    Notes: The 5.5 Mb chromosome 7q21–22 ACHE/PON1 locus harbours the ACHE gene encoding the acetylcholine hydrolyzing, organophosphate (OP)-inhibitable acetylcholinesterase protein and the paraoxonase gene PON1, yielding the OP-hydrolyzing PON1 enzyme which also displays arylesterase activity. In search of inherited and acquired ACHE–PON1 interactions we genotyped seven polymorphic sites and determined the hydrolytic activities of the corresponding plasma enzymes and of the AChE-homologous butyrylcholinesetrase (BChE) in 157 healthy Israelis. AChE, arylesterase, BChE and paraoxonase activities in plasma displayed 5.4-, 6.5-, 7.2- and 15.5-fold variability, respectively, with genotype-specific differences between carriers of distinct compound polymorphisms. AChE, BChE and arylesterase but not paraoxonase activity increased with age, depending on leucine at PON1 position 55. In contrast, carriers of PON1 M55 displayed decreased arylesterase activity independent of the − 108 promoter polymorphism. Predicted structural consequences of the PON1 L55M substitution demonstrated spatial shifts in adjacent residues. Molecular modelling showed substrate interactions with the enzyme variants, explaining the changes in substrate specificity induced by the Q192R substitution. Intriguingly, PON1, but not BChE or arylesterase, activities displayed inverse association with AChE activity. Our findings demonstrate that polymorphism(s) in the adjacent PON1 and ACHE genes affect each other's expression, predicting for carriers of biochemically debilitating ACHE/PON1 polymorphisms adverse genome–environment interactions.

    Notes: An integral equation approach for a binary hard-sphere mixture interacting with a planar semipermeable wall (membrane) is formulated by using the second-order nonuniform or pair Ornstein–Zernike equation as well as the usual singlet Ornstein–Zernike equation. The results of the pair theory are compared with those obtained from the singlet theory and with computer simulation data.     Notes: An extension of the singlet-level equations for the density profile to the case of adsorption of polydisperse fluids on solid surfaces is presented. Explicit calculations and comparisons with canonical ensemble Monte Carlo data have been performed for a polydisperse hard sphere fluid in contact with a hard wall by using the Percus–Yevick, hypernetted chain, and a modified Verlet approximation. A numerical solution of the density profile equations makes use of the orthonormal polynomials with the weight function corresponding to the distribution function of the diameters of fluid particles. © 1999 American Institute of Physics.

    Notes: We study the adsorption, including wetting, of a Lennard-Jones gas on a rough surface consisting of rough layer on a smooth substrate with which the gas interacts via a 9-3 potential. The rough layer is two molecular diameters thick and consists of a disordered quenched matrix of hard spheres. As well as interacting with the other matrix spheres by the hard-sphere potential, the matrix molecules interact with the gas molecules by the hard-sphere potential. Hence, on average, the degree to which the gas molecules can approach the substrate depends only on the density of the matrix layer. The density of this rough layer has a significant effect. As the density of the matrix layer increases, the adsorption isotherms pass from wetting to a prewetting transition, and, if the layer is dense enough, to partial wetting. It is interesting that the prewetting transition remains first order. © 1999 American Institute of Physics.

    Notes: We have investigated adsorption of a hard-sphere fluid in a slitlike pore with composite walls that permit penetration of fluid particles into the wall body for a given depth. Each of the walls is prepared by quenching of a layer of an ideal gas on a hard wall substrate. Our interest is in the adsorption isotherms and in partitioning of the fluid between random microporous layers of a given thickness and an empty space between these layers. The inhomogeneous replica Ornstein–Zernike equations and the Percus–Yevick approximation in conjunction with the Born–Green–Yvon equation for the density profile are used to study the system. The theoretical predictions are compared with the grand canonical ensemble Monte Carlo simulations. The adsorption isotherms and in-plane correlation functions are evaluated and compared with the reference system. We have shown that the presence of random matrix layers deposited on the pore walls significantly influences the structure of the confined fluid. The adsorption isotherms reflect decreasing penetrability of the fluid into a pore with microporous walls. In-plane correlations for two particles located at a rough wall are stronger than the correlations at a hard wall. © 1998 American Institute of Physics.

    Notes: Mycobacterium tuberculosis (Mtb) persists for prolonged periods in macrophages, where it must adapt to metabolic limitations and oxidative/nitrosative stress. However, little is known about Mtb's intermediary metabolism or antioxidant defences. We recently identified a peroxynitrite reductase-peroxidase complex in Mtb that included products of the genes sucB and lpd, which are annotated to encode the dihydrolipoamide succinyltransferase (E2) and lipoamide dehydrogenase (E3) components of α-ketoglutarate dehydrogenase (KDH). However, we could detect no KDH activity in Mtb lysates, nor could we reconstitute KDH by combining the recombinant proteins SucA (annotated as the E1 component of KDH), SucB and Lpd. We therefore renamed the sucB product dihydrolipoamide acyltransferase (DlaT). Mtb lysates contained pyruvate dehydrogenase (PDH) activity, which was lost when the dlaT gene (formerly, sucB) was disrupted. Purification of PDH from Mtb yielded AceE, annotated as an E1 component of PDH, along with DlaT and Lpd. Moreover, anti-DlaT antibody coimmunoprecipitated AceE. Finally, recombinant AceE, DlaT and Lpd, although encoded by genes that are widely separated on the chromosome, reconstituted PDH in vitro with Km values typical of bacterial PDH complexes. In sum, Mtb appears to lack KDH. Instead, DlaT and Lpd join with AceE to constitute PDH.
