Notizen: We present results of a model Hamiltonian approach to CO single molecule adsorption on a semi-infinite NiAl(110) surface. CO adsorbs on the Ni rows in agreement with experimental findings. The calculated electron–hole excitation spectrum compares reasonably with direct and inverse photoemission data. The nature of the chemisorption bond is analyzed by partitioning the density matrix and the energy into components which are accessible to physical intuition. The results demonstrate that the d electrons are as important on the alloy as they are on the pure Ni component, although they play a different role. The extended nature of the alloy d wave functions determines the adsorption geometry. The character of the chemisorption bond is discussed in comparison to CO adsorption on the components Al and Ni and to CO/Cu.

Notizen: The interaction of atomic hydrogen with Al(110), Cu(110), Ni(110), and NiAl(110) is investigated using a spin-unrestricted screened Hartree–Fock method. As Al is a pure sp-metal, Cu and NiAl have completely filled d-bands, and Ni provides unfilled d-states, this allows study of the influence of the d-electrons on the hydrogen adsorption mechanism. The band structures of Cu and NiAl are rather similar and we can address the problem of whether the adsorption is dominated by the local character or the band structure. A detailed analysis of the chemisorption bond is given, based on a density matrix partitioning technique. The adsorption energy is found to increase from 1.8 eV via 2.1 eV to 2.4 eV and 3.0 eV in the order Al, NiAl, Cu, Ni in qualitative agreement with the available experimental data. On NiAl(110), hydrogen adsorbs with approximately the same energy above the Ni and Al rows, although the components of the adsorption energy vary significantly. The reduced adsorption energy on NiAl compared to Cu and Ni is traced back to the increased sp-electron density and the more diffuse character of the d-orbitals in NiAl. Adsorption strength is determined by the competing effects of energy gain due to polarization of metal electrons toward the attractive proton potential and energy loss due to Pauli repulsion. Comparison with the mechanism of formation of gas phase chemical bonds reveals a novel mechanism of the chemisorption bond which is uncommon in the gas phase. The long-range interaction with the image potential, the high polarizability, and the spillover of metal electrons play a decisive role in determining the new mechanism of hydrogen chemisorption on metal and alloy surfaces. © 1996 American Institute of Physics.

Notizen: This paper describes the optical design of the general-purpose Spanish beamline for absorption and diffraction (SpLine), which will be installed on a bending-magnet port of the ESRF. This beamline is planned to cover the needs of the Spanish synchroton radiation community with broad scientific fields of interest, covering physics, chemistry, materials science and biology. By using the advantages of a wide front end (9 mrad), the Spanish beamline will be split into two branches. Both branches will be equipped with focusing optics and experimental stations. Thus, each branch can be operated simultaneously and independently from each other. One branch (A) will have two experimental stations, one for high-resolution powder diffraction (HRPD) and the other for X-ray absorption spectroscopy (XAS) and X-ray standing waves (XSW). The other branch (B) will have facilities for macromolecular crystallography and for single-crystal diffraction analysis, including that of surfaces and interfaces, as well as an X-ray diffraction/scattering camera for non-crystalline specimens.

Notizen: Abstract  Bacillus licheniformis MIR 29 has been isolated and produces extracellular proteases. It is able to grow at temperatures up to 60 °C and at pH values up to 9.0. Casein was the best carbon source for production of a thermostable protease activity which, in some conditions, is 90% extracellular. The synthesis of alkaline protease is not constitutive; different levels of production were found with different carbon and nitrogen sources. Casein was thought to be an inducer of enzyme synthesis. The optimal pH and temperature of the enzyme activity were 12 °C and 60 °C, respectively. The enzyme was stable up to 60 °C in the absence of stabilizers. The protease activity was inhibited with phenylmethylsulphonyl fluoride, indicating a serine-protease activity. The proteolytic activity was lowered by molecules present in the culture supernatant, which include amino acids and peptides, indicating end-product inhibition. Electrophoresis assay on denaturating gels showed two bands with alkaline protease activity, in the 25 to 40-kDa molecular mass range.

Notizen: 515 microorganisms of the genus Bacillus which produce starch-degrading enzymes were isolated from natural environments in the course of a screening programme. Treating the Bacillus strains on agar plates with iodine reagent for 30 seconds led to the selection of clones with increased amylolytic activity. This methods is very convenient for the screening of large numbers of amylolytic strains.

Notizen: An acid α-amylase hyperproducing strain, designated as MIR-61, was isolated in a screening procedure from South American soil samples. MIR-61, a 60°C thermoresistant strain, was identified using 98 biochemical and morphological tests and characterized as Bacillus licheniformis by numerical taxonomy. Batch cultures of B. licheniformis MIR-61 showed extracellular α-amylase and α-glucosidase activities during the exponential growth phase. The production of α-amylase was studied at free and constant pH values at 37 and 45°C. Maximum α-amylase activity (4,767 kU/dm3 in a liquid medium) was detected at 45°C at a constant pH (7.0) in the late exponential phase. The α-amylase production by B. licheniformis MIR-61 is 10 to 300 times higher than the enzyme production reported in strains of the same species.Optimum α-amylase activity was found at 50 to 67°C in an acid pH range from 5.5 to 6.0. These properties would allow its use in starch industry processes.

Notizen: α-Glucosidase was detected in four wild-type amylolytic strains belonging to the Bacillus genus. The strains showed α-glucosidase activity in extracellular and membrane-bound fractions. Kinitic studies of the α-glucosidase synthesis in the batch cultures of four strains of the Bacillus genus showed two profiles: partially and totally growth-linked synthesis. The presence of different activities and production profiles of α-glucosidase in the strains at high or low glucose concentrations in the medium would indicate that α-glucosidase may have a role in the regulation of the metabolism of α-polysaccharides.
