    Notizen: Summary. A high frequency shoot regeneration (80%) was developed from callus of leaf disc and stem internodes of Moricandia arvensis. Leaf discs were shown to be a preferable starting material for transformation experiments. Agrobacterium tumefaciens strain GV3101/pMP90 used in this study contained a binary vector with genes for kanamycin resistance, hygromycin resistance and β‐glucuronidase (GUS). Maximum transformation efficiency (10.3%) was achieved by using kanamycin at the rate of 200 mg/l as a selection agent. Presence of tobaco suspension culture during co‐cultivation and a pre‐selection period of seven days after co‐cultivation was essential for successful transformation. Transgenic plants grew to maturity and exhibited flowering in a glasshouse. GUS activity was evident in all parts of leaf and the presence of GUS gene in plant gemone was confirmed by PCR analysis.

    Notizen: Abstract The vegetative-to-floral transition ofBrassica campestris cv. Osome was induced by vernalization. Poly(A)+RNA was isolated from the transition shoot apex after 6 weeks of vernalization, the floral apex after 12 weeks of vernalization and the expanded leaves just before vernalization, and cDNAs were synthesized. These cDNAs were used for subtraction and differential screening to select cDNA preferentially present in the transition and floral apices. Nucleotide sequences of the resulting 14 cDNA clones were determined, and northern blot analysis was carried out on six cDNAs. Two cDNA clones which did not show significant similarity to known genes were shown to be preferentially expressed in the floral apex.

    Notizen: Abstract  The sporophytic self-incompatibility (SI) system in Brassica campestris (syn. rapa) is controlled by multiple alleles of a single locus, S, in which SLG and SRK are located. SLG and/or SRK appear to comprise the female component of the recognition reaction of SI. The gene encoding the male component must also be located at the S locus but has not been identified to date. RNA gel blot analysis with regions flanking SLG 9 and SRK 9 revealed two anther-expressed genes 3’ of the SLG 9 gene. Two cDNA clones were isolated using the 3’-flanking region of SLG 9 as probe. Because of sequence similarity with the Brassica SLL2 gene, one cDNA clone was designated SLL2-S 9 (for S Locus-Linked 2-S 9 gene). The 5’-region of SLL2-S 9 was not present in other SLL2 genes examined. The second gene was novel and was designated SAE1-S 9 (for S locus Anther Expressed 1-S 9 gene). SAE1-S 9 and SLL2-S 9 were located approximately 2.5 kb and 5.5 kb, respectively, downstream of SLG 9 . A 1.4-kb transcript for SLL2-S 9 was detected in both vegetative and reproductive tissues, whereas 1.9-kb and 2.4-kb transcripts were specifically detected in the anther of S 9 haplotype. SAE1-S 9 was specifically expressed in the anther at the uninucleate stage. This gene encoded a novel polypeptide of 257 amino acids. Database analysis revealed a homologous sequence in the upstream region of SRK 3 of B. oleracea. We discuss possible relationships between SLL2-S 9 and SAE1-S 9 and SI.

Three-week-old scutella calli served as an excellent starting material. These were infected with an Agrobacterium tumefaciens strain EHA101 carrying a plasmid pIG121Hm containing genes for β-glucuronidase (GUS) and hygromycin resistance (HygR). Hygromycin (50 mg/l) was used as a selectable agent. Inclusion of acetosyringone (50μM) in the Agrobacterium suspension and co-culture media proved to be indispensable for successful transformation. Transformation efficiency of Basmati 370 was 22% which was as high as reported in japonica rice and dicots. A large number of morphologically normal, fertile transgenic plants were obtained. Integration of foreign genes into the genome of transgenic plants was confirmed by Southern blot analysis. GUS and HygR genes were inherited and expressed in R1 progeny. Mendelian segregation was observed in some R1 progeny.

    Notizen: Abstract We have isolated a rice cDNA clone that is homologous to the gene for the maize NADP-dependent malic enzyme (EC 1.1.1.40; NADP-ME). The deduced amino acid sequence coded for by the cDNA indicates a high level of homology to chloroplast type NADP-ME, including a transit peptide with pronounced hydrophobic properties at the amino terminus. Northern blot analysis indicates that the expression of this gene is regulated by external stress such as submergence.

    Notizen: Abstract A cDNA library of rice (Oryza sativa L.) has been constructed from anthers at an early stage of pollen development. By differential screening of the library, we have isolated cDNAs of two genes, designated as Osc4 and Osc6, that are abundantly expressed in anthers containing tetrads and uninucleate microspores, but are not expressed in leaves or roots. Expression of Osc4 is absent in mature anthers, while Osc6 is present although the expression decays during pollen maturation. A comparison of the nucleotide and deduced amino acid sequences with those in data banks has not shown significant homology to known molecules.

    Notizen: Abstract In situ localization of mRNA was carried out on two cDNAs (Osc4 and Osc6) that had been isolated from rice anthers at the microspore stage. The mRNA corresponding to each cDNA was shown to be localized only in the tapetal cells of the rice immature anthers, but not in the microspores or the mature pollen. The corresponding genomic clone, Osg6B, was isolated, and its 5′-upstream region was found to regulate β-glucuronidase expression in the tapetum of transgenic tobacco. A set of 5′ deletions was also generated and a 1095 bp 5′ region was revealed to be necessary for activation of the Osg6B promoter in transgenic tobacco.
