    Notes: Standard 25-μm-thick polyvinilydene fluoride (PVDF) piezoelectric gauges and new 450-μm-thick P(VDF 70%, TrFE 30%) piezoelectric copolymer have been used to record shock profiles at the back face of metallic targets irradiated by laser pulses of 2.5 and 0.6 ns duration at a 1.06 μm wavelength. The records are fully explained with simplified space–time diagram analysis. The pressure profile applied at the front face of the target has been determined from these records combined with numerical simulations of wave propagation through the target. A numerical code describing the interaction of laser with matter (FILM) has also been used for computing the applied pressure. Both methods lead to very close results. The peak pressure dependence on incident laser intensity is determined up to 30 GPa at 1012 W/cm2. © 1996 American Institute of Physics.

    Notes: Piezoelectric elements made of polyvinylidene-fluoride-trifluoroethylene copolymer P(VDF/TrFE) have been used to measure the pressure induced on the rear surface of metallic foils irradiated by infra-red laser pulses of 1.5 ns duration, with intensities up to 3 TW/cm2. The application of such piezoelectric materials under those conditions is new, and a special effort has been made for their improvement and characterization under plate impact loading. Then, the laser experiments have been carried out. First, peak pressures of 0.5–7.5 GPa transmitted in the copolymer have been derived from the peak voltage measured at each shot, using simple assumptions. Next, a more accurate analysis of the data, involving computer simulations, has provided the pressure profiles characterizing the laser shocks driven on the front surface of the targets, over a wide range of laser intensities. Peak pressures of 7–60 GPa on the irradiated surface have been inferred from that analysis. The results have been compared to values obtained from a classical scaling law on one hand, and to predictions of a laser-matter interaction simulation code on the other hand. An overall coherence has been obtained, despite some discrepancies that have been discussed. © 1996 American Institute of Physics.

    Notes: Galanin was infused intravenously into eight healthy volunteers at a dose of 40 pmol kg−1 min−1 for 1 h to investigate the pharmacological effects of this peptide on the postprandial sialagogical response in man. Galanin significantly increased the salivary volume and the saliva output of sodium, chloride and bicarbonate compared to control saline infusion, but had no effect on the output of potassium and α-amylase. An increase in salivary pH was also observed. The increase in salivary volume may indicate a physiological role of galanin in the control of salivary secretion.

    Notes: Test sites on healthy controls and on the clinically uninvolved skin of psoriatic patients were stripped with tape, and eight variables were quantified at intervals during the subsequent healing process.In the control groups, the stratum corneum regenerated at a constant rate and the underlying skin showed elevations of metabolic activity peaking around days 2–4. In the psoriatic groups, we observed that (I) the response of the keratinizing zone is identical to that of the controls, (2) the proliferative response is initially normal but remains elevated rather longer than usual, and (3) the dermal capillaries (indicated by alkaline phosphatase activity) show a gross hyper-reactivity which is already apparent after I day and which persists for more than a week.These findings support our previous conclusion that metabolic alteration of the dermal capillary precedes epidermal hyperplasia in the pathogenesis of the psoriatic lesion.

    Notes: Abstract: Different subpopulations of mast cells are characterized by their abundant contents of either tryptase or in addition chymase. These two neutral proteases are found in mast cells and may thus hold a key to the understanding of mast cell dependent reactions. Such studies are however hampered by the lack of readily available supplies of chymase. We have therefore studied the simultaneous purification of both proteases from hairless moro hr/hr mouse skin, using a sequence of salt extractions and chromatographic separations. After three steps of extraction, a 13-fold purification with an 82% yield was obtained for tryptase and a 15-fold purification with a 90% yield for chymase. Further one step purification on conventional sephadex, sephacryl and octyl sepharose columns was unsatisfactory because of further protein contamination of the fractions. Heparin affinity chromatography caused a high loss of tryptase and residual protein contamination. Gradient elution on a benzamidine sepharose 6B column resulted however in a single, low yield (17.9%) tryptase peak and a broader, high yield (〉90%) chymase peak, and a 34% yield high purity fraction. The proteases thus purified exhibited their typical inhibitor profile. On Western blot analysis and on autoradiography in the presence of the serine protease inhibitor diisopropylfuorophosphate (DFP), only one 28 kD molecule with chymase activity was identified, whereas a broad 32-38 kD band of tryptase monomers was noted. Taken together, these data show that, after salt extraction and a single benzamidine affinity chromatography step, both mast cell chymase and tryptase can be separated and in case of chymase also highly purified, allowing thus for the study of biological activities of this molecule.
