    Notes: Summary Background Chronic exposure to ultraviolet (UV) radiation induces changes in the skin structure which are mostly found in the superficial dermis and at the dermal–epidermal junction. Keratinocytes and fibroblasts contribute both to the synthesis and to the degradation of the molecules important for the integrity of this skin site. While several studies have reported on alterations of dermal components and of the functions of fibroblasts in vivo and in vitro after UV exposure, recent data suggested that keratinocytes could be the main skin cell type involved in the photoageing process. Objectives In this study, we analysed the expression of two keratinocyte molecules namely, β1 integrin (a proliferation marker) and involucrin (a differentiation marker) in sun-exposed and sun-protected facial skin of 16 healthy patients undergoing facial lifting. Methods Methods included histology, immunohistochemistry and quantitative reverse transcriptase–polymerase chain reaction analysis. Results Sun-exposed skin displayed the characteristic morphological and molecular features of dermal photoageing, compared with sun-protected skin, including dermal elastosis, diminished fibrillin and type VII collagen expression. Analysis of the epidermis in sun-exposed vs. sun-protected skin showed no histological differences, but dramatic changes in the expression of β1 integrin and involucrin. In sun-exposed skin, expression of β1 integrin protein by epidermal basal cells was reduced, paralleling a downregulation of β1 integrin mRNA, whereas involucrin protein expression was greatly enhanced in the superficial epidermal cell layers. Interestingly, the ratio between involucrin and β1 integrin protein expression was consistently increased in sun-exposed skin sites. Conclusions Collectively these results demonstrate that epidermal homeostasis is impaired by chronic UV exposure, and define β1 integrin expression as a molecular marker of the epidermal photoageing process.

    Notes: Background  Photodamage is characterized by degradation of collagen and accumulation of abnormal elastin in the superficial dermis. Mast cells and macrophages, which are found in higher numbers in photoaged skin, have been implicated in this process.Objectives  To analyse the phenotype of haematopoietic-derived infiltrating cells in photodamaged skin.Methods  Chronically sun-exposed (preauricular) and control sun-protected (postauricular) skin was recovered from eight healthy subjects undergoing cosmetic surgery (facial lifting).Results  Histological analysis showed that sun-exposed skin harboured more infiltrating mononuclear cells than sun-protected skin. Cellular infiltrates were found at the periphery of areas of elastolysis around hair follicles in sun-exposed sites, whereas they were found in the interfollicular dermis around blood vessels and around hair follicles in sun-protected samples. Immunohistochemical analysis revealed an increased number of mast cells, macrophages and CD4+ CD45RO+ T cells in sun-exposed dermis as well as a higher number of CD1a+ dendritic cells in sun-exposed epidermis, compared with the sun-protected samples. Thus photoageing displays histological features of chronic skin inflammation. However, no molecular sign of inflammation was observed and we even found a decreased expression of interleukin-1β mRNA in sun-exposed compared with sun-protected sites. Furthermore, the patients' skin looked normal and did not display any clinical inflammation.Conclusions  Collectively, these data show that chronic ultraviolet irradiation induces alterations of innate immune cells which are recruited in sun-exposed skin without being activated.

    Notes: In anaerobiosis, pyruvate carboxylase activity can be greatly reduced by limiting the availability of biotin to yeast cells. In these conditions an incorporation of carbon from exogenous l-malic acid can be observed. The main pathway used for this anabolic utilization goes through malate dehydrogenase and the oxidative reactions of the tricarboxylic acid cycle. This anabolic function of l-malic acid is strongly inhibited by a residual glucose concentration in the fermenter. This inhibition may be due, in this condition, to a higher intracellular concentration of some negative effectors of malate dehydrogenase such as: 1,6-fructose diphosphate, glyceraldehyde 3-phosphate, ATP and ADP.

    Notes: Abstract A comparison between isothermal and anisothermal alcoholic fermentation is made in this paper. Important differences were observed: in some anisothermal operations maximum rates of CO2 production were reached towards the end of fermentation. Cultures with different initial nitrogen or biotin concentrations showed the importance of thermal conditions for the completion of the reaction. They indicated that the notion of the limiting nutrient does not have the same technological significance with respect to the mode of temperature processing. Thus, some studies at the laboratory scale should not be carried out under isothermal conditions, especially not within temperature ranges which may be critical for cell viability. This is, for example, the case with enological studies about red wine processing.

    Notes: Summary Between January 1987 and January 1991, 110 detubularized U-shaped ileocystoplasties (Camey II) following radical cystectomy were carried out in our Department of Urology (CMC Foch Suresnes, France). Our first evaluation of this procedure was carried out in 1989 and reviewed initial 57 patients operated on. These data were compared with those of the Camey I operation. The improvement in neobladder capacity as well as nighttime urinary control achieved by the detubularization required in the Camey II operation was obvious. In this article we review the first 110 patients treated by Camey II bladder replacement following cystectomy.

    Notes: Abstract Flow cytometric analysis performed on two different crosses of dura×pisifera oil palm gave an accurate estimation of nuclear DNA content. The genome size of Elaeis guineensis was found to be 2C=3.76±0.09 pg and therefore ca. 3.4×109 bp. Embryogenic calli and plants showed the same ploidy level, but the measured 2C DNA values differed significantly. No variation in the ploidy level between three different types of calli originating from foliar explants, namely nodular compact callus, fast-growing callus and friable callus was observed. Since fast-growing callus (FGC), already identified as a source of `mantled' phenotype variants, did not show any difference in their ploidy level, these results are consistent with the hypothesis of an epigenetic origin for this type of somaclonal variant.

    Notes: Abstract A yeast flocculation gene was isolated from a genomic library of an FLO5 strain of S. cerevisiae on the basis of its ability to trigger flocculation in a non-flocculent strain. Characterization of the cloned gene by restriction mapping, Southern analysis, and chromosome mapping have shown that it corresponds to a FLO5 gene previously located on chromosome I and that this gene is related to the already described. FLO1 gene. A study of gene expression in different yeast strains has indicated that, while this gene is dominant, its expression can be suppressed in some genetic backgrounds. A Northern-blot analysis has demonstrated that the same 5000-nt transcript was present in an FLO5 and an FLO1 strain. A gene disruption experiment has led to the conclusion that another flocculation gene is present and can be active in the FLO5 strain we used.
