    Notes: —Dexamethasone administration to newborn rats resulted in an increase in phenylethanolamine N-methyltransferase activity in superior cervical ganglia. The same treatment resulted in a decrease in tyrosine hydroxylase and choline acetyltransferase activities. The decline in choline acetyltransferase activity after dexamethasone treatment was only seen when the drug was given before 5 days of age. When dexamethasone was given to pregnant rats it caused an age-dependent decrease in choline acetyltransferase in the adrenals, superior cervical ganglia and para-aortic chromaffin tissue of the offspring.These results suggest that dexamethasone administration may delay the development of the preganglionic neurons of sympathetic ganglia. This in turn would delay the development of tyrosine hydroxylase but not phenylethanolamine N-methyltransferase.

    Notes: —Three days after superior cervical ganglionectomy of adult Sprague-Dawley rats, the levels of endogenous norepinephrine, the uptake process for [3H]norepinephrine and the activity of tyrosine hydroxylase decreased 99 per cent in the ipsilateral salivary gland. In contrast, the activity of dopamine-β-hydroxylase and DOPA decarboxylase fell to 30 per cent of the activity of the contralateral innervated gland. Examination of the cofactor requirements, the characteristics of activation by cupric ion and the immunologic identity of this residual hydroxylase activity indicated that it was authentic dopamine-β-hydroxylase. The residual dopamine-β-hydroxylase in the denervated gland had the same subcellular distribution as the enzyme in the innervated salivary gland. Procedures that caused atrophy or hypertrophy of the acinar cells did not affect the total content of dopamine-β-hydroxylase in the denervated salivary gland. Chemical sympathectomy with 6-hydroxy-dopamine caused a 40 per cent decrement in the serum levels of dopamine-β-hydroxylase but a 30 per cent increase in its activity in the denervated salivary gland. Although denervation caused a complete loss of endogenous norepinephrine in the salivary gland, it resulted in only a 15 per cent decrement in the levels of endogenous octopamine and β-phenylethanolamine, two other products of dopamine-β-hydroxylase.

    Notes: Abstract— An assay for 3,4-dihydroxyphenylacetic acid (DOPAC) capable of detecting as little as 1 pmole of DOPAC is described. The basis of the assay is the O-methylation of DOPAC utilizing S-[methyl-3H]adenosyl-l-methionine and a partially purified catechol-O-methyl transferase to form [methyl-3H]homovanillic acid. The [methyl-3H]homovanillic acid is purified with ion exchange resin and either solvent partitions or TLC. Utilizing this assay, the effect of either chlorpromazine or reserpine upon the content of DOPAC both in the caudate nucleus and in the substantia nigra was determined. Both of these drugs increase the level of DOPAC in the caudate nucleus but have minimal effects upon the level of DOPAC in the substantia nigra.

    Notes: Abstract— Treatment of neonatal rats with dexamethasone resulted in the appearance of phenylethanolamine-N-methyltransferase (PNMT) and numerous small, intensely fluorescent (SIF) cells in abdominal paraganglia and in sympathetic paravertebral ganglia. These cells may be derived from a primitive stem cell precursor, but because of their unusual anatomical features, origin from ganglion cells cannot be altogether ruled out. Associated with the proliferation of the cells was a marked increase in the PNMT activity of the tissues. The PNMT response to the glucocorticoid was limited to the first few days of life, as was the SIF cell response. After discontinuance of dexamethasone, the enzyme activity fell very rapidly, while the number of cells declined at a slower rate.

    Notes: Abstract— Octopamine is a normally occurring amine in several species of animals. Particularly high concentrations are found in the crustacean central nerve cord. In the rat it is specifically localized to sympathetic nerve endings, has a subcellular distribution similar to that of norepinephrine, and is asymmetrically distributed in the CNS. It has a turnover rate in heart about six times that of norepinephrine. The physiological role of octopamine has not been established but it appears likely that it is a cotransmitter together with norepinephrine in the peripheral sympathetic nervous system.

    Notes: The uptake and storage of L-[3H]norepinephrine at various stages of development was examined in homogenates of rat brain. For the adult animal, active uptake accounted for 80 per cent of the total uptake. At 14 days of gestation, no active uptake was demonstrable At 18 days of gestation, saturable uptake of L-[3H]norepinephrine with a Km of 3 × 10 −7m was first demonstrable; the Km value did not vary during subsequent development. The Vmax. of uptake increased five-fold between 18 days of gestation and 28 days postnatally, at which stage it was the same as the adult value. The development of saturable uptake paralleled but preceded the increase in endogenous norepinephrine. When homogenates were incubated with l-[3H]norepinephrine and subjected to centrifugation on linear sucrose gradients, there was a peak of tritium in the synaptosomal fractions; the magnitude of the peak increased with maturation of the brain. The increase in the peak of tritium paralleled the increase in particulate LDH activity and was distinct from the peak of MAO activity. Desipramine, a compound that blocks the initial uptake of norepinephrine, first exhibited inhibition of uptake at 19 days of gestation; the degree of inhibition did not vary during subsequent development. In contrast, reserpine, a compound which inhibits the intra-neuronal storage of norepinephrine, exhibited a progressive increase of inhibition with maturation of the brain at and subsequent to 19 days of gestation.

    Notes: Abstract— Phenylethanolamine and octopamine have been detected in the developing rat brain. Maximum concentration of these amines occurs early in development (16-17 days of gestation). At this developmental stage, the brain concentration of these amines is higher than that of norepinephrine. There is a sharp decline in the phenylethanolamine and octopamine concentrations on day 18 of gestation to approximately those of the adult. This decrease coincides with an increase in-monoamine oxidase activity of fetal brain, with an increase in the activities of tyrosine hydroxylase and dopamine-β-hydroxylase, and with the appearance of a saturable active uptake mechanism for norepinephrine. The administration of iproniazid, a monoamine oxidase inhibitor, to pregnant rats produced an increase in phenylethanolamine, octopamine and norepinephrine concentrations in the fetal rat brain at 16 days of gestation. p-Chlorophenylalanine, an inhibitor of phenylalanine hydroxylase, decreased fetal brain norepinephrine; this drug increased brain levels of phenylethanolamine and octopamine. The combined administration of iproniazid, p-chlorophenylalanine and phenylalanine to pregnant rats resulted in increased concentrations of octopamine and in a several-fold increase of phenylethanolamine levels; norepinephrine concentrations were sharply reduced. The possible significance of these findings in relation to pathological conditions such as phenylketonuria is discussed.
