Notes: Abstract The separation of enantiomers by gas chromatography on a chiral stationary phase is stored and conveniently retrieved in the molecular database Chirbase/GC. According to the present information content (version 3/96), a great variety of analytes has been published in several hundred journals. A selected review is given on 364 enantiomer separations of 102 different molecules containing at least one four-membered saturated carbocyclic ring. Factors determining the degree of enantioseparation are outlined and analyzed.

Notes: Following an extended chiral drug screening program by capillary zone electrophoresis (CZE), the enantioseparation of 86 racemic drugs was tested with γ-cyclodextrin as a chiral solvating agent. Unified conditions were applied to all experiments. In total, 18 drug racemates were separated, 13 entries thereof that had not been separated at the lower CSA concentration applied in an earlier stage of the project. A comparison of the data with the results obtained for α- and β-cyclodextrin points to the significance of partial penetration (“side-on binding”) of aryl groups into the cyclodextrin cavity. Notes: ---Following an extended screening project, 86 racemic drugs were investigated by capillary zone electrophoresis in the presence of the chiral solvating agent (CSA) hydroxypropyl-β-cyclodextrin. A total of 42 drugs out of 86 tested was thereby separated into enantiomeric pairs. Statistical analysis of the numerous experiments performed un der identical conditions reveals a loose correlation of the migration separation factor (αm) with the migration retardation factor (Rm). For a subset of 23 drugs, a drop in the concentration of the CSA was also studied, showing the way to further optimization.

Notes: ---Orthogonal design and uniform design were used for the optimization of separation of enantiomers using 2,6-di-O-methyl-β-cyclodextrin (DM-β-CD) as a chiral selector by capillary zone electrophoresis. The concentration of DM-β-CD, buffer pH, running voltage, and capillary temperature were selected as variable parameters, their different effects on peak resolution were studied by the design methods. It was concluded that orthogonal design offers a rapid and efficient means for testing the importance of individual parameters and for determining the optimum operating conditions. However, for a large number of both factors and levels, uniform design is more efficient. The effect of addition of methanol and citric acid buffer on the separation of enantiomers was also examined.

Notes: In an extended chiral drug screening program, enantioseparation of 86 racemic drugs was tested with hydroxypropyl-γ-cyclodextrin as chiral solvating agent (CSA). A total of 30 drugs out of 86 could be resolved in this straightforward approach. The number of experiments performed under identical conditions allows a statistical treatment of the data. The enantioseparation of the analytes is correlated with their interaction strength with the CSA. Hence, the concentration of the CSA is a crucial parameter for optimization of the enantioseparation, as shown by a subset of 23 examples.
