The determination of polymer conformation by means of ORD and CD requires the existence of COTTON effects and consequently of absorption bands. For polysaccharides, chromophoric groups must be furnished which make no significant alteration of the polymer's properties.Polysaccharide-Congo Red complexes fulfil this postulate; their ORD- and CD-spectra exhibit several strong induced COTTON effects. In case of complete saturation, 15 glucose units are equivalent to one dye molecule in the amylose-Congo Red complex, and the equilibrium constant of complex formation is found to be K ≥ 3 · 104 [1 Mol-1].The complex structure comprises monomeric dye molecules bound separately to the polymer. The dissymmetry factor determined for λ = 51 5mμ ( g 515 = 2,6 · 10-4) shows the induced COTTON effects to be originated by interactions between Congo Red and asymmetric carbon atoms of the base units. Evidently, this is the cause for opposite-sign COTTON effects in the case of Congo Red-salep mannan, a polysaccharide with a stretched secondary structure.

Die Bestimmung von Polymerkonformationen durch ORD- und CD-Messungen ist nur möglich, wenn Absorptionsbanden und demzufolge COTTON-Effekte vorhanden sind. Im Falle der Polysaccharide müssen Chromophore eingeführt werden, ohne daß jedoch dadurch die Eigenschaften des Polymeren entscheidend verändert werden dürfen. Polysaccharid-Kongorotkomplexe erfüllen diese Bedingung; ihre ORD- und CD-Spektren weisen mehrere starke induzierte COTTON-Effekte auf. Bei vollständiger Sättigung liegt im Amylose-Kongorotkomplex ein Verhältnis von 15 Glucoseresten pro Kongorotmolekül vor; die Gleichgewichtskonstante der Komplexbildung ist K≥3 · 104 [1 Mol-1].Der Komplex entsteht durch die Anlagerung isolierter Kongorotmoleküle an das Polymere. Aus dem Dissymmetriefaktor bei 515 mμ (g515 = 2,6 · 10-4) ist zu schließen, daß die induzierten COTTON-Effekte auf der Wechselwirkung der Farbstoffmolekeln mit asymmetrischen Kohlenstoffatomen der Grundbausteine beruhen. Offenbar ist dies auch der Grund dafür, daß Mannan, das eine gestreckte Sekundär-struktur besitzt, einen Kongorotkomplex mit entgegengesetzten COTTON-Effekten bildet.
