    Notes: We report here on studies of the growth mechanism of CdTe during molecular beam epitaxy on (100) oriented CdTe substrates by reflection high energy electron diffraction (RHEED). The growth rate of CdTe as a function of the Cd/Te ratio in the impinging molecular beam was investigated in detail. The growth rates were determined by RHEED intensity oscillations. Fluxes were calibrated by film thickness measurements. From the growth rates dependence on the Cd/Te flux ratio we determined the Cd and Te sticking coefficients by comparing the results with kinetic models of molecular beam epitaxial growth. Both Cd and Te sticking coefficients are dependent on the surface concentration of free Cd and Te sites, respectively. The main result is that the influence of a precursor state has to be taken into account to describe the experimental results.

    Notes: High-pressure nuclear magnetic resonance (NMR) is an established method in NMR spectroscopy: on-line coupling of high-performance liquid chromatography with NMR, for example, reveals structural information which cannot be obtained with any other method. However, applications has been focused solely on high-pressure NMR spectroscopy, even though high-pressure NMR imaging allows in situ studies of processes such as the fluid exchange in porous media. A versatile high-pressure autoclave for NMR imaging is described in this article. The autoclave allows measurements in any horizontal NMR imager using magnetic field coil systems with an inside diameter of more than 70 mm. Any sample with a diameter up to 28 mm and a length of about 200 mm can be investigated. The autoclave is constructed for operating pressures up to 10 MPa and is temperature controlled between 10 and 60 °C. The materials of the high-pressure cell which are the thermoplastic polyetheretherketon (PEEK) for the pressure tube and brass (63% Cu, 37% Zn) for the caps also permit investigations with aggressive fluids such as supercritical carbon dioxide. Inlet and outlet valves allow replacement of fluids and pressure variations in the autoclave during the NMR measurement. FLASH NMR images of the fluid exchange of methanol for liquid carbon dioxide in silica alcogels at 6.5 MPa are presented in order to demonstrate possible applications. © 1999 American Institute of Physics.

    Notes: Purpose This in vitro study was carried out to examine the fracture strength of metal- free posterior inlay fixed partial dentures (IFPDs). The 3-unit IFPDs were made of either a polyethylene fiber–reinforced composite, 3 glass fiber–reinforced composites, or an all-ceramic material.Materials and Methods Eight IFPDs were fabricated of each material in accordance with the manufacturer's instructions and luted to extracted human molars with a dual-cure adhesive system. The molars were positioned in PMMA resin 10 mm apart mesiodistally. Inlay cavity preparations with enamel finishing lines were used. After thermal cycling and mechanical loading in an artificial oral environment, the cemented IFPDs were mechanically loaded until failure. Visual and radiologic examinations were done out to discern the different forms of fracture. Median and 25%/75% percentile values were calculated. Statistical analysis was performed using the Mann-Whitney U test and the Kruskal-Wallis test (p≤0.05).Results With a median (and 25%/75% percentile) fracture strength of 368 N (234 N/424 N), the FibreKor/Conquest Sculpture showed significantly lower values than the Connect/BelleGlass [898 N (736 N/1033 N)], Vectris/Targis [723 N (692 N/806 N)], Everstick/Sinfony [634 N (532 N/673 N)], and Empress2 [520 N (385 N/706 N)].Conclusions Assuming maximum chewing forces of ≥500N in posterior areas, all systems showed sufficient fracture strength in most cases and warrant further investigation for potential clinical use.

    Notes: Abstract: We have reported previously that oxysterols inhibit astrogliosis and intracranial glioblastoma growth. To elucidate the mechanism of action of these molecules in vivo, we have investigated their effect on the cholesterol biosynthesis in the injured brain. In a bilateral lesion model, injection of liposomes containing 7β-hydroxycholesterol decreased [3H]acetate incorporation into neutral lipids and cholesterol by 30% and 40%, respectively. Structural analogues were tested using a unilateral lesion model. The injury did not significantly affect cholesterogenesis; injection of 7β-hydroxycholesterol or 7β-hydroxycholesteryl-3-oleate reduced acetate incorporation into cholesterol by 47% and 43%, respectively. Both 7-ketocholesteryl-3-oleate and 7α-hydroxycholesteryl-3-oleate inhibited cholesterogenesis by 32%. As cholesterol and by-products of the cholesterol pathway play a key role in cell division, we have assessed the effect of oxysterols on reactive astrocyte proliferation. The incorporation of bromodeoxyuridine showed that up to 46% of astrocytes were proliferating 24 h after the injury. Injection of 12 nmol of 7β-hydroxycholesterol or 7β-hydroxycholesteryl-3-oleate reduced the labelling index to 26%, whereas the labelling index in the 7-ketocholesteryl-3-oleate-treated cortex was 37%. These findings demonstrate that oxysterols are potent inhibitors of the endogenous cholesterol biosynthesis in brain and show a correlation between cholesterogenesis and reactive astrocyte proliferation.

    Notes: Abstract: A simple and efficient transfection technique based on lipopolyamine-coated DNA that can be used for gene transfer in cerebellar granular neurons is described. Gene transfer is achieved by exposure of cells to a DNA/lipid complex obtained by simple mixing of lipopolyamine and plasmid DNA. This procedure may represent a general tool of physiological investigations in primary cells. We show that the promoters of the introduced chimera genes are regulated by their respective trans-acting factors and may be modulated via membrane receptors and second messengers. This procedure has no noticeable toxic effects, nor does it seem to interfere with complex physiological behavior like neuronal differentiation.

    Notes: In endemic areas, clinical manifestations of Plasmodium falciparum infection range from asymptomatic parasitaemia to life-threatening severe syndromes. Immune differences that could account for this disparity are poorly understood. Using tight criteria to classify patients into non-overlapping clinical categories, we showed that cerebral malaria and severe anaemia were distinct immunological syndromes and that a proper quantitative description of cytokine profiles in the various clinical groups is essential to the understanding of the activation of immunocompetent cells.Due to the limited size of paediatric blood samples, we chose to measure cytokine mRNA using real-time RT-PCR. We showed that RT efficiency displayed intra-and intergenic variations that have to be taken into consideration for reliable absolute RNA quantification when comparing clinical cases. We thus developed a SYBR Green I-based real-time RT-PCR method using synthetic external RNA standards specific for each gene. Absolute RNA quantification is achieved by reverse transcribing known copy numbers of this RNA standard in parallel with cellular RNA. Strictly specific primers were designed to allow the quantification of any RNA in the same thermocycling parameters for future automation. Our method gave similar results for a lower cost when compared with TaqMan, and led to reproducible and reliable absolute RNA quantification. We validated it in vitro on naïve PBMC stimulated by LPS and ex vivo on PBMC from malaria patients. This new method raises the unprecedented possibility to compare cytokine mRNA levels between different clinical groups and is a powerful tool to further study the inflammation processes associated to malaria.
