    Notes: Oxidative stress is implicated in the death of dopaminergic neurons in sporadic forms of Parkinson's disease. Because oxidative stress can be modulated endogenously by uncoupling proteins (UCPs), we hypothesized that specific neuronal expression of UCP2, one member of the UCP family that is rapidly induced in the CNS following insults, could confer neuroprotection in a mouse model of Parkinson's disease. We generated transgenic mice overexpressing UCP2 in catecholaminergic neurons under the control of the tyrosine hydroxylase promoter (TH-UCP2). In these mice, dopaminergic neurons of the substantia nigra showed a twofold elevation in UCP2 expression, elevated uncoupling of their mitochondria, and a marked reduction in indicators of oxidative stress, an effect also observed in the striatum. Upon acute exposure to 1,2,3,6-methyl-phenyl-tetrahydropyridine, TH-UCP2 mice showed neuroprotection and retention of locomotor functions. Our data suggest that UCP2 may represent a drug target for slowing the progression of Parkinson's disease.

    Notes: Activation of ion channel-linked glutamate receptors, especially N-methyl-d-aspartate (NMDA) receptors, mediates the excitotoxic effects of glutamate upon central neurons. We examined the hypothesis that activation of group I metabotropic glutamate receptors (mGluRs) would increase NMDA receptor-mediated cortical neuronal death. Addition of the selective group I mGluR agonists, dihydroxyphenylglycine (DHPG) or trans-azetidine-2,4-dicarboxylic acid (t-ADA) potentiated NMDA-induced neuronal death, and application of the group I mGluR-selective antagonist, aminoindan-1,5-dicarboxylic acid (AIDA), as well as the non-selective antagonists methyl-4-carboxyphenylglycine (MCPG) or 4-carboxyphenylglycine (4CPG) reduced NMDA- and kainate-induced neuronal death in murine cortical cultures. The pro-excitotoxic effect of group I mGluR activation may be mediated largely by enhancement of glutamate release, as DHPG potentiated high potassium-stimulated glutamate release, and the protective effects of both AIDA and MCPG were abolished when NMDA and alpha-amino-3-hydroxy-5-methyl-4-isoxazole proprionic acid (AMPA) receptors were blocked immediately after toxic NMDA receptor overstimulation. The present data support the possibility that antagonizing group I mGluRs may be a useful strategy for attenuating excitotoxic neuronal death in certain disease states.

    Notes: Nuclear factor I (NFI) proteins are DNA-binding transcription factors that participate in the tissue specific expression of various genes. They are encoded by four different genes (NFI-A, B, C, and X) each of which generates multiple isoforms by alternative RNA splicing. NFI-like binding sites have been identified in several genes preferentially expressed in olfactory receptor neurons. Our prior demonstration that NFI binds to these elements led to the hypothesis that NFI is involved in the regulation of these genes. To analyse the role of NFI in the regulation of olfactory neuron gene expression we have performed transient transfection experiments in HEK 293 cells using constructs that place luciferase expression under the control of an olfactory marker protein (OMP)-promoter fragment containing the NFI binding site. In vitro mutagenesis of this site revealed a negative modulation of luciferase expression by endogenous NFI proteins in HEK 293 cells. In addition, we have used in situ hybridization to analyse the tissue and cellular distribution of the four NFI gene transcripts during pre- and postnatal mouse development. We have simultaneously characterized the expression of Pax-6, and O/E-1, transcription factors known to regulate the phenotype of olfactory receptor neurons. We demonstrate that all of these transcription factors vary in specific spatio–temporal patterns during the development of the olfactory system. These data on NFI activity, and on transcription factor expression, provide a basis to understand the role of NFI in regulating gene expression in olfactory receptor neurons.

    Description / Table of Contents: Abstract The Geological Institute of Bonn University since 1975 investigates in connection with DSDP/IPOD the Cretaceous of the Atlantic coastal basin in south-western Morocco. 30 main stratigraphic sections and numerous auxiliary ones were taken along several traverses, running along distances of 150 to 200 km in west-east direction. The recognized epicontinental facies-pattern along the traverse of the southern High Atlas mountain range depicts clearly, that we cut here the critical region of intertonguing marine, fine-clastic-carbonaceous sediments and continental detritic deposits. The littoral zone successively progrades with three major transgressions (Neocomian, Upper Aptian, Turonian) towards the African Continent. Sedimentation processes, stratigraphy, paleomagnetics, geochemistry, sedimentology and paleogeography are investigated, taking selected horizons on one hand and complete stratigraphic columns on the other. A new sedimentation model reflecting transgression and regression events in the Cretaceous “Atlas Gulf” show some probabilities of eustatic sea-level changes. There are similarities between the Atlas Gulf and other coastal basins on both sides of the Atlantic Ocean. There are certain relations between the sedimentary record of the proand regrading littoral zone on the continent and in the nearby deep sea deposits. There is an exciting coincidence between the stratigraphie succession of the downwarping Atlas Gulf and the sedimentary cover of the interior of Africa, especially the Mesozoic Sahara Basin.
    Abstract: Résumé L'institut géologique de l'université de Bonn fait des recherches géologiques dans le Crétacé du Bassin Côtier Atlantique dans le sudouest du Maroc. Ces études font partie du «Deep-Sea-Drilling-Project» (DSDP), programme de recherche international. 30 coupes principales et un grand nombre de coupes partielles sont levées suivant trois tracés pénétrant de 150–200 km vers l'est, à partir de la côte. Un schéma des facies dressé au sud du Hautatlas, met en évidence qu'il s'agit d'une région montrant le passage complet de sédiments marins épais argilo-carbonatés à des dépôts terrestres. La zone littorale pénètre par étapes de plus en plus dans la région continentale au cours de trois grandes transgressions (Néocomien, Aptien supérieur, Turonien). Ces transgressions sont interrompues par des périodes de régression. Dans le but d'illustrer les modalités de la sédimentation accompagnant ces événements, les études portent de façon détaillée sur la stratigraphie, le paléomagnétisme, la géochimie, la sédimentologie et la paléogéographie d'horizons choisis et de parties entières de la colonne stratigraphique. Un modèle de sédimentation est conçu pour la région du Golfe d'Atlas au Crétacé. Les rapports paléogéographiques entre ce golfe et les bassins de sédimentation côtiers d'Amérique et d'Afrique d'une part, les dépôts abyssaux de l'Atlantique d'autre part et finalement le Mésozoique du Sahara sont ensuite discutés.
    Notes: Zusammenfassung Im Rahmen des deutschen Beitrages zum internationalen Deep Sea Drilling Project (DSDP) untersucht das Geologische Institut der Universität Bonn seit 1975 die Kreide des atlantischen Küstenbeckens in Südwestmarokko. 30 Hauptprofile sowie viele Zwischen- und Teilprofile wurden entlang 150–200 km landeinwärts verlaufenden Trassen aufgenommen. Aus dem Faziesschema am Südrand des Hohen Atlas geht hervor, daß hier der Verzahnungsbereich mächtiger mariner, pelitisch-karbonatischer Sedimente mit festländischen Ablagerungen vollständig erfaßt wird. Die Litoralzone rückt vom Liegenden zum Hangenden in drei ausgreifenden Transgressionen (Neocom, höheres Apt, Turon) — unterbrochen von Regressionen — schrittweise gegen den Kontinent vor. Mit Zielrichtung auf die damit verbundenen Sedimentationsabläufe werden Stratigraphie, Paläomagnetismus, Geochemie, Sedimentologie und Paläogeographie einzelner ausgewählter Horizonte und ganzer Schichtsäulen eingehend untersucht. Für den einheitlichen Sedimentationsraum eines kreidezeitlichen Atlas-Golfs wird ein Sedimentationsmodell entwickelt. Paläogeographische Beziehungen zu den Küstenbecken Amerikas und Afrikas, zu Tiefseeablagerungen des Atlantischen Ozeans und zum Sahara-Mesozoikum werden diskutiert.

It was found that the basal activity of AChE was increased in myotonic nerves while its proximodistal transport was inhibited.
