    Notizen: In an attempt to elucidate the pattern of spoilage in rock lobster (Jasus novae-hollandiae), samples of abdominal musculature consisting of flexor, extensor and enveloping muscles were subjected to two treatments; half the musculature, after bisecting along the mediosaggital plane, was immersed for 4 min in 3% sodium chloride solution (untreated) and the other half in 3% sodium chloride containing 15 ppm oxytetracycline (treated). After being drained, the muscles from different lobsters were stored under nitrogen, some at 20°C or 15°C for 72 hr, and some at 0°C for 216 hr.In the untreated samples stored at 20°C or 15°C the concentration of total volatile bases (TVB) and trimethylamine (TMA) rose sharply after 24 hr storage. In both the untreated and treated samples at 0°C for up to 168 hr post mortem and in treated samples at 15°C for up to 56 hr post mortem TVB concentrations remained well below 30 mg n/100 g muscle. The TMA concentrations did not rise above 5 mg n/100 g muscle during the entire experimental period in samples held at 0°C.The increase in total bacterial counts correlated well with the rise in the concentrations of TVB and TMA in the untreated samples.Changes in the concentrations of free amino acids showed that bacterial breakdown of arginine into ornithine and ammonia accounted for a very high proportion of ammonia present in the TVB.The significance of these observations to rock lobster spoilage is discussed.

    Notizen: Glycolysis in rock lobster comprising the oblique extensor, oblique flexor and enveloping muscles of Jasus novae-hollandiae was studied under anaerobic conditions at 20°, 15° and 0°C. After bisecting the muscles along the mediosaggital plane one section was treated by dipping for 4 min in 3% sodium chloride solution containing 15 ppm oxytetracycline (treated). The other (untreated) was immersed in sodium chloride solution containing no antibiotic. In the treated muscles lactate production showed a linear relation to the fall in pH over a range of 7·2–6·0, a fall in one unit of pH corresponding to 118 μmol of lactate per g of tissue. In spite of a relatively high production of lactate the ultimate pH did not fall below 6·0 because of a high buffering capacity of the muscle.Breakdown of phosphoraginine and ATP and accumulation of lactate and inorganic phosphorus at 20° and 15°C followed patterns similar to those observed in mammalian and poultry muscle but at 0°C these processes were slowed down to a much greater extent. There was no accumulation of glucose 6-phosphate.Concentrations of arginine rose as a result of phosphoarginine breakdown in both treated and untreated samples. However, in the untreated samples at 20° and 15°C, arginine concentrations started to fall 24 hr post mortem probably due to bacterial attack; the fall being more rapid at 20°C.When mounted in a specially designed rigorometer at 20° and 15°C the muscle strips started to shorten concurrently with only a slight drop in ATP concentrations. The extent of shortening did not exceed 10% of the original and in some cases the contraction was immediately followed by a slow lengthening.The findings of this study are discussed in relation to glycolysis and rigor mortis in mammalian and fish muscles.

    Notizen: Sputum specimens were received for microbiological examination from 110 patients following open heart surgery. The isolation of Haemophilus influenzae occurred significantly more often in those patients who had pre-existing chest disease, but was not associated with postoperative chest problems. There was a significant association between pre-operative heart failure and subsequent severe pulmonary complications.

    Notizen: Histamine and virus-specific IgE appear to play some role in the pathogenesis of wheezing which occurs during viral infection. Respiratory viruses can enhance IgE-mediated histamine release in vitro partly by inducing interleron (IFN). To investigate the role of histamine, IgE, and IFN in the pathogenesis of asthma associated with infection, we prospectively studied hospitalized infants and children (8 months to 17 years of age) with acute episodes of asthma. Patients were enrolled and acute samples were collected within 2 days of admission; convalescent samples were obtained 1 to 4 weeks later. Patients were separated into virus/mycoplasma-assoeiated and noninfectious asthma groups by comprehensive virologic and serologic techniques. One third of acute asthma episodes proved to be virus/mycoplasma-associated. In both asthma groups, nasal wash histamine values were significantly higher during acute attack, compared to convalescence. However, during acute attack, there was a trend for more detectable histamine and in higher concentrations from patients with infectious asthma, compared to the noninfectious group. Histamine was correlated with nasal wash and serum IgE during acute episodes of only infectious asthma. Nasal wash or serum IFN levels did not differ between the 2 patient groups. However, high IFN titers were detected in sera of all 3 patients with asthma associated with RSV infection, although RSV is known to be a poor IFN inducer. Our findings suggest an agreement with the hypothesis that histamine and IgE may play a role in attacks of asthma which arc associated with viral or mycoplasma infection. Further studies are required to confirm our obsmution and to evaluate the role of other inflammatory mediators in the mechanism of infectious asthma.

    Notizen: Summary Twenty-eight outpatients receiving sulfasalazine for inflammatory bowel disease were monitored. Assessment of acetylator phenotype according to the percentage of acetylated sulfapyridine in serum provided a clear distinction between rapid and slow acetylators. In comparison, the percentage of acetylated sulfapyridine in saliva or urine was a less precise index of phenotype. Determination of saliva concentrations of sulfapyridine and N4-acetylsulfapyridine did not provide a reliable estimate of serum levels. Slow acetylators had significantly higher serum concentrations of sulfapyridine (21.9±14.0 [SD] µg/ml) than rapid acetylators (8.8±4.3 µg/ml) and had a higher incidence of toxicity (not statistically significant,p〉0.05). Serum concentrations of sulfapyridine were significantly higher in patients with symptoms of toxicity (23.2±15.9 µg/ml) than those without (13.9±9.5 µg/ml) (p〈0.05). However, serum concentrations of total sulfapyridine (sulfapyridine plus N4-acetylsulfapyridine) were not significantly different in patients with (32.9±21.2 µg/ml) or without (22.8±12.0 µg/ml) toxicity (p〉0.05). For all patients serum concentrations of sulfapyridine (total sulfapyridine) ranged from 3.5 to 73.1 (5.7 to 95.1) µg/ml in patients with controlled disease and 6.3 to 38.0 (14.0 to 54.7) µg/ml in patients with active disease. A significant correlation between clinical status of disease and serum drug concentrations was only apparent for rapid acetylators (p〈0.05). The daily sulfasalazine dosage (mg/kg of body weight, log value) and serum drug concentrations (log values) were highly correlated (p〈0.05). For clinical evaluation of inflammatory bowel disease patients determination of serum sulfapyridine concentrations appears to be more important for monitoring toxicity than therapeutic efficacy of sulfasalazine. Assessment of acetylator status appears to be useful for predicting serum sulfapyridine levels in patients receiving sulfasalazine therapy.
