    Notizen: Abstract The effect of long-term misoprostol — a synthetic prostaglandin E1 analogue — ingestion on rat gastric morphology and cell turnover was studied. Misoprostol in a daily dose of 90 μg/kg or 9,000 μg/kg was intragastrically administered to rats. Control rats were treated with the vehicle only. Following 90 days of treatment, 3H thymidine was injected i. v. and rats were sacrificed 1 h later. Tissue sections were prepared from the stomach body and subjected to autoradiography. Misoprostol treatment significantly reduced body weight gain. High dose misoprostol treatment induced significant increases in gastric wall thickness and in gastric gland length. On the other hand, the labelling index was significantly reduced by treatment with high dose misoprostol. These results indicate that chronic administration of misoprostol in high doses increases gastric wall thickness and decreases gastric cell turnover, suggesting that administration of prostanoids causes an increase in cell survival and a decrease in cell shedding.

    Notizen: Abstract Background.Physiologic monitors present an influx of numerical data that can be overwhelming to the clinician. We combined several parameters in an effort to reduce the amount of information that must be continuously monitored including oxyhemoglobin saturation by pulse oximetry, end-tidal CO2 concentration, arterial blood pressure, and heart rate into an integrated measure – the health stability magnitude (HSM). The HSM is computed for a pre-determined basal period, the reference HSM (RHSM), and recalculated continuously for comparison with the baseline value. In this study we present the HSM concept and examine changes in the HSM during abdominal aortic aneurysm surgery. Materials and methods.After IRB approval, nine patients were studied. The anesthesiologist recorded all significant intra-operative events. Within a defined time interval, data were recorded and used to calculate a combined parameter, the HSM. The baseline or reference value of this index (RHSM) was calculated after the induction of anesthesia. Individual HSM values were repeatedly calculated for ten second periods after the RHSM value was established. A 〉 30% deviation of the HSM from the RHSM was considered significant. Deviations in the HSM were compared with events recorded by the anesthesiologist on a paper record and with there cord from an electronic record-keeping system. The deviation observed between two consecutive HSMs, called dHSM, was plotted against HSM to construct a contour diagram of data from all patients to which individual cases could be compared. Results.The plot showed that dHSM vs. HSM values were tightly clustered. The inner contour on the distribution plot contained 90% of values. Individual patient’s time course, projected on this diagram, revealed deviations form “normal” physiology. Fifty-nine events led to 〉 30% deviations in the HSM; 27 were anticipated events and 32 were unanticipated. Conclusion.The correlation between HSM and dHSM depicts changes in multiple monitored parameters that can beviewed using a single graphical representation. Projection of individual cases on the contour diagram may help the clinician to distinguish relative intraoperative stability from important events. Data reduction in this manner may guide clinical decision-making in response to unanticipated or unrecognized events.

    Notizen: Abstract Animal dietary studies related to human colorectal carcinogenesis are usually based on AIN-76A diet, which is dissimilar to human food in source, preparation, and content. Our aim was to examine colonic epithelial proliferation in rats fed a diet based on the mean daily food intake of adenoma patients. Foods were prepared as reported by the adenoma patients and dehydrated; 64 Sprague-Dawley rats were fed either “human adenoma” or AIN-76A diet and every eight weeks, eight from each group were sacrificed. Both groups gained weight equally, had no colonic histological changes, but during the study showed progressive lengthening of colonic crypts (P〈0.01) and decreased proliferation (P〈0.05) in distal colons. Compared to controls, rats fed human adenoma diet had significantly longer crypts (P〈0.01) and more labeled cells (P〈0.05) at 32 weeks; overall they had increased proliferation (P〈0.01), most significantly in the distal colon. Thus, food eaten by adenoma patients induced hyperproliferative changes in the rat colon during growth and maturity, especially the distal colon, as found in humans at risk for neoplasia.

    Notizen: Abstract Hepatocytes are hypothesized to continuallystream from the portal tract to the terminal hepaticvein. By this model, when a cell divides, one of itsprogeny replaces the dividing ancestor and the other is displaced into a more remote location. Thepresent experiment aims to demonstrate thathypothyroidism affects liver cell turnover. Thirty maleadult rats were divided into two groups. One receivedmethimazole for two weeks and the other served as control.Each rat was injected intraperitoneally with 18.5 KBq[3H]thymidine/g body weight. Rats were killedafter 1 hr and two and four weeks. Autoradiography was done. The distance of the labeled cells fromthe portal tract was measured. The mean TSH levels ofthe methimazole-treated group and controls were 1.45 and0.25 mM/liter, respectively (P 〈 0.01). Hepatocyte streaming was lower in hypothyroid (1.8μm/day) than in untreated rats (2.5 μm/day) (P〈 0.01). The respective labeling indices 1 hr afterlabeling were 0.9% and 1.24% (P 〈 0.05). We concludethat hypothyroidism diminishes hepatocyte and littoral cellturnover and slows down their streaming.

    Notizen: Summary The inner enamel epithelium (IEE) covers the labial tooth aspect as a one cell layer which, when cut sagittally, appears as a longitudinal cell column extending from the tooth origin toward the periphery. Following sudden tooth shortening, the IEE responds by an increased cell production which later declines below normal values. The perturbation affects all cell kinetic parameters; the progenitor compartment, which initially increases, diminishes in size toward end of the experiment. The cell cycle transition times, which initially decline, rise toward the end of the experiment. The mean normal daily cell production rate of 70 cell % (i.e. 70 cells are produced by 100 progenitors) increases to 111 cell % and then declines to a low of 51 cell %. The IEE response typifies the behavior of other cell renewal systems such as intestinal epithelium and epidermis.
