    Notes: Abstract Photosensitised haemolysis of red blood cells (RBC) by sodium hypericin, haematoporphyrin, protoporphyrin, Photofrin® and benzoporphyrin derivative monoacid ring A were investigated for broad spectrum near-ultraviolet (UVA)-visible light and monochromatic 630 nm irradiations. The delayed photohaemolysis rate (1/t 50) increased with the approximate square of the incident light dose, wheret 50 is the dark incubation time required for 50% haemolysis. The power dependence of 1/t 50 on the sensitiser concentration varied with the specific sensitiser and the irradiation conditions. Energy calculations for protoporphyrin show that the quantum efficiency of delayed photohaemolysis at 630 run is independent of the sensitiser concentration at constantt 50 and increases witht 50 The protective effects of water-soluble singlet oxygen quenchers are indicative of a Type II photosensitisation pathway for these agents. Pretreatment of RBC with hydrogen peroxide suppressed photosensitisation by protoporphyrin accompanied by lipid peroxidation and oxidation of haemoglobin. The photohaemolysis curves are analysed with a target theory model which postulates that haemolysis requires thermal activation of photochemical damage. The model calculations indicate that photosensitised haemolysis requires two photochemical hits and a highly cooperative thermal process.

    Notes: Abstract The recursive estimation of the regression function m(x) = E(Y/X = x) and its derivatives is studied under dependence conditions. The examined method of nonparametric estimation is a recursive version of the estimator based on locally weighted polynomial fitting, that in recent articles has proved to be an attractive technique and has advantages over other popular estimation techniques. For strongly mixing processes, expressions for the bias and variance of these estimators are given and asymptotic normality is established. Finally, a simulation study illustrates the proposed estimation method.

    Notes: Abstract Abstract. A variety of T, B and natural killer (NK) cell subsets defined by surface markers were analyzed by double immunofluorescence flow cytometry in the peripheral blood of patients following autologous bone marrow transplantation (ABMT, n=14), autologous peripheral blood stem cell transplantation (PBSCT, n=10) and allogeneic bone marrow transplantation (allo-BMT, n=6). Patients following ABMT were divided in 2 groups, those who did not received G-CSF post-transplant (ABMT, n=6) and those who did (ABMT+G, n=8). All patients following PBSCT or allo BMT received G-CSF. In all the groups prolonged significant decreases with respect to normal numbers were observed for the T CD3+, CD2+ and CD25+ subsets, more profound for the CD4+ subset but less for the CD8+ subset, especially following PBSCT (only decreased at 1 month). A significant expansion of the CD3+CD57+ and CD8+CD57+ phenotypes was noticed between 9 and 12 months following ABMT, the group of longer follow-up. Long-lasting expansion of the NK-like CD3+CD56+ and CD3+CD16+ subsets was also observed. The B CD19+ and CD20+ subsets had a significant overexpression from 4 months after ABMT, showing a normally balanced Igk+ : Ig1+ ratio. Concordantly, the HLA-DR+ and HLA-DQ+ subsets showed significant increases. The NK CD56+ and CD16+ subsets had a faster recovery than the T or B subsets in all the groups. However, the CD3-CD56+, CD3-CD16+, CD16+CD56+, CD3-CD8+, and especially the CD3-CD57+, CD16+CD57+, and CD56+CD57+ subsets had a slower recovery than the global CD56+, CD16+, or CD57+ subsets. The biological and clinical implications of these findings are discussed.

    Notes: Summary The effect of insulin on the synthesis of free fatty acids from glucose in the skeletal and heart muscles of chicken is examined. 10 min after glucose-(U-14C) administration, labeled free fatty acids (FFA) appeared in both skeletal and heart muscles. 0.75 IU of insulin kg−1 b. wt significantly increased the labeled FFA at the 30, 60 and 120 min intervals, with a maximum at 60 min.

    Notes: Abstract Since the beginning of this century evidence has accumulated which demonstrates that non-mammalian vertebrates possess photoreceptors situated deep within the brain. While many attempts have been made to localize these sensory cells, studies have either failed or been inconclusive. In this report we have used several experimental approaches to localize the deep brain photoreceptors of the lizard Anolis carolinensis. Using 3 antibodies that bind vertebrate cone opsins, we have immunolabelled cerebrospinal fluid (CSF)-contacting neurons located at the ventricular border within the nucleus ventromedialis of the septum. Western blot analysis indicates that these antibodies recognized a single 40 kD protein in ocular, anterior brain, and pineal extracts. Immunoblots of rodent brain did not show a similar protein band. We have also identified specific retinoids associated with phototransduction (11-cis and all-trans-3,4-didehydroretinaldehyde) within anterior brain extracts. This combined data provides the most detailed analysis of deep brain photoreceptors in any vertebrate. Consequently, we feel Anolis provides an excellent model to study this unexplored sensory system of the vertebrates.

    Notes: Abstract.  A portable fibre optic instrument for oxygen sensing based on luminescence lifetime is presented. The instrument is based on measurement of the quenching by oxygen of the room temperature phosphorescence (RTP) emitted by aluminium tris(8-hydroxy-7-iodo-5-quinolinesulfonic acid) chelate incorporated in an inorganic matrix by sol–gel technology. The comparatively long RTP lifetime of this sensing material (450 μs) and the large singlet–triplet splitting (λexc 390 nm, λem 590 nm) allow the use of simple opto-electronic circuits and low-cost processing electronics. Standard electronic components have been applied in development of the low-cost lifetime-measurement instrumentation presented here. Two optical sensor configurations, “flow-through cell” and “probe”, have been designed and evaluated for the determination of very low levels of oxygen, in gaseous argon streams and in waters. The basic technology, design parameters and performance characteristics of the optical sensors are discussed. Applications to determination of dissolved oxygen in river, tap and sewage waters are described. The advantages of luminescence lifetime measurements over conventional RTP intensity measurements for oxygen sensing are discussed.

    Notes: Abstract  Neurotrophins and their receptors (p75 and Trk family of receptors) play an important role in the survival of different populations of neurons in the central and peripheral nervous system. Expression of p75, TrkA, TrkB and TrkC was examined in mouse retinas by means immunohistochemistry in the postnatal development of normal and rd/rd mice (C57BL/6J). The rd/rd mice suffer a degeneration that causes a massive lost of photoreceptor cells. Results showed immunoreactivity to all three Trk proteins in both normal and rd/rd mice during the first 21 postnatal days, but some variations in intensity and localization were found. p75 immunoreaction was only present in rd/rd mice at the end of the degeneration process. These results could indicate a role of neurotrophins and their receptors in both the postnatal development of mouse retina and the degeneration process of rd/rd mice.
