    Notes: Summary The distribution of the preexisting and of thede novo synthesized proteins among soluble, and insoluble fractions, as well as between immunoprecipitable and non-immunoprecipitable soluble proteins has been studied in sea urchin embryos at different stages of development. In the S-100 fraction, which represented about 20% of total proteins, only a minor part of radioactivity was found. The majority of newly synthesized proteins was insoluble at neutral pH. Such distribution was practically invariant for all investigated stages of development, and was not markedly affected by Dactinomycin nor by 5-azacytidine. Only a small percentage of the total radioactivity of the S-100 fraction was found in the antigen-antibody complexes of soluble proteins. No “shift” of newly synthesized proteins towards the type of old, preexisting antigenic proteins was detected, and the majority of soluble newly synthesized proteins was found to be related to the non-immunoprecipitable soluble proteins.

    Notes: Summary The changes of histones related to the development of sea urchin embryos from blastula to pluteus stage were studied by an electrophoretic method. The observed alterations were found to be quantitative ones and related to the transition of embryos from blastula to gastrula stage. During this period an increase in the relative amount of the F-1 and F-3 histones on account of the F2b + F2a2 was observed. The patterns of gastrula and pluteus histones were found to be similar.

    Notes: Summary The changes of the patterns of soluble proteins during early embryogenesis have been studied by acrylamide and agar-agar gel electrophoresis. Immunological analyses of electrophoretically separated antigens have been done by diffusion-in-gel methods. It has been shown that immunoprecipitable proteins of the S-100 fraction represent a part of preexisting, bulk proteins. During embryogenesis quantitative changes take place in the distribution of these proteins. These changes are more pronounced after gastrulation, and are differently timed for particular antigens. Qualitative changes were not evident by the methods used in these experiments.

    Notes: Abstract We have studied the effect of soman intoxication on serum acute phase reactants (APR) levels, and the relationship of the APR and corticosterone concentrations and the immunosuppressive activity of the serum. One day after the injection of 1.8 LD50 soman the concentrations of α2-macroglobulin (α2-MG) and α1-acid glycoprotein (AGP) in the serum of antidote protected rats increased 4- and 7-fold, respectively, whereas those of hemopexin (Hx), haptoglobin (Hp) and cysteine protease inhibitor (CPI) were two to three times higher than in the controls. A similar magnitude of increase of serum acute phase reactants levels was observed when 0.3 LD50 soman was administered at 24-h intervals over the 5-day period. The relationship of changes in the APR concentration, corticosterone level and immunosuppressive activity of the serum was also comparable to that observed in the acute phase response to tissue injury.

    Notes: Abstract The effect of soman on rat brain ribosomes organization and translational activity of mRNA in cellfree system was studied in rats exposed to 1.3 LD50 soman (120 μg/kg body weight) and in rats repeatedly injected with 0.4 LD50 soman (35 μg/kg). Fifteen minutes after the injection of 1.3 LD50 soman the heavy polyribosomal fraction from rat brain was found to be enriched and translational activity of mRNA was enhanced. In rats administered five injections of 0.4 LD50 soman at 24-h intervals, the low density ribosomes appeared as the predominant fraction whereas the activity of mRNA in all cell-free system was significantly impaired. It is concluded that soman intoxication expresses a stimulatory or inhibitory effect on the processes of protein synthesis in the rat brain, depending on the dose schedule of soman administration.

    Notes: Summary Lysis of mitochondria from sea urchin embryos with Triton X-100 led to a complete conversion of DNA-containing mitochondrial residues into protein-DNA complex with a density higher than 1.22 g/cm3 in sucrose solutions. This complex banded isopycnically in metrizamide gradients at a density of 1..26 g/cm3. Exposure to mixtures of Triton X-100 with Tween 80 resulted in progressively less delipitated and disorganized mitochondria over Tween/Triton weight ratios from 1 to 2, with the retention of the starting buoyant density in sucrose of approximately 1.16 g/cm3 at Tween/Triton ratios above 2.5. The DNA-internal protein complex sedimented with the bulk of the surviving mitochondrial structure under all conditions studied. No free DNA could be detected under any conditions of membrane removal.

    Notes: Abstract A 29 kD soluble rat liver nucleoprotein (p29) has increased binding affinity for the hormone responsive element (RE) of the rat haptoglobin (Hp) gene during the acute-phase reaction. In this work the possibility of its structural and functional homology to the high mobility group 1 (HMG1) nonhistone protein constituent of chromatin was examined. The results of two-dimensional gel electrophoresis, Southwestern and Western immunoblot analyses, showed that p29 and HMG1 are homologous protein species. On the basis of in vitro and in vivo phosphorylation/dephosphorylation experiments, we discuss the modulatory role of phosphate groups in view of the structure and function of p29.
