    Notes: Purpose - Seeks to evaluate the link between task and relationship conflict, and their influence on some employees' affective reactions such as satisfaction, wellbeing, and propensity to leave a job; and to analyse the mediated and moderated role of relationship conflict. Design/methodology/approach - The study involved 169 employees from six service organizations (hotels) in Andalusia (Spain). A questionnaire was used containing different measures: task and relationship conflict, wellbeing, job satisfaction, and propensity to leave the job. Findings - The two types of conflict have different consequences. Data show that relationship conflict is negatively associated with affective reactions, while task conflict does not relate directly to affective reactions in a predictable way; relationship conflict has a positive influence on the desire to leave the current job, while task conflict does not affect it negatively; the interactive effect of relationships and task conflict shows that this interaction contributes substantially to predict the propensity to leave the current job; and relationship conflict mediates in the link between task conflict and affective reactions. Research limitations/implications - A high level of task conflict may backfire by boosting relationship conflict as well, thus having a negative effect on affective reactions. Thus some conclusions can be drawn with a view to improving conflict management in teams. First an attempt must be made to understand the type of conflict that is taking place. Second, managers should encourage open discussion of task-related issues. Third, special attention should be paid to the level of each conflict because of its interactive effects on some affective outcomes. Thus, in spite of the generally beneficial effects associated with task conflict, the intensification of task-related conflict may backfire when interacting with dysfunctional affective-dissent. Originality/value - Serves too analyze the mediated and moderated role of relationship conflict and to test the role of types of conflict on affective reactions such as wellbeing and propensity to leave the job.

    Notes: Purpose - In an effort to directly manufacture devices with embedded complex and three-dimensional (3D) micro-channels on the order of microns to millimeters, issues associated with micro-fabrication using current commercially available line-scan stereolithography (SL) technology were investigated. Design/methodology/approach - Practical issues associated with the successful fabrication of embedded micro-channels were divided into software part preparation, part manufacture, and post-cleaning with emphasis on channel geometry, size, and orientation for successful micro-fabrication. Accurate representation of intended geometries was investigated during conversion from CAD to STL and STL to machine build file, and fabricated vertical and horizontal micro-channels were inspected. Additional build issues investigated included accurate spatial registration of the build platform, building without base support, and Z-stage position accuracy during the build. Findings - For successful fabrication of micro-channels using current technology, it is imperative to inspect the conversion process from CAD to STL and STL to machine build file. Inaccuracies in micro-channel representation can arise at different stages of part preparation, although newer software versions appear to improve representation of micro-geometries. Square channel cross-sections are most easily sliced and vertical channels are most easily stacked together for layered manufacturing. While building, a means should be developed for building without base and internal supports, providing feedback on Z-stage position, and having the capability for cleaning the micro-channels. Research limitations/implications - This research demonstrates that commercial SL technology is capable of accurately fabricating embedded vertical square cross-section micro-channels on the order of 100?µm (with reasonable advancements to smaller scales on the order of 10?µm achievable). Additional practical limitations exist on other channel geometries and orientations. The research used a single resin and additional material resins should be explored for improved micro-fabrication characteristics. Practical implications - Practical issues associated with micro-fabrication of embedded channels with appropriate solutions using available SL technology were provided. It is expected that these solutions will enable unique applications of micro-channel fabrication for micro-fluidic and other devices. Originality/value - This work represents an original investigation of the capabilities of current line-scan SL technology for fabricating embedded micro-channels, and the solutions provide the means for applying this technology in micro-fabrication.

    Notes: Abstract. Five major soluble nuclear proteins associated with cell proliferation were identified in Allium cepa L. root cells. One of them, of 64 kDa, was revealed by Western blotting with anti-mammalian nucleolin antibodies. A polyclonal antibody raised against this protein, which we have named NopA64, localised it in the nucleolus as well as in nuclear coiled bodies. Together with NopA64, the antibody also revealed a smaller form, called NopA61. Both proteins were present in the soluble ribonucleoprotein fraction and in the nuclear matrix of proliferating cells, but NopA61 was the only form revealed in differentiated cells. NopA64 contained epitopes also present in other plants, in mammalian nucleolin and in its yeast homologue, gar2. In mammals, the highest homology was with 50-kDa nucleolin fragments containing the RNA-binding motifs and the glycine-arginine-rich (GAR) domain. NopA64 was moderately phosphorylated in vitro by exogenous casein kinase II and cdc2 kinase, whereas NopA61 was highly phosphorylated by casein kinase II. Furthermore, NopA61 was the only band detected after dephosphorylation as well as after endoproteolysis of NopA64. This protein could be one of the various functional homologues of mammalian nucleolin in plant cells.

    Notes: Abstract. The nucleolar protein fibrillarin has been studied in onion cells; it is detected as an Mr 37,000 protein by immunoblotting using a human autoimmune serum. Quantitative immunoelectron microscopy showed that most fibrillarin is localized in the transition zone between the fibrillar center (FC) and the dense fibrillar component (DFC) as well as in the proximal zone of the DFC, where the labeling shows a gradual decrease outward until it reaches insignificant levels in the distal zone of the DFC. Thus, fibrillarin is not uniformly distributed throughout the DFC of plant cells. This result supports the hypothesis that the morphologically homogeneous DFC may not be uniform in function; it is also in agreement with the hypothesized vectorial flow of ribosome biogenesis through the same compartments. Data are also presented showing that the amount of fibrillarin increases when nucleolar activity increases in G2, and probably decreases when nucleolar activity decreases during differentiation.

    Notes: Abstract Intranucleolar DNA, including ribosomal DNA (rDNA), was localized in situ in proliferating onion cells under the electron microscope using an anti-DNA monoclonal antibody and a postembedding indirect immunogold procedure. In the interphase nucleolus of this species, characterized by a very high amount of rRNA genes, we found DNA concentrated mostly in fibrillar centres (FCs) and in the region of the dense fibrillar component (DFC) immediately surrounding them. Clusters of gold particles were frequently seen covering both of these structural components of the nucleolus at the same time. Moreover, the same technique, applied to transcriptionally arrested quiescent onion cells, showed the nucleolar DFC devoid of DNA. Also, in mitotic cells at telophase, the prenucleolar material, which has the same morphological and cytochemical features as the DFC, does not contain DNA. These data suggest the existence of at least two subcomponents of the DFC in the onion cell nucleolus, one associated with pre-rRNA synthesis, and the other, with further processing of transcripts, already released from the rDNA template. We conclude that the first subcomponent forms part of the “transition between FC and DFC”, which is the in situ structural counterpart of pre-rRNA synthesis. This transition is morphologicaly sizeable in onion cells, because of their high number of rRNA genes and the large size of the DFC mass; however, it would be barely detectable in situ in other cell systems, where the whole DFC comprises only a thin layer and the amount of rDNA is considerably reduced.

    Notes: Abstract The pattern of nucleolar silver staining was determined in the interphase and mitosis of plant cells using two cytological techniques, the selectivity of which was tested densitometrically. For complete accuracy, silver grains were removed and the sections were then observed under uranyl and lead staining. During interphase, the highest density of silver grains occurred in the portions of the dense fibrillar component nearest to the fibrillar centres; the latter were also stained except for the condensed chromatin inclusions typical of the heterogeneous type. During mitosis, not only the nucleolar organiser region (NOR) was stained, but also the prenucleolar material in anaphase and telophase. Two different proteins or sets of proteins are suggested to be responsible for this staining pattern; one of them is associated with NOR chromatin (which is not contained in the mitotic prenucleolar material) and the other with early ribosomal precursors (which have never been found in fibrillar centres). From these two proteins, the “Ag-NOR” reveals either transcriptionally active or potentially active chromatin, characterized by an uncoiled, extended structure, regardless of the state of transcription. This is the structural feature detected by Ag-NOR staining.
