    Notes: NGFI-B (Nur77/Nr4a1) is a member of a nuclear steroid receptor subgroup that includes the related factors Nurr1 (Nr4a2) and NOR-1 (Nr4a3). These proteins do not have recognized ligands and in fact function independently as orphan receptors with transcriptional regulatory activity. In the present study, expression of the NGFI-B gene in the rat pineal gland was found to exhibit a robust circadian rhythm, with elevated levels of NGFI-B mRNA occurring at night. The rhythm of NGFI-B mRNA is translated into a circadian rhythm of NGFI-B protein, which accumulates in the nucleus of pinealocytes. In addition, there is a parallel marked nocturnal increase in pineal DNA binding activity to a NGFI-B response element (NBRE, AAAGGTCA). Pharmacological studies indicate that NGFI-B mRNA and protein levels are elevated via activation of adrenergic receptors. NGFI-B protein levels are also elevated by dibutyryl cyclic AMP, as in other systems. In the pineal gland, regulation of NGFI-B expression also involves the AP-1 protein Fra-2, based on studies with a transgenic Fra-2 knockdown rat, in which pineal NGFI-B expression increases. This set of observations extends the number of pineal genes that are known to be regulated by Fra-2, and also provides the first indication that a member of the NGFI-B group of nuclear receptors is involved in controlling gene expression in the pineal gland.

    Notes: Recent analysis of endocrine gene transcripts has revealed that several hormone mRNAs exhibit regulated size changes (due to alterations in length of the 3’poly(A) tail) which may function as an additional level of control in the determination of gene expression. We have now shown, through the novel application of an organ culture technique, that prolactin mRNA exhibits a similar regulated change in poly(A) tail length when rat anterior pituitary glands are explanted. The effect is observed in glands of either male or female rats and is specific with respect to growth hormone and α-tubulin mRNAs. Furthermore, we have also found that the size change in prolactin mRNA is attenuated in the presence of bromocriptine, indicating regulation through a dopaminergic pathway.

    Notes: Forebrain vasopressin (VP) neurons of the bed nucleus of the stria terminalis (BNST) contrast with hypothalamic VP neurons in exhibiting nuclear gonadal steroid receptors which may directly effect steroid-induced changes in VP gene expression observed in BNST cells. A transcription and Northern mRNA analysis has been performed to determine the mechanism through which gonadal steroids regulate VP gene expression in the BNST. In addition to confirming a distinctive, sexually dimorphic pattern of VP mRNA expression in the BNST as compared with the hypothalamic supraoptic nucleus (SON), our results show that the marked decrease in BNST VP mRNA levels observed two weeks after castration is not associated with a change in transcriptional activity of the VP gene. Similarly, VP gene transcription is not increased, relative to castrated animals, in the BNST of castrated rats treated with testosterone which exhibit normal or somewhat elevated levels of VP mRNA in the BNST. A post-transcriptional mechanism therefore appears to underlie the gonadal steroid-regulated changes in VP gene expression in the BNST. Since modulation of mRNA size (due to changes in poly (A) tail length) was also observed following castration and testosterone treatment it is apparent that the post-transcriptional mechanism may involve regulated changes in VP mRNA poly (A) tail length.The present findings contrast with the osmotic up-regulation of VP mRNA levels in the SON which is primarily a transcriptional response, and provide a demonstration of the potential physiological importance of post-transcriptional mechanisms of hormonal gene regulation.

    Notes: The description of vasoactive intestinal peptide (VIP) immunoreactivity in the rat anterior pituitary gland has stimulated much interest in possible autocrine and/or paracrine roles as well as in the regulation of its biosynthesis (1–6). The detection of two VIP transcripts in this tissue, together with the knowledge of the rat VIP cDNA sequence (7), has led us to speculate that they are products of the same gene but as a result of differential transcriptional termination, differ in the length of the 3’untranslated region. In this study, we present evidence for the differential utilization of poly(A) addition sites which accounts for the generation of two distinct VIP RNA species.

    Notes: Leukaemia inhibitory factor (LIF) is the generally recognized nomenclature for a pleiotropic cytokine that exerts multiple biological effects in different systems. However, its role in the neuroendocrine system is relatively unexplored, with the exception of one report indicating that LIF may act to determine the unique vascular organization of the pituitary gland. In the present study the expression of LIF mRNA has been demonstrated for the first time in the pituitary gland of the rat, in all three pituitary lobes. However, LIF gene expression is restricted to tissues in explant culture where LIF transcripts are readily detectable by Northern Techniques. Similar studies using the reverse transcriptase-polymerase chain reaction (RT-PCR) technique also failed to detect LIF transcripts in fresh tissue from adult animals. These results are consistent with the hypothesis that LIF acts to mediate the response to trauma, a response that may include the induction of neuropeptide expression in the anterior pituitary.In further studies it has also been shown that anterior pituitary LIF mRNA is super-induced in the presence of protein synthesis inhibitors, a response that indicates a role for a labile protein in the attentuation of LIF expression. Both the explantation response, and the response to protein synthesis inhibitory drugs are decreased in the presence of dexamethasone indicating that a glucocorticoid receptor mechanism acts as a general modulatory influence on LIF mRNA levels. These findings are consistent with the presence of multiple regulatory controls on the expression of LIF that serve to restrict its expression to pathological conditions, and indicate that LIF does not participate in normal endocrine physiology.

    Notes: A transgene consisting of the rat vasopressin structural gene containing a reporter in exon III, flanked by 5 kb of upstream and 3 kb of downstream sequences, is expressed in vasopressinergic, but not oxytocinergic, magnocellular neurons of rats. Functionally appropriate physiological stimuli increase transgene expression in magnocellular neurons in an exaggerated fashion; the magnitude of the transgene response to osmotic challenge exceeds that of the endogenous gene by 10–15 fold. Magnocellular vasopressinergic neurons in the rat are now accessible to rational and precise genetic perturbation of function and regulation.

    Notes: Vasopressin ribonucleic acid (VP RNA) accumulates in the posterior lobe of the rat pituitary in response to an osmotic stimulus. The accumulation of posterior pituitary VP RNA can be prevented by stalk transection or the intracerebroventricular injection of colchicine. The hypothalamic and pituitary VP RNAs however, have different structures and are independently regulated. Depletion of serotonin with parachlorophenylalanine blocks the osmotically induced accumulation of both VP and oxytocin RNA in the hypothalamus but does not affect the accumulation of VP RNA in the posterior pituitary gland. In Brattleboro rats, posterior lobe oxytocin RNA but not VP RNA, increases after osmotic stimulation. Although axonal transport of RNA may occur, our results are also consistent with VP RNA synthesis in pituicytes controlled by hypothalamic neuroendocrine factors released by neurohypophyseal terminals in response to osmotic stimuli.

    Notes: To investigate the expression of nerve cell-specific transgene products in neural transplants, we implanted into the hippocampus of immunosuppressed adult Sprague – Dawley rats cell suspensions obtained from the septal region of the fetal brain of mice that carry the human neurofilament-light (hNF-L) gene. In grafts examined between 3 weeks and 7 months after transplantation, axons and nerve cell somata immunoreacted to antibodies specific to the human NF-L subunit. Thus, the hNF-L protein appears to be a suitable marker of these grafted neurons. Transgenic mice bearing the hNF-L gene may be a convenient source of donor tissue or be used as hosts for neural transplantation studies. Furthermore, the hNF-L promoter/enhancer elements in this transgene may help direct neuronal expression of heterologous genes that could influence nerve cell responses in either the transplant or host tissues.
