    Notizen: Abstract An isolated brain preparation was used to characterize neurons innervating the accessory lobe (AL) of the spiny lobster (Panulirus argus). Four distinct classes of neurons responded to electrical stimulation of the olfactory (antennular) nerve. These cells responded to electrical stimulation with a long and variable latency; they also responded to odor stimulation in a nose-brain preparation. Neurons connecting the AL with the olfactory lobe branched in the central AL layer and selectively innervated olfactory lobe glomeruli. These cells had response latencies which were significantly shorter than those of other AL neurons. Intrinsic AL interneurons were heterogeneous as a population, and most arborized in irregular but circumscribed regions of either the lateral or medial layers. The final class of neurons branched ipsilaterally in the deutocerebral neuropil and bilaterally innervated only a few AL glomeruli. The physiology and morphology of these four classes of neurons confirm an olfactory function for the AL and identify the input and output regions of the lobe. Based on these findings, we propose that the AL processes odor information in the context of higher order multimodal input.

    Notizen: Abstract Antisera to GABA and histamine (HA) label distinct populations of interneurons that innervate glomeruli in the olfactory lobe (OL) of the spiny lobster. GABA-immunoreactive interneurons branch most heavily in the cap of the glomeruli, while HA-immunoreactive interneurons branch mostly in the glomerular subcap. Perfusing GABA or HA into the isolated brain increases the intensity of electrical stimulation of the antennular nerve necessary to elicit action potentials in OL projection neurons. The GABA receptor antagonist picrotoxin (30–100 μmol · l−1) and the HA receptor antagonist cimetidine (1–5 mmol · l−1) both reduce the stimulus intensity needed to elicit action potentials. However, cimetidine also eliminates the hyperpolarizing phase of the evoked response and reveals a delayed, prolonged excitation of up to 10 s, whereas picrotoxin enhances the hyperpolarization and, at higher concentrations, transiently suppresses all phases of the evoked response. We conclude that GABA- and HA-ergic interneurons constitute two overlapping, yet functionally distinct inhibitory pathways in the OL, an organizational feature which may be fundamental to processing at this level of the olfactory pathway.

    Notizen: Abstract Whole-cell patch-clamp recording was used to characterize olfactory projection neurons in an isolated brain preparation of the spiny lobster, Panulirus argus. Responses to electrical stimulation of the olfactory afferents were recorded from projection neuron somata using biocytin-filled electrodes. All projection neurons were multiglomerular, innervating up to 80% of all olfactory lobe glomeruli, but the innervation was heterogeneous. Most neurons densely innervated only 3–4 glomeruli; the remaining glomeruli in their dendritic arbor were sparsely innervated, thereby creating two distinct patterns of intraglomerular branching. Projection neurons responded to orthodromic stimulation with an initial depolarization and spiking followed by a 1–3 s hyperpolarization. The inhibitory phase of the response was lower in threshold and longer in latency than the excitatory phase, a response pattern also reported in olfactory projection neurons of insects and vertebrates. The somata of the projection neurons supported voltage-activated currents and TTX-sensitive action potentials, suggesting that the soma, although spatially separated from the axon and dendrites, may play a significant functional role in these cells. Dye coupling between some projection neurons correlated with the presence of multiple amplitude action potentials, suggesting that at least some projection neurons may be coupled via gap junctions.

    Notizen: Abstract Neurons in the olfactory deutocerebrum of the spiny lobster, Panulirus argus, were recorded intracellularly and filled with biocytin. Recorded neurons arborized in the olfactory lobe (OL), a glomerular neuropil innervated by olfactory and some presumptive mechanosensory antennular afferents. The neurons responded to chemosensory input from the lateral antennular flagellum bearing the olfactory sensilla but not the medial flagellum bearing many non-olfactory chemosensory sensilla. Many neurons received additional mechanosensory input. Thus the OL integrates specifically olfactory with mechanosensory input. OL neurons had multiglomerular arborizations restricted to one or two of the three horizontal layers of the columnar glomeruli. OL local interneurons comprised “core” neurons with tree-like neurites and terminals in the base of the glomeruli and “rim” neurons with neurites surrounding the OL and terminals in the cap/subcap. The somata of OL local interneurons lay in the medial soma cluster (100000 somata). OL projection neurons arborized in the base of the glomeruli and ascended via the olfactory glomerular tract to the lateral protocerebrum. A parallel projection pathway is constituted by projection neurons of the accessory lobe, a glomerular neuropil without afferent innervation but intimate links to the OL. The projection neuron somata constituted the lateral soma cluster (200000 somata).

    Notizen: Abstract Neurons in the brain of the spiny lobster that respond to chemical and mechanical stimulation of the antennule (antenna I) were recorded and stained intracellularly. Described here are neurons that do not arborize in the olfactory and accessory lobes of the deutocerebrum, but rather primarily target the lateral and/or the median antennular neuropils of the deutocerebrum. Some of the neurons also extend into the antennal and tegumentary neuropils of the tritocerebrum and the neuropils of the median protocerebrum. Included are antennular sensory afferents, antennular motoneurons, projection neurons descending from the central brain, projection neurons ascending from the central brain and projection neurons descending from the eyestalk ganglia. Collectively, these neurons consitutute a novel antennular sensory pathway that is parallel to and independent of the antennular olfactory pathway. The novel pathway integrates mechanosensory and non-olfactory chemosensory information in the lateral and/or the median antennular neuropils, which also serve as lower motor centers of the antennule. Division of the arthropod deutocerebrum into two, functionally distinct chemosensory pathways may reflect differences in how chemosensory information is processed that is fundamental to understanding the origin of the sense of smell.

    Notizen: Abstract This study characterizes odor-evoked responses of the glomerular output neurons of the spiny lobster olfactory lobe, and implicates previously identified γ-aminobutyric acid (GABA)- and histamine-mediated inhibitory pathways in shaping these responses. Odor-evoked responses were more complex than electrically evoked responses, with up to three distinct components: a brief, short-latency (fast) depolarization, a longer-duration, longer-latency (slow) depolarization, and a slow hyperpolarization. Seventy-seven percent of all responses contained the hyperpolarization, while only 31% and 23% contained the fast and slow depolarizations, respectively. The broader tuning of the hyperpolarization relative to the other two components suggests that the hyperpolarization mediates lateral inhibitory interactions across olfactory glomeruli. Perfusing the brain with the GABA-receptor antagonist picrotoxin increased the amplitude of the hyperpolarization, while the histamine-receptor antagonist cimetidine decreased the hyperpolarization in some instances but increased it in others. Pharmacological enhancement or suppression of the hyperpolarization could mask or unmask, respectively, the slow depolarization. Both antagonists could also cause the appearance of the fast depolarization when it was not apparent prior to treatment. We conclude that GABA- and histamine-mediated inhibition contributes to the overall pattern of the response of projection neurons to odors by regulating the relative strength of these three distinct types of input.
