    Notes: Abstract. Assertions that the ``conventional'' rate of mitochondrial DNA (mtDNA) evolution is reduced in poikilotherms in general and turtles in particular were tested for side-necked turtles (Pleurodira: Chelidae). Homologous data sets of mitochondrial 12S rRNA gene sequences were used to compare the average divergence between the Australian and South American species for two Gondwanan groups: the chelid turtles and the marsupials. The mean nucleotide divergences between continental groups for both the turtles and the marsupials are remarkably similar. These data suggest that the rate of evolution of mitochondrial 12S rRNA gene is not substantially slower in turtles than in the homeothermic marsupials.

    Notes: Abstract Ten individuals of Uromys caudimaculatus sampled from Queensland gave evidence for the occurrence of two distinct chromosome races characterised by marked differences in their pattern of C-banding. In all four individuals from the north, thirteen of the twenty three chromosome which make up the standard haploid set had substantial distal C-blocks in addition to the smaller centric blocks which characterise all chromosomes other than the Y. Additionally two pairs had an interstitial block. By contrast none of the six southern individuals had fixed distal blocks though all of them except the Y carry pro-centric C-blocks and again one pair showed an interstitial block. The southern karyotype was, however, characterised by the presence of from six to nine mitotically stable supernumerary chromosomes all of which were totally C-positive despite the fact that at least five morphologically distinguishable types have been defined. While the relationship of these two types of constitutive heterochromatin remains to be clarified the large amount present in both northern and southern animals suggests that heterochromatin plays an important role in the basic biology of this species.

    Notes: Abstract The presence of rat kappa-chain allotype specificities (RI-1a and b) has been studied in 13 subspecies of the seven native Australian species ofRattus. RI-la reactivity was not detected among these rats. On the other hand, extensive cross-reactivity was seen with RI-1b, some sera cross-reacting totally (R. leucopus cooktownensis), some not at all (R. colletti), and the remainder showing at least two distinct levels ofpartial cross-reactivity, confirming the existence of multiple specificities for Rl-lb. Three subspecies show polymorphism with respect to Rl-lb cross-reactivity (R. sordidus, R. colletti, andR. l. leucopus) and in one case (leucopus) breeding studies have indicated that there is allelic inheritance of this trait. The segregation of RI-1b reactivity has been studied in crosses and backcrosses made between species differing in their RI-1b reactivity, and the results are consistent with the existance of codominant alleles at a single locus. The fact that these species differ extensively in their karyotype opens the door to possible chromosomal localization of this and other genetic traits.

    Notes: Abstract Twenty-six inbred strains of the laboratory rat (Rattus norvegicus) were examined for electrophoretic variation at an estimated 97 genetic loci. In addition to previously documented markers, variation was observed for the enzymes aconitase, aldehyde dehydrogenase, and alkaline phosphatase. The genetic basis of these markers (Acon-1, Ahd-2, and Akp-1) was confirmed. Linkage analysis between 35 pairwise comparisons revealed that the markers Fh-1 and Pep-3 are linked. The strain profiles of the 25 inbred strains at 11 electrophoretic markers are given.

    Notes: In this study we show how the use of exon-primed, intron-crossing (EPIC) polymerase chain reaction (PCR) of a diploid intronic region, in conjunction with temperature gradient gel electrophoresis (TGGE), can be used to detect and rapidly assess allelic variation at the nucleotide level. We developed passerine-specific primers to amplify and sequence a 762 bp region including the second intron of the myoglobin gene in the Gouldian Finch, Erythrura gouldiae. A POLAND plot based on this sequence indicated that TGGE in combination with heteroduplex analysis (TGGE/HA) should reveal nucleotide variation in the 160 bp low-melting domain. Sequencing of the entire fragment from 19 Er. gouldiae revealed five nucleotide substitution differences within the low-melt domain, all of which could be detected and differentiated by TGGE/HA, and an additional substitution in a section of the high-melt domain which characterised another allele. A total of 181 individuals from four populations were screened for these six alleles.
