    Notizen: The chemisorption of PF3, PF2, and PF on Ni(111) is examined, using tight binding extended Hückel calculations on a three-layer model slab. We have studied various adsorption sites. Calculations indicate that PF3 should be preferentially bound to the onefold on-top Ni sites, with P–F bonds azimuthally oriented over neighboring Ni atoms. PF2 is indicated to bind in a "perpendicular'' manner at a twofold bridging site or possibly at a threefold site; and PF to the threefold hollow sites. These calculations are in agreement with experimental findings. PF3 remains a σ donor and a π acceptor (through backbonding) on the surface; in the bonding of PF2 and PF the π interaction is more important.

    Notizen: Background Asthmatic inflammation is mediated by a network of cytokines, chemokines and adhesion molecules. Corticosteroids are the only effective agents available to control asthmatic inflammation. We investigated the effect of high-dose montelukast (MK), a selective cysteinyl leukotriene receptor 1 antagonist, on mediators of airway inflammation.Objective The aim of this study was to determine the effect of a 3-day course of high-dose MK on mediators of airway inflammation induced by a single allergen challenge in sensitized mice.Methods Ovalbumin (OVA)-sensitized BALB/c mice were treated with 25 mg/kg of MK or saline intravenously for 3 days. On the third day, a single inhalation challenge with OVA was given. Cellular infiltration was assessed in the bronchoalveolar lavage (BAL) and in the lung. Expression of IL-4, IL-5, IL-13 and eotaxin in the BAL, and the lung was determined. Serum IL-5 and total IgE was measured. IL-5 and eotaxin mRNA expression in the lung was determined. Finally, eotaxin and VACM-1 expression in the lung was assessed by immunohistochemistry.Results MK reduced the number of eosinophils in the BAL by 〉 90%. There was also significant reduction in IL-5 in the BAL, lung and the serum, and IL-5 mRNA expression in the lung. IL-4 level in the lung and BAL, and IL-13 level in the lung also significantly decreased. Serum IgE level and lung VCAM-1 expression was also significantly lower in treated animals, but eotaxin protein and mRNA expression in the lung remained unchanged.Conclusion MK exerts its anti-inflammatory effect through the suppression of T helper type-2 (Th2) cytokines. The use of high-dose MK as an anti-inflammatory agent in acute asthma should be further explored.

    Notizen: Summary The effects of chronic administration of 2 types of liquid diets on brain thiamine pyrophosphate (TPP) levels have been investigated. With the Lieber-DeCarli diet, rats in the control group had significantly lower TPP levels compared with those of the ethanol group. The Nutrament diet used in mice was apparently adequate in the thiamine supply.

    Notizen: Abstract Several neurochemical parameters were studied in brain regions of rats chronically treated with a high concentration of manganese chloride (20 mg MnCl2.4H2O per ml. of drinking water) throughout development until adulthood. Large increases in Mn accumulation were found in all brain regions (hypothalamus, +530%; striatum, +479%; other regions, +152 to +250%) of Mn-treated adult rats. In these animals, Ca levels were decreased (−20 to −46%) in cerebellum, hypothalamus, and cerebral cortex but were increased (+186%) in midbrain. Mg levels were decreased (−12 to −32%) in pons and medulla, midbrain, and cerebellum. Fe levels were increased (+95%) in striatum but were decreased (−28%) in cerebral cortex. Cu levels were increased (+43 to +100%) in pons and medulla and striatum but Zn levels were decreased (−30%) in pons and medulla. Na levels were increased (+22%) in striatum but those of K and Cl remained unchanged. Type A monoamine oxidase activities were decreased (−13 to −16%) in midbrain, striatum, and cerebral cortex, but type B monoamine oxidase activities decreased (−13%) only in hypothalamus. Acetylcholinesterase activities were increased (+20 to +22%) in striatum and cerebellum. The results are consistent with out hypothesis that chronic manganese encephalopathy not only affects brain metabolism of Mn but also that of other metals.

    Notizen: Injection molding can be used to fabricate fiber-reinforced polymer composites by impregnating a continuous fabric mat preplaced in a mold cavity with a polymer resin. The mold-filling time is dependent on the flow and heat transfer behavior in the mold. A model is proposed that considers the non-Newtonian How through the porous fabric mat and the heat transfer between mold, fabric mat, and flowing fluid. The model was simulated for the mold filling of a carbon fiber mat with a pseudoplastic polymer solution. The results from the simulation provide Information for optimizing mold-filling parameters through proper selection of inlet fluid pressure, heat source temperature, and type of polymer-solvent system.

    Notizen: The aggregation and structural properties of the synthetic C-terminal half [Ala330, Ala350)270-373; 104-mer)] polypeptide from HIV-1 p24gag were studied. In concentrated solutions the synthetic polypeptide aggregated to tetramers which, upon dilution, gave a mixture of monomeric and dimeric species. These results correlated well with the in vitro aggregation properties of recombinant p24. The tetrameric form of the synthetic polypeptide had a pI which differed by about four units from that of the mixture of monomeric and dimeric species. CD studies indicated that the latter contained, in aqueous solutions, a compact molecule lacking, however, a defined tertiary structure. Addition of MeOH to aqueous solutions of both tetramer and monomer/dimer mixture induced a more defined structure, which was assigned to that of an α+β protein in agreement with secondary structure predictions. A model of the dimeric form of the 104-mer, which takes into account the results presented here and those from a study on the specificity of a set of anti-104-mer MoAbs, is presented.Finally, the results indicated that the structure of the 104-mer in its dimeric form is similar to that adopted by the same sequence when part of full-length p24. © 1997 European Peptide Society and John Wiley & Sons, Ltd.
