Notizen: Activity of acetyl CoA carboxylase was determined in germinating and maturing sunflower (Helianthus annuus L.) cv. Rum Sun Record seeds. A constant decrease was observed in germi nating embryos. Exposure of etioplasts to light intensity of 500 (74 mW . cm−2) lux for different time intervals decreased the activity of the enzyme in the embryos. A reverse trend was observed in ripening embryos where the ATP-Pi exchange rate was found to be directly correlated to the activity of acetyl CoA carboxylase.

Notizen: The polar lipid components in the roots of spinach (Spinacia oleracea) plant were identified as phosphatidylcholine, phosphatidylserine, phosphatidylethanolamine, monogalactosyl-diglycerides, digalactosyldiglycerides, polygalactosyldiglycerides, sulpholipids and three unknown glycosides. Phospholipids constituted only 10% of the polar lipid fraction and 90% was of nonphosphatidic nature. This latter fraction was highly unsaturated and contained linoleic acid as the major fatty acid component.

Notizen: In developing seeds of mustard (Brassica juncea L. cv. RLM 198) the period between 20 and 30 days after fertilization (DAF) was identified as the period of active lipid biosynthesis, although dry matter continued to accumulate until maturity. The period of lipid synthesis was associated with a decrease in starch, soluble sugars and protein, thus, giving rise to precursors for the biosynthesis of lipids. Besides decreasing the dry matter content (on both % and seed basis), Zn and S deficiency caused a significant (P 〉 0.05) reduction in oil content. As compared to control, the decrease in oil content was 11, 12 and 18% at 30 DAF and 4, 9 and 16% at maturity in Zn, S and (Zn+S) deficient treatments, respectively. Throughout the period of seed development, a significant decrease in starch and protein with a slight accumulation of soluble sugars was observed due to deficiency of Zn or S. The rate of [l-14C]-acetate incorporation into total lipids, which was maximal at 30 DAF, also displayed a significant decrease due to the abovementioned mineral deficiencies. Addition of Zn or S in vitro, enhanced the lipid synthesis at all stages of seed development. Under Zn and S deficiency, the phospholipids increased from 10 to 30 DAF and then declined until maturity. However, the proportion of glycolipids and free fatty acids increased, with a corresponding decrease in total glycerides. Further, in deficiency treatments, there was an increase in 22:1 with a corresponding decrease in 18:1, 18:2 and 18:3 in developing and mature mustard seeds.

Notizen: The rates of carboxylation, photophosphorylation and acetate incorporation have been compared in the intact and broken chloroplasts of Hydrilla verticillata Royle leaves in the presence and absence of certain inhibitors and metabolites. The intact chloroplasts showed low rates of photophosphorylation, high rates of carboxylation, and exhibited normal capacity for fatty acid biosynthesis. In broken chloroplasts a drastic decrease was observed in the rates of carboxylation and acetate incorporation. However, the rate of photophosphorylation was considerably increased. In the presence of light, inhibitors such as iodoacetamide, arsenite and sodium azide decreased the photophosphorylation rate. F-1,6-di-P and PGA stimulated CO2 fixation rate. In the absence of artificial light, inhibitors such as sodium arsenite, gluconate-6-phosphate, sodium azide and iodoacetamide decreased the rate of CO2 fixation. CoA, ATP, G-6-P, F-1,6-di-P Stimulated the synthesis of fatty acids. Exogenous supply of ADP. NADH, NADP and NADPH did not stimulate fatty acid biosynthesis probably because these compounds could not gain entry into the chloroplasts. Light was necessary for the in vitro fatty acid biosynthesis.

Notizen: Lipid depletion of berseem seed treated with 100 mg/1 gibberellic acid or maleic hydrazide was studied during germination. During the first 3 days after germination there was little lipid consumption but thereafter the depletion of lipid was rapid. Gibberellic acid was found to enhance the lipid utilization of total lipids and triglyceride components during germination. On the other hand, maleic hydrazide reduced the utilization of triglycerides of the germinating seed. The rate of utilization was rapid for unsaturated fatty acids and slower for saturated ones in treated seeds as compared to untreated seeds.

Notizen: Abstract Qualitative and quantitative analysis of polar lipids in sun flower (Helianthus annuus L.) seed is reported in the present study. Ten classes of polar lipids i.e. phosphatidic acid, phosphatidyl inositol, phosphatidyl choline, phosphatidyl ethanolamine, phosphatidyl glycerol, diphosphatidyl glycerol, digalactosyl diglyceride, monogalactosyl diglyceride, esterified sterol glycoside and free fatty acids have been identified. Phosphatidyl choline and phosphatidyl inositol formed the major phosphatide fractions. Only four fatty acids i.e. palmitic, stearic, oleic and linoleic acids were present in measurable amounts in the total polar lipids. There was a great variation in the fatty acid composition in different classes of polar lipids. Arachidic acid was present in all the individual phosphatide fractions. Phosphatidyl glycerol was highly unsaturated followed by esterified sterolglycoside, phosphatidic acid, phosphatidyl ethanolamine + phosphatidyl glycerol, diphosphatidyl glycerol and monogalactosyl diglyceride.

Notizen: Abstract In the developing peanut (Arachis hypogaea L.) kernels, the period between 15 and 35 days after podding (DAP) was identified as the active period of oil-filling. The period of active oil-filling was associated with a decrease in the starch, soluble sugars and proteins so as to make available the energy and carbon skeleton for the synthesis of oil. The oil content in the mature kernels decreased by 11, 12 and 25 per cent with Zn, S and Zn+S deficiency, respectively. In addition, proteins and starch content decreased significantly while that of soluble sugars increased slightly. The activity of malate dehydrogenase and glucose-6-phosphate dehydrogenase also decreased due to Zn as well as S deficiency. The deficiency treatments resulted in a decrease in phospholipids, free fatty acids and triacylglycerols in mature kernels. Further the proportion of 16∶0 and 18∶2 decreased while that of 18∶1 increased in developing kernels.
