    Notes: Abstract The benefits of using animal or human cell cultures have been clearly demonstrated in diagnostic and therapeutic research and in their application for manufacturing. Cell cultures serve as a tools for the production of vaccines, receptors, enzymes, monoclonal antibodies and recombinant DNA-derived proteins. They represent an integral part of drug development for which corresponding facilities, equipment and manufacturing processes are required. Although the cells themselves offer no particular risk to workers in laboratories and production areas or to the environment, the cell cultures may be contaminated with viruses, mycoplasma, bacteria, yeast and fungi or might contain endogenous viruses. The containment level for animal and human cells is therefore determined by the risk class of these agents. The history of animal and human cell cultures has proved that they can be handled safely. The recommendations in this publication concern the safe handling of cell cultures (tissue explants, primary cell cultures) and permanent cell lines of animal and human origin. A classification system of safety precautions has been elaborated according to the potential for contamination with the pathogenic agents involved.

    Notes: Abstract Treatment efficacy with netilmicin sulphate/methylprednisolone sodium succinate in a severe septic shock baboon model, using an LD100 of liveEscherichia coli, was evaluated. All the animals treated with both netilmicin and methylprednisolone were premanent (≥ 7 days) survivors, whereas none of the untreated baboons lived more than 24 hours. These results indicate that, in a baboon model, netilmicin is an effective alternative to gentamicin (with methylprednisolone) in the treatment of severe septic shock.

    Notes: Abstract Single-source metal precursors designed to form crystalline perovskite phase mixed metal oxides at low temperatures have been synthesized using α-hydroxycarboxylates and alkoxides as ligands. Divalent salts of Ba, Sr, Ca and Pb, [A(O2CCR2OH)2], R = H, Me, were formed from the reaction between ACO3 and HO2CCR2OH in which the hydroxyl group is free to react with metal alkoxide compounds B(OR′)4 where B=Ti, Zr, Sn. These compounds react to eliminate two equivalents of alcohol producing precursors with fixed stoichiometry as shown by the equation: A(O2CCR2OH)2+B(OR′)4 → A(O2CCR2O)2B(OR′)2+2HOR′. The reaction product is only slightly soluble in pyridine, however upon hydrolysis these compounds form clear solutions from which yellow powders can be isolated by removal of the solvent. Thermolysis of these powders in the temperature range 300–350°C yields perovskite ABO3. Aqueous solutions of these powders can also be used to form sub-micron sized particles of ABO3 materials via aerosol processing techniques. In control experiments it was shown that Pb(O2CCH2OPh)2 does not react with metal alkoxide compounds. As a result perovskite PbTiO3 is not formed below 550°C. Solutions for spin coating thin films can be formed by dissolution of the powder in ethanol, but the crystallization behavior is quite different compared to the bulk powders.

    Notes: Abstract Beginning with a formal statement of the conservation of probability, we derive a new differential constitutive equation for entangled polymers under flow. The constitutive equation is termed the Partial Strand Extension (PSE) equation because it accounts for partial extension of polymer strands in flow. Partial extensibility is included in the equation by considering the effect of a step strain with amplitude E on the primitive chain contour length. Specifically, by a simple scaling argument we show that the mean primitive chain contour length after retraction is L=L 0 E 1/2, not the equilibrium length L 0 as previously thought. The equilibrium contour length is infact recovered only after a characteristic stretch relaxation time λ s that is bounded by the reptation time and longest Rouse relaxation time for the primitive chain. The PSE model predictions of polymer rheology in various shear and extensional flows are found to be in good to excellent agreement with experimental results from several groups.

    Notes: Summary IG 1, a temperate phage ofBacillus subtilis, is strongly induced from its lysogens by 6-(p-hydroxyphenylazo)-uracil, an azopyrimidine known as selectively inhibiting theB. subtilis DNA polymerase III. IG 1 phages originated either by induction or infection multiply, abundantly, in the presence of that azopyrimidine, in spite of the drastic decline of cell viability. Chloramphenicol completely suppresses the induction effect and also blocks the formation of spontaneously induced phage particles.

    Notes: Abstract Although it is well known that informants often disagree about the degree of psychopathology in children, this issue has not been systematically evaluated in children with autism. The objective of this paper is to estimate the extent of agreement between parents and teachers on the assessment of autistic symptoms and adaptive behavior skills. We assessed 83 children, 4–6 years of age, with a diagnosis of pervasive developmental disorder (PDD), using the Autism Behavior Checklist (ABC) and the Vineland Adaptive Behavior Scales (VABS). Parents and teachers rated each child on each measure. While there was good agreement between informants on the VABS, teachers tended to rate the PDD children higher that parents. In contrast, there was virtually no agreement on the ABC. High levels of stress experienced by parents appeared to be associated with parents reporting more autistic behaviors and less adaptive skills than teachers. As with other child psychiatric disorders, caution must be exercised in combining information from several informants.

    Notes: Summary The temperate phages IG1, IG3, and IG4 ofBacillus subtilis were found to mediate specialized transduction of a λ-like type. High frequency transducing lysates were also obtained. In many instances the transductants were shown to be heterogenotes, segregating the mutant alleles. They are presumably the result of transduction by addition. These findings are discussed together with data on other members of the group III of temperate bacteriophages aiming to a deeper understanding of that group, and of its evolutionary history.

    Notes: Summary TheBacillus subtilis bacteriophages AR9, 3NT, and I10 belong to the same group as the widely used bacteriophage PBS1. These four phages have in common, among other properties, flagellotropic adsorption to host cells and total substitution of thymine by uracil in their DNAs. We tested the action of different restriction endonucleases on the four genomes. Among 14 enzymes, which did cut the DNAs into discrete fragments, six are known as having at least one thymine residue in their recognition sequences. The electrophoretical patterns of the digested DNAs from the four phages show strong similarities. Nevertheless, the differences are sufficient to allow a clear distinction between them.
