    Notes: The association of incomplete microtubule assemblies with either another incom-plete structure or complete microtubules was studied in two organisms, the phytoflag-ellate Polytoma papillatum and the phorid fly Megaselia scalaris, using transmission electron microscopy. In the alga, hook-shaped appendages on cytoplasmic and spindle microtubules were detected. These resulted from the lateral association of a curved ribbon of protofilaments with the surface of a complete microtubular wall. In the fly, an S-shaped protofilament sheet was found embedded in the kinetochore plate of a prometaphase I spematocyte. Tracing of the S-shaped element towards the spindle pole revealed that it was formed by the lateral junction of two curved protofilament sheets. In all cases, the C-shaped protofilament sheets represented the endings of complete micro-tubules. Incomplete microtubules are generally considered as representing intermediates of microtubule assembly and disassembly. Since high molecular weight proteins are believed to be responsible for maintaining microtubule-microtubule spacing, it is hypo-thesized that the endings of growing and shrinking microtubules are sparsely studded with these proteins; their depletion allows lateral microtubule contacts. In addition, the microtubule-microtubule contacts may be rendered possible by the flexibility of the slender elongated microtubule-associated molecules. Relatively long C-shaped proto-filament appendages (0.6-1.4 μm) were detected in this study. Therefore, it is plausible to assume that the protofilament sheets are stabilized by contact with one another or with an intact tubule wall.

    Notes: Abstract Spermatogenesis was analysed in a cricket,Eneoptera surinamensis (Gryllidae, Orthoptera), using ultrathin serial sections and transmission electron microscopy. Special attention was placed on documentation of the development and structure of synaptonemal polycomplexes (PCs) within spermatid nuclei. Pachytene spermatocytes showed the usual tripartite synaptonemal complexes in the nuclear lumen. PCs were situated close to chromosomes at the periphery of spindles in prometaphase I spermatocytes, where microtubule density was low. The PCs are probably incorporated into the daughter nuclei of both meiotic divisions by adhesion to chromosomes. Finally, PCs end up within spermatid nuclei. Analysis of serial sections through three nuclei of young spermatids revealed at least one PC within each. The PCs were intimately attached to an electrondense spherical nuclear body. This topographical correlation was confirmed through inspection of random sections. The PCs may have an affinity to the spherical bodies. In more developed spermatids, PCs and nuclear bodies were missing. Disassembly products of the PCs may play a role in spermatid maturation. In a series of other Orthoptera species, PCs have been reported to occur in the cytoplasm or the nuclei of spermatids. In most other systematic groups, PCs do not form at all or disassemble earlier. The presence of PCs in young spermatids, therefore, seems to be typical of Orthoptera.

    Notes: A comparative investigation of testicular eupyrene cysts (in larvae) and apyrene cysts (in pupae) of Ephestia kuehniella laboratory strains was conducted using light and electron microscopy. Eupyrene cysts in the first meiotic division contained 64 spermatocytes, which showed only moderate asynchrony. In one of the strains, a wild-type strain, L, normal-sized cysts occurred together with abnormally large cysts. These are called giant cysts in this article. One of the premeiotic cysts, early giant cysts, studied in detail, contained approximately a fourfold number of cells compared with the number in a eupyrene cyst of the same stage. In cysts harboring spermatocytes and spermatids, late giant cysts, cell differentiation was highly asynchronous.Failure in one of two control mechanisms in early cyst development may have caused the appearance of the cysts. Control of cell division might have been sloppy in apyrene spermatogonia. Hence, the spermatogonia within the cyst could have passed through additional division cycles. Alternatively, the giant cysts may have originated from more than one predefinitive gonial cell enclosed in a common envelope of sheath cells. As a third possibility, giant cysts could have arisen by fusion of normal cysts at a later stage. In either case, this is evidence that separation of eupyrene and apyrene pathways is earlier than was previously expected.In two other Ephestia strains, apyrene sperm development proceeded without formation of giant cysts. One was a mutant strain, a, and the other one was a recently established wild-type strain, Sbr. Apyrene sperm development is considered an example of degenerate evolution in which enhanced variability between species and even between populations of one species is a common phenomenon.
