    Notes: SUMMARY.— The fine structure of periderm and germinative cells of the bilaminar epidermis of a 14 mm. human embryo is described. Desmosomes at different stages of differentiation are present at intervals along the apposed plasma membranes of cells of both layers, but hemi-desmosomes have not yet developed along the basal plasma membranes of the germinative cells. Cytoplasmic tono-filaments are not diffusely distributed within the germinative cells, but a continuous band of filamentous material lies just within, and parallel with the basal plasma membrane. Cells occasionally present between periderm and germinative layers exhibit features suggesting they may be immigrant mesenchymal elements.

    Notes: Abstract Using a clonogenic assay in vitro, it has been shown that exposure to azelaic acid “1-100 mM” for 24 hours has a dose-dependent effect on the survival of the colony-forming ability of murine “B16” and human “HMB2, and SK23” melanoma cells as compared with a non-melanotic non-tumoral Chinese hamster cell line “CHO”. Both human cell lines were more sensitive to the diacid than the murine cells, and the HMB2 cells were more sensitive than the SK23 cells. These differences may be partly correlated with differences in pigmentation and doubling times between the three melanoma cell lines. The two human lines were more pigmented than the B16, and the SK23 more than the HMB2; the human lines had a longer doubling time than the others.

    Notes: SUMMARY.— Clinical, histological and ultrastructural features of digital fibrous swellings in children are described. The clinical progress of these cases indicates that the lesions tend to regress spontaneously, and that they are recurrent only in the sense that growth may continue after incomplete surgical excision. A characteristic feature of the lesions is the occurrence of cells with dense cytoplasmic deposits. These cells appear to be abnormal fibroblasts, but electron microscopy provides no clues as to the origin, nature, or causation of the cytoplasmic deposits. No evidence was obtained to support a suggestion that the lesions may have an infective viral origin. Elastic fibres were very scarce within the swellings, and showed some evidence of abnormality. Collagen fibres, constituting the bulk of the swellings, appeared essentially normal.

    Notes: Lentigo maligna and malignant melanoma can be treated by dicarboxylic acids (C9 and C12, which are competitive inhibitors of tyrosinase. We therefore studied the intracellular location and possible sites of action of dodecanedioic acid (C12) in murine melanoma cells, using EM autoradiography and biochemical analysis of lipid extracts by HPLC. Significant levels of radioactivity were found in the mitochondria and in the nuclei but not in association with membranes of rough endoplasmic reticulum, Golgi-associated endoplasmic reticulum, or Golgi apparatus, and not in coated vesicles or melanosomes. Biochemical analysis revealed that the diacid underwent β-oxidation, which occurs only in mitochondria. The results suggest that the toxicity of dicarboxylic acids in melanoma cells is not related to anti-tyrosinase activity but may be due to interference with oxidoreductase enzymes in the mitochondria and possibly to inhibition of DNA synthesis in the nucleus.

    Notes: Summary Large numbers of dendritic cells similar in structure to Langerhans cells of normal epidermis and other epithelia were observed within nodules of a tumour identified as a bronchiolar-alveolar tumour of primary alveolar origin. Though Langerhans cells have never yet been seen in normal lung, it is argued that the present observations provide circumstantial evidence for their probable presence, perhaps in very small numbers, in this situation. Such a suggestion would also render more explicable the characteristic feature of pulmonary lesions of Histiocytosis-X.

    Notes: Summary Freeze-fracture studies have been made on the sciatic nerve of rats with chronic streptozotocin-induced diabetes mellitus. The density of intramembranous particles was reduced in both the P and E faces of the axolemma of myelinated and unmyelinated axons, in myelin and in the perineurial cells. This may reflect a general reduction in protein synthesis, or excessive protein degradation, related to the diabetic state. The perineurial cells also showed gap junctions which are not normally present in adult rat peripheral nerve. These may represent a reaction to changes in perineurial activity consequent to alterations in the endoneurial tissue fluid.

    Notes: Summary Junctional complexes have been investigated in the epiblast of young chick embryos by examination of freeze-fracture replicas and of sections of comparable specimens stained with lanthanum nitrate. By means of freeze-fracture, tight junctions were shown to be present in the unincubated embryo (stage 1 of Hamburger and Hamilton). The number of ridges or grooves was found to vary between 2 and 10 near the dorsal border, whereas isolated ridges were found more ventrally. Lanthanum was unable to penetrate between the cells in the region of the dorsally situated tight junctions. Similar tight junctions were found in incubated embryos (stage 3) examined by both techniques. Tight junctions were also seen in cleavage (pre-laying) embryos examined in section. Gap junctions were extremely uncommon in unincubated embryos, though occasional aggregates of gap junction particles were seen on the lateral cell membranes close to the dorsal surface. In only one instance were associated pits visible. By contrast, gap junctions were more frequently encountered by stage 3, and these junctions possessed both pits and particles. Desmosomes were never seen in the freeze-fracture replicas at either stages 1 or 3, though structures which might be developing desmosomes were visible in sections. The functions of both the tight and gap junctions in the young chick embryo are discussed. The results are also considered in relation to recent theories about the way in which gap junctions are formed.
