    Notizen: Mycoplasma-like organisms (MLOs) were detected by fluorescence microscopy in a number of pear cultivars and pear seedlings growing under a range of different conditions. In the cultivar Conference, MLOs were consistently associated with symptoms of Parry's disease, a decline-like disease of young trees with quince rootstocks. MLO-free pear seedlings rapidly became infected when they were planted outside. Experimental transmission of MLOs to pear seedlings and Conference trees was achieved using pear psyllids, caught outside or raised on infected plants under controlled conditions. Conference trees in an orchard trial remained free from the severe spring symptoms of Parry's disease when they were protected from feeding insects during spring of the previous year. MLOs were graft transmissible, but were not perpetuated by the standard propagation practices of budding or grafting when quince rootstocks were used. Parry's disease appears to be similar to pear decline, an MLO-induced disease well established in several other parts of the world. It is suggested that Parry's disease should be referred to as pear decline.

    Notizen: Pear explants (Pyrus communis cv. Pine) infected with mycoplasma-like organisms (MLOs) were maintained for more than 3 years by micropropagation on Murashige and Skoog medium supplemented with gibberellic acid, indole butyric acid and benzyl amino purine. MLOs reached consistently higher concentrations in micropropagated explants than in samples from field-grown plants, although explants remained free from symptoms. MLOs could be eliminated by incorporation of 100/μg/ml oxytetracycline into the growth medium for a period of 4 weeks. These results have implications for plant propagation schemes of symptomless MLO-infected explants raised by micropropagation.

    Notizen: Cuttings from sugar-cane plants with symptoms of whiteleaf disease and bermuda-grass plants also showing whiteleaf symptoms were collected in Thailand and transferred to a glasshouse at East Mailing. Using these plants as source material, procedures were devised for the partial purification of MLO-associated immunogens. Antisera were raised separately to the sugar-cane whiteleaf and bermuda-grass whiteleaf immunogens. These antisera exhibited specificity for their homologous MLO antigens in F(ab′)2 indirect enzyme immunosorbent assays (ELISA). Detection of MLO infection was possible in crude tissue extracts diluted several hundredfold. No cross-reactions were observed in reciprocal tests between these two MLO sources and their antisera, nor were there reactions between either antiserum and extracts of other graminaceous hosts showing symptoms of natural whiteleaf infection. No cross-reactions were obtained in reciprocal tests with Spiroplasma citri or aster yellows MLO and their homologous antibodies.

    Notizen: Abstract Methods are described for pre- and post-embedding immunogold labeling of mycoplasmalike organisms (MLOs) in thin sections of infected plants. Antisera against primula yellows (PY), tomato big bud (TBB) and bermudagrass white leaf (BGWL) MLOs, and a monoclonal antibody (mab) against PY were tested with the three serologically unrelated MLOs. Labeling was specific for each MLO and was localized to the outer surface of the MLOs. The antisera performed well in both pre- and post-embedding experiments; the mab reacted well in pre-embedding conditions but gave no labeling with post-embedding. Glutaraldehyde fixation reduced levels of labeling in post-embedding conditions. The results show that these techniques can be used to differentiate MLOs reliably, and extend the usefulness of electron microscopy in this area.
