    Notes: A Ga2Te3 interfacial phase has been observed in a ZnTe/(001)GaSb heterostructure by high resolution electron microscopy under special imaging conditions. This phase exists in domains 5–10 nm in size on the ZnTe side of, and usually 2–4 nm away from, the interface. A structural model has been proposed for this phase that is derived from the sphalerite cell with cation sites occupied either fully (occupancy 1) or partially (occupancy 5/9) by Ga atoms. The fully occupied Ga sites form a regular array of uninterrupted chains along the [110] direction of the sphalerite unit cell. The partially occupied Ga sites can also be considered as forming chains containing both Ga atoms and vacancies along the [110] direction. Within these chains vacancies are highly mobile, resulting in an average Ga occupancy of 5/9. The unit cell of Ga2Te3 is orthorhombic with the space group Amm2. The lattice parameters of the unit cell have been derived from electron diffraction data.

    Notes: 0.1% B2H6 diluted in hydrogen is used as the p-type dopant gas in Si1−xGex grown by ultrahigh vacuum chemical vapor deposition (UHVCVD) using Si2H6 and GeH4. The boron concentration is evaluated by secondary ion mass spectrometry (SIMS). The boron concentration of Si1−xGex increases with the increase of the GeH4 flow rate, that is, Ge fraction, by keeping Si2H6 and B2H6 flow rates constant. The result may be due to the increase of the vacant surface sites which is caused by the increase of the hydrogen desorption rate when a higher Ge fraction epilayer is grown. © 1995 American Institute of Physics.

    Notes: Misfit dislocations in [001] GaAs/In0.25Ga0.75As/GaAs multiquantum well heterostructures are studied by transmission electron microscopy. Two sets of misfit dislocation networks have been observed; one is a set of conventional 60° misfit dislocations lying along 〈110〉, and the other is a set of edge dislocations lying along 〈100〉. The location of these misfit dislocations with respect to the strained layers is investigated. A qualitative explanation for the generation mechanism of the misfit dislocations lying along 〈100〉 is given.

    Notes: Using rosetting methods, we have purified rabit B cells and studied their interactions with T cells purified by passage over an anti-immunoglobulin-coated Degalan beads column. B cells enhance the responce of T cells in concanavalin A (Con A) and phytohaemagglutinin. In regulation of the response to Con A, an adherein cell is a third participating cell B-cell preparations, contain a minority of cells that can respond to T mitogens with the help of non-proliferating T cells, but the proportion of these responding cells is small, and the involvement of a T-cell impurity cannot be excluded.

    Notes: The proliferative response, induced in rabbit spleen cell by concanavalin A (Con A) and phyto-haemagglutinin (PHA), is abolished when T cells are killed with antibody againsi rabbit thymus lymphocyte antigen (RTLA) in the presence of complement. The response was examined witb puritied spleen T cells, to whieh various helper cell factions were added; it could he shown that B cells help the responding T cells, The helper effect in ihe response to PHA is abolished imd ihe response to Con A is reduced by any manoeuvre which destroys or removes B cells. Help by B cells is given when helper cells have lost proliferative capacity as a consequence of mitomycin-C treatment. Spleen cells adhering to the walls of culture tubes help suspended T spleen cells in their response to Con A. This help could be abolished by complement mediated coll kill with antibody to rabbit bursal equivalent lymphocyte antigen (RABELA). On the other hand, the helper effect in Con A response was increased when T cells were removed. Thus the response of T cells to Con A is regulated by helper B cells and suppressor T cells.

    Notes: Immunoglobulin was obtained from a hybridoma cell line, which was a reclone of a hybrid between a rabbit cell and mouse myeloma cell (X63-Ag8). The immunoglobulin, isolated from the cell culture medium was found to be homogeneous by isoelectrofocusing and immunoelectrophoresis and consisted of mouse heavy and rabbit light chains, linked by disulphide bonds. All immunoglobulin molecules carried both mouse and rabbit determinants; mouse determinants were associated only with the heavy chains while rabbit determinants were only associated with the light chains. The rabbit light chains were of Ab4 allotypical specificity, but possess only some of the Ab4 determinants normally present in Ab4/Ab4 animals. It was sugested that the restriction in allotypical specificity may be a general property of light chain Ab allotypes; the normal serum immunoglobulins may be heterogeneous with respect to the allotypic determinants and any one molecule may possess only a proportion of determinants detected by a conventional anti-allotype antiserum.

    Notes: Background: Candida albicans has been implicated in human allergic disorders. However, many of its immunoglobulin E (IgE)-reacting components have not yet been identified. The purpose of the present study is to characterize a novel 29 kDa IgE-binding protein from C. albicans.Methods: The 29 kDa protein was partially purified and its tryptic digests subjected to mass spectrometric analysis. The cDNA encoding this protein was isolated and heterologously expressed in Escherichia coli. Monoclonal antibodies (MoAbs) were raised against the 29 kDa protein purified from C. abicans extracts.Results: We isolated a 29 kDa IgE-reacting component from C. albicans. The protein was digested on-gel with trypsin and the masses of the resulting fragments were determined in a MALDI-TOF mass spectrometer. The data were searched against protein sequences deduced from the C. albicans genome. An open reading frame that possibly encodes the 29 kDa IgE-reacting component was identified. The cDNA corresponding to the open reading frame was isolated. It encodes a 236 residues protein that has 62% sequence identity to that of a hypothetical protein (YDR533c) from Saccharomyces cerevisiae. Conserved domain search suggests that the encoded protein belongs to the ThiJ/PfpI family. The cDNA isolated was inserted into a pQE-30 vector for protein expression in Escherichia coli. The recombinant protein can react with IgE antibodies in sera from asthmatic patients and two MoAbs that were generated against the purified native 29 kDa protein from C. albicans.Conclusions: We identified and cloned a novel 29 kDa IgE-reacting component (Cand a 3) from C. albicans. The recombinant proteins produced from this clone and the MoAbs prepared may be useful in the standardization of diagnostic extracts. They are also instrumental in elucidating the role of C. albicans in clinical allergy.

    Notes: Computer software for specimen orientation adjustment relative to the incident electron beam has been developed. The difficulties encountered when tilting severly strained, highly beam-sensitive, or small-grain-size specimens with known structures using either a double-tilt or a rotation holder in transmission electron miscroscopy (TEM) can be minimized. Combined with computer programs for obtaining the reduced bases from diffraction patterns, the software is also useful for unknown crystals. The algorithm is introduced and examples are given.
