    Notes: The occurrence of acquired inhibitor against factor IX:C is infrequent in haemophilia B patients and is very rare in previously healthy subjects, in whom it is often related to underlying diseases.We describe the case of a 2-year-old girl, who was referred to our hospital with haematomas, without previous bleeding history. Prolonged APTT, normal PT and a factor IX:C level below 1% were found. An inhibitor against factor IX:C was detected (5.5 U mL−1). Her father and mother showed normal factor IX:C levels. Treatment with high-dose immunoglobulin (400 mg kg−1 day−1 for 5 consecutive days by intravenous infusion) and dexamethasone (4 mg three times a day by intravenous injection for 4 consecutive days) normalized factor IX:C levels and overcame the inhibitor.In conclusion, high-dose immunoglobulin and high-dose dexamethasone are a successful and safe immunosuppressive approach for recovery from inhibitor occurrence.

    Notes: Until recently the Younger Dryas cooling event was thought to be restricted to the North Atlantic region. However, preliminary evidence based on magnetic susceptibility and stable isotope data from Lake Hetongchahannor, a hypersaline alkaline lake in Inner Mongolia indicates that this event is observed in NE Asia. In addition we find indications of wetter climatic conditions between 9000 and 6000 yr BP, possibly due to increased monsoon activity, followed by a progressive aridification towards the present time.

    Notes: Summary Lymphocytes infiltrating human ovarian carcinoma obtained directly from the tumour mass (tumour-infiltrating lymphocytes, TIL) or from the carcinomatous ascites (tumour-associated lymphocytes, TAL) were expanded in vitro in long-term cultures with interleukin-2 and tested for their specific cytolytic activity. Killing of the autologous tumour was detected only in a proportion of the patients, less frequently in TIL compared to TAL. In fact two out of ten TIL and four out of nine TAL cultures tested showed significant levels of lysis against the autologous tumour. This cytotoxic activity was not restricted to the autologous tumour, as other tumour cell lines, including non-ovarian ones, were lysed as well. The cultures that were not cytotoxic against the autologous tumour were in most cases able to lyse other tumour cell lines of ovarian or other histology. Cloning of TIL from one patient was performed: of 22 clones tested, 4 displayed higher cytotoxicity against the autologous tumour compared to the uncloned population and 3 out of these 4 did not kill an irrelevant carcinoma cell line. In order to stimulate the expansion of putative specific effectors we performed mixed lymphocyte/tumour cultures (MLTC) with autologous or allogeneic tumour cells. No stimulation of cytotoxicity against the autologous tumour was detected after MLTC in nine different TAL populations, using autologous or allogeneic tumours as stimulators. On the contrary, peripheral blood lymphocytes from two patients after MLTC with the autologous tumour showed increased killing of the autologous and decreased killing of an allogeneic target. In conclusion TIL and TAL from ovarian carcinoma expanded in vitro with interleukin-2 usually have non-MHC-restricted cytotoxicity and variable degrees of reactivity against the autologous tumour. A preferential killing for the autologous tumour was not observed even after MLTC. These results do not exclude the existence of tumour-specific cytotoxic T lymphocytes in ovarian carcinoma; nevertheless they suggest that putative specific effectors have very low frequency and that culture techniques for expanding their growth more selectively are still to be optimized.

    Notes: Abstract Intraventricular injection of 5,7-dihydroxytryptamine, selective destruction of descending serotoninergic neurons by 5,6-dihydroxytryptamine or electrolytic and chemical lesions of the nucleus raphe dorsalis did not affect the electroconvulsive threshold in rats. No effect was observed after the systemic administration of drugs known to increase central serotonin transmission, such as quipazine, m-chlorophenylpiperazine, and moderate doses of d-fenfluramine, whereas p-chlorophenylalanine, an inhibitor of serotonin synthesis, decreased seizure susceptibility. The anticonvulsant activity of carbamazepine was not modified in animals with the same experimental lesions. The results, in relation to the high selectivity of the experimental procedures employed to deplete brain and spinal cord serotonin, do not bear out any involvement of serotonin in the tonic component of electrically induced convulsions or in the action of carbamazepine.

    Notes: Abstract Lake Neuchåtel is a medium sized, hard-water lake, lacking varved sediments, situated in the western Swiss Lowlands at the foot of the Jura Mountains. Stable isotope data (δ18O and δ13C) from both bulk carbonate and ostracode calcite in an 81 cm long, radiocarbon-dated sediment core represent the last 1500 years of Lake Neuchåtel's environmental history. Comparison between this isotopic and other palaeolimnologic data (mineralogical, geochemical, palynological, etc.) helps to differentiate between anthropogenic and natural factors most recently affecting the lake. An increase in lacustrine productivity (450–650AD ca), inferred from the positive trend in δ13C values of bulk carbonate, is related to medieval forest clearances and the associated nutrient budget changes. A negative trend in both the bulk carbonate and ostracode calcite δ18O values between approximately 1300 and 1500AD, is tentatively interpreted as due to a cooling in mean air temperature at the transition from the Medieval Warm Period to the Little Ice Age. Negative trends in bulk carbonate δ18O and δ13C values through the uppermost sediments, which have no equivalent in ostracode calcite isotopic values, are concomitant with the recent onset of eutrophication in the lake. Isotopic disequilibrium during calcite precipitation, probably due to kinetic factors in periods of high productivity is postulated as the mechanism to explain the associated negative isotopic trends, although the effect of a shift of the calcite precipitation towards the warmer months cannot be excluded.

    Notes: Summary Some di- and tripeptides containing homoleucine (2-amino-4-methylhexanoic acid) have been synthetized. These compounds have been submitted to biological tests on insects for their hormonal activity in comparison with the known tripeptide pivaloyl-alanyl-p-aminobenzoic acid ethyl ester. Only 2 of them (3 and 4) caused morphological changes on larvae ofPyrrhocoris apterus.

    Notes: Summary The ability of the nocardioform actinomycete Rhodococcus rhodochrous to metabolize selected lignin model compounds was studied. The compounds studied included cinnamic and ferulic acids and dimers possessing intermonomeric linkages that are characteristic of the lignin molecule. R. rhodochrous reduced the carbonyl group of anisoin, a 1,2-diarylethane (β-1) structure to (1R,2R)-1,2-bis(4-methoxyphenyl)ethane-1,2-diol with an enantiomeric excess of .98%. Cleavage of 1,2-diarylethane and β-O-4 structures by this strain could not be detected under our metabolic conditions.

    Notes: Abstract  Pseudomonas fluorescens strain FE2 isolated in the presence of ferulic acid was able to grow on hydroxylated and methoxylated compounds bearing the hydroxyl group in the para position. By ethylmethansulphonate (EMS) and transposon mutagenesis, mutants unable to utilize ferulic acid have been selected. The metabolic characterization of the wild-type strain and its mutants indicates that ferulic acid was degraded through the formation of vanillic acid. Mutant FE2B in co-oxidation experiments with glutamate, is able to transform ferulic and dihydroferulic acid into vanillic acid, 4-hydroxycinnamic acid and 3 (4-hydroxyphenyl)propanoic acid into 4-hydroxybenzoic acid, and 3-hydroxycinnamic acid into 3-hydroxybenzoic acid. The bioconversion of hydroxylated aromatic substrates by the FE2B mutants suggests that the presence of a hydroxyl group on the aromatic ring is required for deacetylase activity.
