    Notes: Experimental colitis was induced in rabbits by exposing the colon mucosa to 1% formalin followed by i.v. injections of soluble immune complexes made with antigen in excess. The animals were preimmunized with Esherichia coli 014:K7:H — inducing antibodies cross-reactive to intestinal epithelium.Animals with this colitis were divided in two groups. One group was treated with sulphasalazine and the other was given vehicle only. Sulphasalazine was administered daily at 125.5 μmol (50 mg) per kg body weight. The administration was started at the same day as the colitis was initiated. At Day 6, 13 and 30 following induction of colitis, biopsies were sampled and histologically evaluated. Inflammation was assessed by scores for inflammatory cells, crypt distortion, decreased crypt number and presence of crypt abscesses, thus corresponding to the picture seen in humans. A statistically significant lower score of inflammation was seen on Day 6 and 13 (P 〈 0.01) and on Day 30 (P 〈 0.05) following induction of colitis in animals treated with sulphasalazine.

    Notes: An IgE-binding factor(s) (IgE-BF(s)) was partially purified from the supernatant of human HTLV-II carrying T-cell line MO, This IgE-BF(s) was shown to increase the IgE synthesis in the human myeloma cell line U-266, but did not affect its viability or growth. The of effect of the IgE-BF(s) was dose-dependent and selective for IgE protein synthesis as β2-microglobulin synthesis in the U-266 and the immunoglobulin production in the U-1958 IgG-secreting human myeloma cell line were unaffected. The IgE-BF(s) increased the production of the c heavy chain but not the X light chain production. The IgE-BF(s) was distinct from IL-1β. IL-3, IL-4, IL-5, lL-6, TNF-α, IFN-α -β, -γ, M-CSF, and fragments of CD23.

    Notes: The IgA level in rat bile was significantly decreased by drainage of the thoracic duct, and passively administered IgA antibodies to Escherichia coli O antigen decreased similarity. In contrast, specific IgA antibodies against E. coli O antigen raised by immunization in the Peyer's patches did not diminish significantly in the bile. Rats immunized in the Peyer's patches with sheep erythrocytes had IgA-forming cells in the thoracic lymph nodes, in the mesenteric lymph nodes, and in the spleen. Perfusion of the liver of immunized animals significantly decreased the bile levels of the IgA antibodies. It seems that IgA antibodies reach the bile not only via the thoracic duct but also via lymph ducts originating from thoracic lymph glands and the spleen.

    Notes: The potential for differentiation of the human basophilic leukaemia cell line KUS12 was examined by means of a panel of physiologic and non-physiologic substances used as inducers. The phenotypic characteristics of non-induced KU812 cells included an immature morphology with scanty cytoplasmic granulation, expression of a low amount of high affinity, but no low affinity receptors (CD 23) for IgE, and a capacity for low-rate histamine synthesis. The differentiation process was characterized by a rapid (24 h) increase in histamine production, a slower morphological maturation with the development of Alcian blue stainable granula demonstrable after 72 h. Concomitant with the phenotypic alterations, cell growth was inhibited. Differentiation in KU812 cells was inducible by Ara-C and to some extent by sodium butyrate, but not by dimethyl sulphoxide, retinoic acid, or gamma-interferon. Conditioned medium (CM) from cultured peripheral blood cells from atopic individuals and 18 out of 22 analysed glioma cell lines induced differentiation of the KUS12 cells, whereas supernatant from only 1 out of 21 other cell lines, including carcinoma, melanoma, sarcoma, leukaemia, and normal fibroblasts had this activity. CM from the T-leukaemic cell line. Mo, also induced KU812 differentiation. A primary fractionation of the active substance from this cell line by reversed phase chromatography eluted the active substance at a concentration of 42–44% acetonitrile. Our present study has shown that the KU812 may serve as an appropriate model to study differentiation of basophils. In addition, its fast and specific response to biological factors makes it suitable as a biological assay for determination of active factor produced by atopic individuals.

    Notes: IgA and IgM antibodies were detected in rat milk after immunization wish ferritin in Peyer's patches (Pp) 1 day after parturition but not after intramammary gland or intravenous immunization. The antibody levels decreased from day 9 to day 17 of the nursing period and were undetectable during a second lactation period. Despite the absence of milk IgM antibodies after intramammary gland or intravenous immunization, the serum levels of the IgM antibodies were similar after all three immunization methods. IgA antibodies were not found in serum after any of the immunization methods. IgG antibodies appeared in serum and milk after Pp. intramammary gland, and intravenous immunization. Milk and serum IgG antibodies from all the Pp-immunized animals decreased from day 9 to day 17 of the lactation period. After intramammary gland immunization, however, the IgG antibody levels increased in all the milk samples, but only in four of seven sera. The milk and serum IgG antibody levels were lower but still detectable during a second lactation period. Re-injection of ferritin in the Pp during a third lactation period resulted in higher levels of milk IgA, IgG and IgM antibodies than after the first injection, Rats with serum IgG anlihodies against Escherichia colia 08 naturally present in their gut flora had no corresponding milk antibodies of any isotype The results suggest that milk antibodies of all three isotypes stem from local production in the mammary gland and that blood IgG and IgM antibodies originate at least partly from stimulation in Pp.

    Notes: Intravenous administration of ducts thoracicus lymph with dimeric IgA antibodies against Escherichia coli 06 to lactating rat dams did not result in transfer of IgA antibodies into the milk, although the antibodies were detectable in serum 1 min after the administration and in bile 60 min later. After intravenous injection of serum from bile-duct-occluded (BDO) rats Immunized in the Peyer's patchs into lactating rat dams, IgA antibodies appeared in the serum and remained there up to 230 min. At this time no IgA antibodies were seen in the milk while they were present in bile. IgG and IgM 06 antibodies did not appear in bile or after intravenous administration of lymph or serum from BDO rats.

    Notes: Drainage of the thoracic duct resulted in a decrease in the IgA level in rat bile, but at the same time there was an increase in both the total IgM level and the specific IgM antibody activity to Escherichia coli 06 in the bile of animals immunized in the Peyer's patches with these bacteria. The increase in total IgM was significantly higher in animals immunized with ihe E coli 06 than in unimmunized rats. The level of total IgG was not altered during the drainage. IgM antibodies to E. coli 04 given intravenously during lymph drainage did not appear in the bile, whereas specific IgM antibodies to E. coli 06 occurring after active immunization increased in the bile of the same animal. The data elucidate two aspects of the hepatic IgM turnover. First IgM could take the place of IgA in cases of IgA deficiency, and second the IgM might originate from intrahepatic production.

    Notes: Rats were immunized with ovalbumin, either subculaneously or by aerosol inhalation. The lymphocyte distribution in lymph nodes, peripheral blood, and spleen was investigated by flow cytometry after labelling with T pan (0X19 and W3/13), T helper lymphocytes (W3/25), T cytotoxic/suppressor lymphocytes (OX8), k light chain (MAR 18-5), or MHC class II (0X6) monoclonal antibodies. The influence of the neurotoxic agent capsaicin on the lymphocyte distribution was also analysed. Subcutaneous immunization resulted in an increased number of OX8+ cells in mesenteric lymph nodes, spleen, and peripheral blood but not in the draining lymph nodes, axillary, brachial, and mediastinal lymph nodes. The number of positive cells for the other cell markers used were not affected by immunization. The neuromodulatory effect of capsaicin had no effect on the lymphocyte distribution. The results showed that the type of immunization used, low amounts of antigen without adjuvant given during a prolonged period, selectively induced OX8+ cells. The patterns were unaffected by neuromodulation using capsaicin.

    Notes: Human bone marrow, cord blood, and peripheral blood comntain progenitor cells, which during culture mature to histamine-containing basophilie cells. In bone marrow the histamine content per Alcian blue staining basophilie cell was low before culture. Cultivaiion of BML cells resulted in increased histamine levels in cultures(P〈0.05). whereas the basophilic cells did not increase significantly. Cell cultures were stimulated with conditioned medium (CM) produced with allergen-slimulated cells from atopies and from the Mo T leukaemie cell line. Cells from cultures slimulaled with CM contained less histamine calculated per basophilie cell than did those from unstimulted cultures (P〈5.05), There was a significant correlation between the numbers of basophilic cells and the histamine content in cells on day 0 prior to cultivation and after 14 days of cultivation (P〈0.01 and P〈0.05 respectively). In cord blood there was a correlation between the numhers of basophilie cells and the histamine levels prior to cultivation (P〈0.05). During cultivation the number of basophilie cells increased five-lold (P〈0.02), whereas the histamine levels did not inercase resulting in a decreased histamine level per basophilie cell (P〈0.02). In peripheral blood the basophilic cells contained the highest levels of histamine. The numbers of hasophilie cells and their conntent of hisiamine showed good correlation both hefore and after unstimtilated and stimulated cultivation (P〈0.01). whereas unstimulated cultures did not show such correlation. The results indicate the presence of different proportions of progenitor cells in bone marrow, cord blood. and peripheral blood. all with different ability to produce histamine and become granulated basophilic cells.
