    Notes: The anti-secretory effects and pharmacokinetics of omeprazole were investigated in ten patients with chronic liver disease. Plasma omeprazole concentrations were measured after a 10-mg intravenous dose of omeprazole and on the first and seventh days of a 7-day course of 10 mg oral omeprazole daily. Pentagastrin tests were performed on the day before oral omeprazole was commenced and 24 h after the last oral dose.The pre-treatment basal and peak gastric acid outputs were low (mean rates of 1.44 mmol/h and 9.26 mmol/h, respectively) and following 7 days of oral 10 mg omeprazole daily, were lowered by 95% and 90% respectively. Following 10 mg intravenous omeprazole, plasma clearance was reduced, and plasma half-life and area under the concentration curve were increased, in comparison with previous studies in healthy subjects. The plasma concentration curves for oral and intravenous doses were very similar. After both the first and seventh oral doses, maximum plasma concentration and area under the curve were higher than in healthy subjects. No accumulation of omeprazole was demonstrated.The pharmacokinetics of omeprazole in chronic liver disease could be influenced by low gastric acidity, poor liver function and/or portasystemic shunting. A dose of 10 mg omeprazole daily has been shown to be an effective anti-secretory agent in chronic liver disease.

    Notes: Current interest in the pharmacological manipulation of portal pressure centres on the long-acting somatostatin analogue SMS 201-995. Nine haemodynamically stable cirrhotic patients who had previously bled from oesophageal varices had wedged and free hepatic venous pressures and cardiac index measured, using a Swan—Ganz catheter, before and at 60, 120 and 180 min after beginning a 60-min infusion of 25 μg/h of SMS 201-995. Seven clinically similar patients had the same measurements performed without SMS 201-995. In all patients cardiac index was found to decrease and systemic vascular resistance increase at 60 min, although heart rates and arterial blood pressures were unchanged. The group given SMS 201-995 was significantly different from the control group in sustaining a fall in wedged hepatic venous pressure and trans-hepatic venous gradient at 60 min. SMS 201-995 causes a fall in portal pressure without a significant systemic haemodynamic effect.

    Notes: Crystals of antithrombin were grown both on earth and in microgravity aboard US Space Shuttle Flight STS-67. The quality of crystals grown in both environments was highly variable and many could not be indexed. The microgravity crystals, however, generally diffracted better, as demonstrated by a novel procedure that estimates the resolution of the Bragg scatter from single diffraction images, without requiring knowledge of the cell dimensions of the crystal. Whereas the best earth-grown crystals never diffracted beyond 3 Å resolution, the best microgravity crystal diffracted to 2.6 Å. The improvement, demonstrated here by a comparison of 23 microgravity and 12 earth-grown crystals, is attributed to better ordered crystal growth in microgravity, although other factors may have contributed also.

    Notes: Abstract Gastric acid secretion was studied in conscious cats with gastric fistulas. Dimaprit and Nα,5-dimethylhistamine produced higher maximal responses than histamine. In the presence of pyrilamine, the maximal response to histamine was equal to that of dimaprit. Pyrilamine increased abmaximal but not maximal responses to pentagastrin. dimetidine decreased the potency of dimaprit but did not after the maximal response. Gastric acid secretion continued for 90 min after infusion of dimaprit was stopped. Possible mechanisms for the enhancement of acid secretion by rilamine include: (a) blocking of an inhibitory histamine H1-receptor and (b) inhibition of histamine methyl-transferase.

    Notes: Abstract A daily surface soil moisture model described and tested previously with data from the Phoenix, AR (USA) area, has been applied now to climate data collected in semi-arid Botswana over a multi-year period. The physically based model was calibrated with observed surface moisture data from one growing season and validated with data from another. Good overall agreement (r2 = .89) was found between the observed and modelled data. The seasonal variability of precipitation in this region is reflected in the simulated daily surface moisture. The response of the natural savanna vegetation as observed from monthly composites of satellite-derived Normalized Difference Vegetation Index (NDVI) is also monitored for the same period. It is seen that the NDVI peaks at different times during the rainy season from one year to the next, according not only to the total seasonal precipitation, but its temporal distribution as well. It is also seen that there is a delay in intraseasonal vegetation response. While the NDVI is poorly correlated with the current month's average soil moisture, a distinct relationship with the previous month's average soil moisture was found.

    Notes: Palladium metal powders used for hydrogen isotope pumping were studied for their bulk and surface chemistry changes during cleaning with hydrogen gas at 150°C. Cleaning is necessary to promote rapid hydriding and dehydriding and to prevent contaminant ingrowth in the process gas during use. These studies show that heating the powders in vacuum at moderate temperatures (150°C) in a reducing environment removes surface oxide, leaves a predominately palladium metal surface which presents no significant barrier to hydrogen absorption or desorption, and shows no loss in surface area and pourability. A surface model of PdO is given and XPS results show that the water-forming reaction is the mode of PdO reduction. Thermodynamic data are discussed which support the above results.
