    Notes: Erianthus arundinaceus has a number of important agronomic traits including good ratooning ability, tolerance to both drought and water logging, disease resistance and vigour and is of interest as a potential source of parental germplasm to sugarcane breeders. However, to date, attempts to produce fertile hybrids between sugarcane (Saccharum spp.) and E. arundinaceus have been unsuccessful. Microsatellite markers that generated genus-specific markers and were highly polymorphic within sugarcane were identified. The microsatellite markers and 5S rDNA PCR were used to screen intergeneric (F1) clones from Saccharum officinarum×E. arundinaceus crosses, and two Saccharum backcross progeny (BC1) populations derived from crosses between selected F1 clones and sugarcane (Saccharum spp.), to identify genuine Saccharum spp. BC1 progeny. The 5S rDNA PCR marker and highly polymorphic microsatellites with Erianthus-specific bands confirmed the F1 parent of the two putative BC1 populations was a S. officinarum×E. arundinaceus hybrid and allowed the identification of the genuine BC1 progeny from selfs of the F1 parent. This is the first verification of BC1 progeny from an F1 intergeneric hybrid x sugarcane (Saccharum spp.) clone with molecular markers and confirms the introgression of E. arundinaceus germplasm into sugarcane. It should now be possible to exploit genes of value from E. arundinaceus in sugarcane breeding programmes.

    Notes: The influence of carbon source and growth regulator composition in induction medium on anther culture response was investigated using spring barley genotypes. Anthers were cultured on BAC3, Ficoll-containing medium, supplemented with one of the following carbohydrates: sucrose, maltose, cellobiosc and melibiose (6 % w/v). The use of either maltose or cellobiose resulted in a significantly higher anther response, calli and/or embryoid production and green plant regeneration compared to the incubation of anthers on a medium containing sucrose. Contrary to these results, the replacement of sucrose by melibiose in BAO medium, drastically reduced the efficiency of anther culture. As an average for the three genotypes tested, the frequency of green plants per 100 anthers plated was 9- to 22-fold higher on medium supplemented with sucrose or cellobiose than on medium containing melibiose as a sole carbohydrate.Among the growth regulators tested, the combination of auxin NAA (2 mg/l) and cytokinin BAP (1 mg/1) performed much better than the employment of auxin 2,4-D combined either with zeatin riboside or BAP as cytokinins. The beneficial effect of medium supplemented with NAA and BAP was associated with better embryoid formation compared to the other growth regulator combinations tested. The hormone-free combination gave a similar anther response and production of calli as any medium supplemented with growth regulators, but the regeneration capacity of calli produced on hormone-free medium was much lower, resulting in the drastic reduction of the number of both total and green regenerants.

    Notes: A general diagnosis is introduced to examine whether and under what condition any Reynolds-averaged Navier–Stokes or large-eddy simulation using models based on linear eddy-viscosity νt is mathematically compatible with the incompressible turbulent flows to be modeled. It is shown that νt is governed by two independent second-order linear partial differential equations with the spatial derivatives of mean or filtered velocity as coefficients. In particular, for two-dimensional mean or filtered flows the νt equation is globally hyperbolic except in some degenerated cases. Numerical examples show that when the hyperbolic equation is incompatible with the boundary condition for νt, the characteristics of the same family may self-intersect to form shocks with finite jumps of νt, caused by the strong dissipation gradient near strong shear layers and vortices. Since in any modeled flow the νt field is globally continuous, once the "νt-shocks" occur the modeling must distort the real flow by weakening or even eliminating some vortices. It is further argued that constitutive-type models should contain the mean or filtered vorticity tensor and hence be nonlinear. © 2002 American Institute of Physics.

    Notes: The mutagenic effects of gamma radiation and sodium azide (SA) immersion of explants on culturability and frequency of somaclonal variation in indica rice were investigated. The results showed that young inflorescence and mature embryo were satisfactory explants both for in vitro culture and for mutagenesis. 1 kR gamma rays applied to immature embryos and exposure of calli derived from mature seeds at the dose range of 250—500 R led to a higher rate of callusing and plantlet regeneration, 0.5—1 kR would be the appropriate exposure for young inflorescence to produce a better culture response and higher variability, and 2.5—5.0 kR the optimal dose range for mature embryos. Immersion of mature seed in 2—4 mM sodium azide solution is a concentration level to induce these effects in indica rice. Application of optimal gamma exposure and chemical mutagen to rice explants enhanced the somaclonal variation frequency and provided a large number of variants useful for rice breeding.
