    Notes: Summary The compartmentalization of the parenchyma of the corpus luteum in the dog was studied by both 100 and 1000 KV electron microscopy. The organelles within the luteal cell are oriented with a high degree of consistency towards the pericapillary space. Characteristically, the avascular pole and the lateral margins of the cell possess predominantly stacked and whorled cisternae of agranular ER. In the central medial portions of the cell, pleomorphic mitochondria with tubulo-vesicular cristae and anastomosing tubules of agranular ER predominate. However, the distribution of organelles in this compartment is graded. Mitochondria predominate in the central medial areas while tubular ER is more dominant peripherally. Microfilaments are ubiquitous in this compartment and run a longitudinal course between and around the subcellular components towards the pericapillary space. The Golgi apparatus is large and prominent and is positioned over the pole of the nucleus that faces the basal lamina. Coated vesicles are abundant in the Golgi regions and along the lateral surface of the cell. Three distinct regional specializations of the cell surface exist. The basal surface contains long pleomorphic cytoplasmic folds that fill the pericapillary space, are interonnected by small gap junctions and contain abundant multivesicular bodies. The lateral cell surface is covered with microvilli and is organized into tortuous intercellular channels and canaliculi. These are interrupted at intervals by cytoplasmic protrusions that extend from one cell well into the cytoplasm of the next. Large, well-developed gap junctions line the margins of the cells furthest removed from the pericapillary space. Finally, individual cells exhibit heterogeneity with respect to the amount one subcellular organelle or compartment is expressed relative to another. These observations are discussed in relation to the subcellular compartmentalization of progesterone synthesis and release.

    Notes: Summary The transition from ovarian granulosa to lutein cell during the estrus cycle of 60 pregnant and non-pregnant beagle bitches was analyzed by light and electron microscopy (both 100 and 1000 KV). Early proestrus was characterized by a gradual rise in serum estrogen levels, hyperplasia of the granulosa cells, the accumulation of follicular fluid, and the development of tortuous intercellular channels. During the second half of proestrus, serum estrogen levels continued to rise, but growth, division, and differentiation of the granulosa cells was minimal. Estrus was marked by the first acceptance of the male and a well-defined LH peak In the subsequent 24 hour period, the granulosa-lutein cells hypertrophy rapidly and develop a large Golgi apparatus, small profiles of granular endoplasmic reticulum, numerous microfilaments, and large gap junctions between the cells. Mitochondria also proliferate, enlarge, and elongate, but retain lamelliform cristae. Luteinization of the cells and progesterone secretion begin just after ovulation which in turn occurs about 24 hours after the LH peak. On the third and fourth day of estrus, numerous small vesicles of agranular endoplasmic reticulum fill the cytoplasm and the mitochondria swell up and round off. The vesicles rapidly fuse into whorled and flattened cisternae or anastomosing tubules of agranular endoplasmic reticulum, while the mitochondria develop tubulovesicular cristae. These structures gradually become organized with respect to the basal lamina. The Golgi apparatus is centered over the pole of the nucleus that faces the pericapillary space. Stacked and whorled cisternae of agranular ER develop in the lateral margins and avascular end of the cell while mitochondria and tubular elements of agranular ER predominate in the central medial and most basal portions of the cytoplasm. Microfilaments are ubiquitous and appear to be instrumental in this orientation process. The cell surface develops three distinct regional specializations that coincide with the underlying cellular compartment: interconnecting pleomorphic folds fill the pericapillary space; long tenous microvilli project from the lateral cell surface and form tortuous intercellular channels and canaliculi; and large gap junctions form along the margins of the cell furthest removed from the basal lamina. By the sixth day of estrus, the granulosa-luteal cell transition is nearly complete and serum progesterone levels are on the rise.

    Notes: Summary The role of the paraventricular nucleus (PVN) and biogenic amines (BA) in regulating the level of corticoids in the serum of osmotically stressed mallard ducks (Anas platyrhynchos) was analyzed employing three experimental approaches: 1) pharmacologic alteration of central BA levels, 2) microscopic evaluation of BA distribution, and 3) placement of electrolytic lesions into the PVN. Reserpine and α-methyl-p-tyrosine (αmpt), agents that decrease the amount of BA's in the central nervous system, produced a fivefold increase in the concentration of serum corticoids. Conversely, pargyline and amphetamine, agents that increase the functional pool of BA's, prevented the rise in serum corticoid concentration normally observed in birds challenged with an intraperitoneal injection of hypertonic saline. When the topographic distribution of BA's was analyzed in the brains of osmotically stressed and nonstressed ducks distinct changes in the intensity of catecholamine (CA) fluorescence were observed in only one location, the PVN of the hypothalamus. Additionally, electrolytic lesions stereotaxically placed in the PVN blocked the osmotic stress-induced rise in serum corticoid concentration. These data therefore indicate that the PVN in the mallard duck plays some role in regulating the observed stress-induced rise in serum corticoid concentration, and that this regulatory function is probably inhibited by catecholamines.

    Notes: Summary Exposure to hypoxia caused an increase in the hematocrit and right heart weight of experimental rats, but did not affect calcium-45 uptake by pulmonary arterial smooth muscle cells. However, autoradiographic studies showed that hypoxia apparently caused a shift of 45-Ca from primarily extracellular sites in arteries of control rats to intracellular sites in tissues of hypertensive rats. Cytochemical studies of calcium distributions in pulmonary arterial smooth muscle cells support the autoradiographic data and show that in both rats and swine the majority of pyroantimonate granules occur extracellularly in control tissues. In contrast, hypoxic tissues displayed a greatly reduced number of granules in extracellular sites and an increase in the amount of precipitate in intracellular sites. In pulmonary arterial smooth muscle cells from hypoxic rats most of the precipitate was associated with the caveolae intracellulares, while in corresponding cells from hypoxic swine the majority of the pyroantimonate granules were localized to the sarcoplasmic reticulum. Hypoxia may produce pulmonary hypertension by interfering with the ability of the arterial smooth muscle cells to maintain transmembrane ionic gradients, thus producing an effective increase in cytoplasmic calcium levels. The increased calcium may then activate the contractile apparatus to produce a sustained vasoconstriction.

    Notes: Abstract Two, healthy, 4-day-old calves were fed an enteropathogenic agent and fatal diarrhea ensued. Biopsies and mucosal scrapings from the ileum and jejunum were taken before introduction of the agent and at 24, 48 and 72 hours thereafter. The biopsies and scrapings were used to analyze the changes in cytochemical localization and biochemical activity of a variety of phosphataases. Within 24 hours, adenosine triphosphate phosphohydrolase activity was almost totally inhibited while the activity of nonspecific phosphatase and nucleoside phosphatase was only partially inhibited and that of Na+-K+ ATPase not effected at all. In preinoculation control sections, the localization of ATPase activity was heaviest in the apices of absorptive epithelial cells located at the base of the villi. This reaction was completely lost within 24 hours after the inoculation with virus. The enzymatic inhibition occurred 12 to 24 hours before diarrhea became evident.

    Notes: Uteri taken from 25 bitches at various times during the early stages of pregnancy were studied cytologically to determine how the implantation chamber developed and how fetal-maternal relations were established. On day 13 after the end of estrus, knobs of trophoblastic syncytium formed and became wedged between cells of the uterine luminal epithelium. The syncytium quickly spread along the uterine lumen and into the mouths of the glands, dislodging and surrounding maternal cells. As invasion continued trophoblastic villi, consisting of cores of cytotrophoblast covered by a continuous layer of syncytium, penetrated deeper into the endometrium. The syncytium spread to surround maternal vessels and decidual cells. By day 26 the trophoblast had extended down to the large lacunae. Here syncytial trophoblast covering tips of the villi degenerated, leaving cytotrophoblast exposed to the necrotic zone. These cells possessed characteristics of absorbing cells. Hematomas were formed by focal necrosis of fetal and endometrial tissue at the poles of the implantation sites. Large pools of extravasated blood accumulated and red blood cells were phagocytized by surrounding trophoblastic cells.Therefore, the endotheliochorial relationship in the canine placenta appeared to be established by syncytial trophoblast invading a cellular endometrium. In the necrotic zone and hematomas, cellular trophoblast may have lost its syncytial covering, but elsewhere maternal vessels and decidual cells in the placenta were in direct contact only with syncytial trophoblast.

    Notes: The lobular, compound, branched, tubular, salt-secreting lacrymal glands of two marine turtles, Chelonia mydas and Caretta caretta are similar in structure and in histochemical reactivity. Blood from the centrolobular arteries flows through a rich capillary bed counter to the flow of tubule secretion. The capillary endothelium is reactive for adenosine triphosphatase (ATPase). Nerves containing cholinesterase pervade the connective tissue. At the blind ends of the secretory tubules small basophilic peripheral cells contain an abundance of glycogen, monoamine oxidase (MAO) and phosphorylase but little succinic dehydrogenase (SDH) or cytochrome oxidase (CTO). Non-mitochondrial ATPase is concentrated at the luminal interface of these cells. The larger principal cells, lining the major portion of the secretory tubules, are rich in SDH and CTO but contain relatively little glycogen, MAO or phosphorylase. Broad intercellular channels reactive for mucopolysaccharide are formed by intermeshing, pleomorphic microvilli that fringe the extensive lateral surfaces of the principal cells. The cytoplasm of these cells contains profiles of smoothsurfaced endoplasmic reticulum (SSER), abundant mitochondria, and prominent Golgi membranes. Profiles of SSER and small membrane bound vesicles fill the apical cytoplasm but mitochondria are lacking. The luminal secretory border of the cell is extremely limited in area.Two types of epithelial cells line the duct system: basal cells that react strongly for non-specific esterase and MAO; and goblet cells containing mucopolysaccharide, acid phosphatase, cholinesterase, and ATPase.The principal cells, close to the arterial blood supply, contain the highest concentrations of oxidative enzymes and have special modifications of the cell surface consistent with their role in salt concentration and secretion.

    Notes: The endometrium of 40 cycling bitches was studied using cytological, cytochemical, and morphometric techniques. Two principal phases of growth and differentiation can be discerned. Phase one begins at the end of anestrus as serum estrogen levels begin to rise and is completed just prior to estrus. It is characterized by growth of the crypts and differentiation of the glandular epithelial cells into well-developed, mucus-secreting cells. Growth, initially rapid, gradually slows. The second growth phase does not begin until the middle of estrus as serum progestin levels rise and lasts nearly a week. Both hypertrophy and hyperplasia of the glandular epithelium and growth of the basal glands characterize this stage. The gland cells develop many well-defined characteristics of absorptive and secretory cells. Another phase of growth occurs in pregnant animals at the onset of implantation. During the third week of metestrus in non-pregnant bitches, the uterus begins to involute. Acid phosphatase and the number of lysosomes increase dramatically in the epithelial cells particularly in the basal glands. Cells lining the lumen and crypts accumulate numerous large lipid droplets.The data are discussed in relation to the clear separation of two distinct uterine functions: (1) sperm transport and maintenance and (2) production and secretion of nutritive uterine milk. Extended periods of follicular development, breeding, and preimplantation in the bitch probably impose this separation.
