    Notes: The development of the heart-conducting system has been controversially discussed. The common opinion that these specialized myocytes originate from mesodermal precursors has been challenged when nerve-specific antigens (Leu-7, NF, G1N2) were demonstrated in embryonic hearts of various species, suggesting a neural crest contribution to the embryonic conducting tissue. Anti-Leu-7 (HNK-1) antibodies were reported to reliably mark the conducting system in developing rat, chicken and human hearts. The present investigation was carried out on the hearts of 15 camel fetuses at 35, 45, 60, 75 and 100 cm crown-rump length (three specimens for each stage), in addition to three adult hearts. We investigated the antigenicity of cardiac structures for Leu-7, NSE (Neurone specific Enolase) and PGP (Protein Gene Peptide) 9.5. In all specimens investigated, both NSE and PGP 9.5 were expressed by cardiac nerves and conducting system components. The sinuatrial and atrioventricular nodes, the atrioventricular bundle as well as subendocardial and intramyocardial Purkinje fibers were stained. In contrast, the developing conducting system did not react with anti-Leu-7 antibody, although Leu-7 antigenicity was strongly expressed by the developing cardiac nerves. In adult camel hearts, the same pattern of immunoreactivity for the markers studied was still retained. Our results show that the expression of marker proteins for the developing conducting system is species-specific. Therefore, these markers are of little significance in discussions on the possible neurogenic nature of the heart conducting tissue.

    Notes: Digital cushions were studied in horses with particular reference to vascularization, tissue constituents and matrix components. The cushions mainly resembled a network of coarse collagen bundles. The areas inbetween the bundles were replenished with loosely woven interstitial connective tissue, myxoid tissue, and fibrocartilage. Expected masses of fat lobules were missing: only solitary adipocytes or small groups of adipocytes were seen. Vascular supply to the cushions was remarkably poor. The mucinous myxoid matrix largely consisted of hyaluronan with little sulphated glycosaminoglycans. Myxoid cells were stellate or ramified in shape and showed a tendency to store glycogen and lipid droplets. Myxoid cells reacted for vimentin and stained for S-100 protein. Moreover, myxoid cells often reacted for neuron specific enolase and glial fibrillary acidic protein. Myxoid tissue continuously transformed into loosely organized interstitial connective tissue with fibroblasts, which remained unreactive when tested for neuroectodermal markers. Myxoid tissue also was not clearly demarcated against irregularly interspersed islets of fibrocartilage or hyaline cartilage. Chondrocytes did not stain for neuron specific enolase but reactivity for S-100 protein and glial fibrillary acidic protein was noted in peripheral regions of fibrocartilage. Single or grouped unilocular fat cells were rarely placed into myxoid areas. Unilocular fat cells stained for vimentin, S-100 protein, and occasionally for glial fibrillary acidic protein but not for neuron specific enolase. Continuous transformation of myxoid tissue into cartilage together with corresponding reactivity for neuroectodermal marker proteins of myxoid cells and peripherally located chondrocytes suggest close relationship between myxoid cells and chondrocytes. The same criteria indicate relationship between myxoid cells and adipocytes. Coarse connective tissue, myxoid tissue, fibrous cartilage, and fat cells are functionally combined to absorb mechanical shock in the horse digital cushions.

    Notes: Summary Differentiation of cellular cartilage was studied in the mouse pinna with particular reference to matrix material. Fixation of glycosaminoglycans was performed by the use of acridine orange and elastin was identified by staining thin sections with tannic acid and uranyl acetate. Condensation of mesenchymal cells (“prechondroblasts”) initiates the formation of a blastema of cartilaginous tissue at postnatal day 4. The synthesis of acidic glycosaminoglycans begins at postnatal day 8 when prechondroblasts transform to chondroblasts. Glycosaminoglycans can be detected within secretory vesicles of chondroblasts at postnatal day 8, in the extracellular space at postnatal day 13. Delicate collagen fibrils and elastic fiber microfibrils are seen between prechondroblasts and chondroblasts. Deposition of elastin begins at postnatal day 11. A network of elastic fibers and lamellae is formed, which replaces both collagen fibrils and elastic fiber microfibrils. In the interstice of mature cellular cartilage only elastin and proteoglycans are present (postnatal day 21). These findings indicate that cellular cartilage represents an independent kind of supporting tissue, which may serve as a progenitor of hyaline or elastic cartilage (“transitional cellular cartilage”) but does not differentiate from hyalin cartilage.

    Notes: Summary The microvasculature of the esophagus was studied by scanning electron microscopy of vascular corrosion casts in human infants and rabbits. In both species, segmental circumferential arteries arise from main longitudinal arteries, the latter giving off numerous perforating arteries. The tunica muscularis is supplied by branches of circumferential and perforating arteries, the submucosa and its glands by branches of perforatings. Terminal arborizations of perforating arteries feed a subepithelial capillary network. These capillaries are drained by a venous plexus in the lamina propria which is connected to a submucosal venous plexus. Perforating veins, running parallel to the corresponding arteries, connect the submucosal plexus with circumferential veins, and finally empty into main longitudinal veins. Valves were not present in any of the veins. Submucosal veins were less numerous in man than in rabbit. The number and caliber of equivalent vessels in human submucosal plexus decreased from the pharyngoesophageal to the gastroesophageal junction, suggesting the latter to be at particular risk in portal hypertension. The subepithelial capillary network reveals a longitudinal arrangement in rabbits, while the same network shows no preferential organization in human infants. The microvascular architecture of the esophagus in humans and rabbits is comparable, especially in the lay-out of the venous plexuses and the absence of venous valves. Therefore the rabbit could serve as an experimental model for studies on portal hypertension. The present results strongly suggest particular significance of the venous plexus in the lamina propria for the genesis of esophageal varices.

    Notes: Summary The elastic fiber system in the human tracheal and bronchial mucosa was studied by light and electron microscopy. Elastic fibers, elaunin fibers, and oxytalan fibers were discerned. These fibers were identified by means of their staining characteristics (elastica stains, methods for disulfide-groups) and on account of their fine structural morphology. Elastic fibers consist of elastin and few “elastic-fiber microfibrils”. The relative amount of elastin (compared to the amount of elastic-fiber microfibrils) is large in elastic fibers but small in elaunin fibers. Oxytalan fibers — by contrast — are pure bundles of microfibrils. In the light microscope a well-defined elastic lamina separates the lamina propria and the submucosa of the normal mucous membrane. The elastic lamina is formed by coarse strands of longitudinally running elastic fibers. A delicate network of elastica-positive fibers is attached to the basement membrane of the epithelial layer (subepithelial elastic layer). A few of these elastica-positive fibers branch out, traverse the region of the thickened basement membrane, and insert into the basal lamina of the epithelium. A loose network of elastica-positive fibers is present both in the lamina propria and in the submucosa. Plates of cartilage, glandular epithelium, and bundles of smooth muscle cells are enveloped by delicate elastica-positive fibers. Electron microscopy shows the lamina elastica to be predominantly composed of elastic fibers, whilst elaunin fibers from the subepithelial elastic layer. Fibers penetrating the thickened basement membrane of the epithelium are identified as oxytalan fibers. All three types of fibers are present throughout the lamina porpria and in the submucosa. Elaunin fibers and oxytalan fibers comprise the elastica-positive nets around glandular epithelium, smooth muscle bundles, and cartilage. The preferred location of oxytalan fibers (within the thickened basement membrane), elaunin fibers (subepithelial elastica-positive layer), and clastic fibers (lamina elastica) facilitates the comparison of light microscopic staining reactions and fine structural morphology of these fibers.
