raynaud's syndrome were recruited into a four week randomised double blind crossover study to compare the efficacy of UK-38,485 50 mg, a new thromboxane synthetase inhibitor with that of placebo. With the doses used there was no significant difference between the two treatment periods in the number, severity and duration of ischaemic attacks, the mean hand temperatures, forearm and digital blood flow and red blood cell rheology.

Notes: Summary The influence of 2 weeks oral intake of nifedipine (2×20 mg) on the oxidative metabolism of antipyrine was investigated in 12 normal volunteers, who had 1050 mg antipyrine solution orally before and after the course of nifedipine. There were no statistically significant differences in the saliva pharmacokinetic parameters of antipyrine on both occasions. However, the metabolite profile of antipyrine in urine showed a significant reduction in the amount of norantipyrine excreted after compared to that before nifedipine administration (16.5 vs 19.6%). This may have implications for drugs that share a similar demethylation pathway with norantipyrine.

Notes: Abstract In this paper, the Hilbert-space analogue of a result of Huang, that all the methods in the Huang class generate the same sequence of points when applied to a quadratic functional with exact linear searches, is established. The convergence of a class of direct prediction methods based on some work of Dixon is then proved, and these methods are then applied to some control problems. Their performance is found to be comparable with methods involving exact linear searches.

Notes: Abstract In this paper, the convergence of variable-metric methods without line searches (direct prediction methods) applied to quadratic functionals on a Hilbert space is established. The methods are then applied to certain control problems with both free endpoints and fixed endpoints. Computational results are reported and compared with earlier results. The methods discussed here are found to compare favorably with earlier methods involving line searches and with other direct prediction quasi-Newton methods.

It is established that certain classes of these methods are equivalent and their convergence is proved for certain nonquadratic functionals on a Hilbert space. A computational study of these methods applied to a control problem is also included with particular reference to the equivalence of methods mentioned above.
